Contact and truth: the unfolding predicament in adoption and fostering.

It is now time to examine the crystallizing results of the social experiment of contact, direct and indirect, and to scrutinize the arguments supporting the predominant conviction that the benefits of regular contact are certain. Many adopted and fostered children who have suffered maltreatment at the hands of their birth parents have regular face-to-face meetings with them as well as routine communication in the form of cards or letters. It is commonly maintained that contact has to be beneficial because it is necessarily in the child's best interests in the long run, if not also immediately.

Processes of change in the adoption of consistent condom use.

Descriptions of reproductive and sexual behavior were obtained from interviews with a convenience sample of women at risk of HIV infection o … POC were examined for the range of topics addressed and the POC's perceived function in condom use adoption. Results show that each POC was identified at least once and that all respondents made statements that matched at least two POCs. Only 4 percent of 400 text segments did not fit a POC category. POC constructs appear to be integral components of the …

Adopting a Trauma-Informed Framework in Clinical Practice.

Trauma, including both historical adversity and acute injury, is highly prevalent in the general population. Over 50% of adults report adverse childhood experiences, and surgical patients frequently present with unaddressed psychological distress, which is significantly compounded by maxillofacial injuries and the potentially triggering nature of the care environment. Standard biomedical training typically lacks the necessary competencies for diagnosing, preventing, and managing these complex psychosocial needs. This review proposes and details a comprehensive, evidence-based framework for int

Application of adoptive cell therapy in malignant melanoma.

Cutaneous melanoma is one of the most aggressive skin cancers originating from skin pigment cells. Patients with advanced melanoma suffer a poor prognosis and generally cannot benefit well from surgical resection and chemo/target therapy due to metastasis and drug resistance. Thus, adoptive cell therapy (ACT), employing immune cells with specific tumor-recognizing receptors, has emerged as a promising therapeutic approach to display on-tumor toxicity. This review discusses the application, efficacy, limitations, as well as future prospects of four commonly utilized approaches -including tumor-

Review of Adoptive Cellular Therapies for the Treatment of Sarcoma.

Sarcomas are a heterogeneous group of malignancies with limited therapeutic options, particularly in the metastatic setting. Adoptive cellular therapies (ACTs), including tumor-infiltrating lymphocyte (TIL) therapy, chimeric antigen receptor (CAR) T-cell therapy, and T-cell receptor (TCR) gene-modified T-cell therapy, offer promising novel approaches for these refractory tumors. TIL-based therapy has demonstrated early efficacy in melanoma and myeloma, with ongoing trials exploring its role in sarcoma. CAR T-cell strategies targeting HER2, GD2, and B7-H3 antigens are in development, though cha

An Adoptive Transfer Model of Rheumatoid Arthritis in Mice.

Rheumatoid arthritis (RA) is a chronic, systemic autoimmune inflammatory disorder that may result in joint damage, deformities, disability, and even death. Due to its complex etiology and heterogeneous clinical presentation, current treatment strategies remain inadequate in effectively controlling disease progression, particularly in achieving early diagnosis and providing personalized therapies. Therefore, developing novel therapeutic approaches is crucial. To achieve this, reliable animal models are essential for investigating the pathogenesis of RA. Currently, several animal models of RA ar

Beyond conventional adoptive T-cell therapy.

Reconstitution of the T-cell compartment is essential in the treatment of several immune disorders. Similarly, individuals with hematologic malignancies who are undergoing allogeneic hematopoietic stem cell transplantation experience prolonged T-cell deficiencies, which increase their risk of infections and relapses. Various strategies for addressing T-cell deficiencies are based on adoptive T-cell therapies. However, challenges related to specificity, safety, scalability, and manufacturing have yet to be overcome. Human T lymphoid progenitor-based immunotherapy might be a valuable, complement

Adoptive cellular therapies in non-Hodgkin lymphomas.

Lymphomas are a group of malignant proliferations of B, T or NK-lymphoid cells at different stages of maturation. While they primarily occur in lymph nodes or lymphatic tissues, they can also involve bone marrow, blood, or other organs. Despite advances in treatment, many patients experience relapse, or develop refractory disease, prompting the development of new therapies. One of the most promising innovations is represented by chimeric antigen receptors (CAR) T-cell therapy, that works by genetically modifying a patient's T lymphocytes to better target and kill their cancer cells. Currently,

Challenges and new technologies in adoptive cell therapy.

Adoptive cell therapies (ACTs) have existed for decades. From the initial infusion of tumor-infiltrating lymphocytes to the subsequent specific enhanced T cell receptor (TCR)-T and chimeric antigen receptor (CAR)-T cell therapies, many novel strategies for cancer treatment have been developed. Owing to its promising outcomes, CAR-T cell therapy has revolutionized the field of ACTs, particularly for hematologic malignancies. Despite these advances, CAR-T cell therapy still has limitations in both autologous and allogeneic settings, including practicality and toxicity issues. To overcome these c

Cancer cachexia - adopting a systems wide approach.

PURPOSE OF REVIEW: Cancer cachexia results in the death of approximately 2 million people worldwide annually. Despite the impact of this devastating condition, there is limited therapy and no standard of care. Although multiple definitions exist, confusion remains as a true understanding of the biology has not yet been achieved and distinct phases of cachexia have not been examined. Research has mainly focused on weight loss and muscle wasting, but cachexia is increasingly recognized as a multiorgan disorder involving adipose tissue, liver, brain, gut and heart, with systemic inflammation a ce

Strategies adopted by Salmonella to survive in host: a review.

Salmonella, a Gram-negative bacterium that infects humans and animals, causes diseases ranging from gastroenteritis to severe systemic infections. Here, we discuss various strategies used by Salmonella against host cell defenses. Epithelial cell invasion largely depends on a Salmonella pathogenicity island (SPI)-1-encoded type 3 secretion system, a molecular syringe for injecting effector proteins directly into host cells. The internalization of Salmonella into macrophages is primarily driven by phagocytosis. After entering the host cell cytoplasm, Salmonella releases many effectors to achieve

Adoptive T-cell therapy for Hodgkin lymphoma.

Although CAR T-cell therapy is US Food and Drug Administration-approved for B-cell non-Hodgkin lymphomas, the development of adoptive immunotherapy for the treatment of classic Hodgkin lymphoma (cHL) has not accelerated at a similar pace. Adoptive T-cell therapy with Epstein-Barr virus-specific cytotoxic T lymphocytes and CD30 CAR T cells have demonstrated significant clinical responses in early clinical trials of patients with cHL. Additionally, CD19 and CD123 CAR T cells that target the immunosuppressive tumor microenvironment in cHL have also been investigated. Here we discuss the landscape

A review of adopted colorectal patients: a parent's perspective.

PURPOSE: After treating many adopted patients with congenital colorectal conditions, our goal was to understand if parents were properly counseled about their child's medical needs before adoption. METHODS: A comprehensive questionnaire was developed. Recruitment occurred by social media and colorectal database. RESULTS: 48 parents participated. Adopted children were primarily male (60%), internationally adopted (75%), and a median age of 2.5 years (range newborn-13yo). While 96% of parents received medical records, 41% had incorrect/missing information. Most patients had an anorectal malfor…

Adoptive cell therapy for acute myeloid leukemia.

Acute myeloid leukemia (AML) is the most common tumor in adult patients, most of the patients have a poor prognosis even after high-intensity chemotherapy, especially for relapsed, refractory or elderly patients. Therefore, new methods are needed to change the outcomes. In recent years, an increasing number of immune-therapies are emerging where adoptive cell therapy, a special immunotherapy, has become a promising strategy for AML. Here, we review the clinical application and advancement of donor lymphocyte infusion, chimeric-antigen receptor (CAR) T cell therapy, natural killer (NK) cell the
