Induction of mutations in DNA-repair deficient bacteria by a liver microsomal metabolite of aflatoxin B1.

Certain strains of Salmonella typhimurium and Escherichia coli, particularly those which are very sensitive to u.v. light, are killed when incubated with rat liver mixed function oxidases and aflatoxin B(1). UvrA or recA strains of E. coli are more susceptible than the wild-type strain, while the double mutant uvrA recA is the most sensitive strain yet tested. The aflatoxin B(1) metabolite is also able to induce reverse mutations in 2 histidine auxotrophic strains of S. typhimurium, one strain of which is reverted specifically by frame shift mutagens and the other by agents inducing base pair

Prenatal Aflatoxin B1 Exposure: A Review of Pathogenesis and Impact on Fetal Skeletal Development and Ossification.

Prenatal exposure to aflatoxin B1 (AFB1) poses a significant risk to fetal development and is associated with reduced birth weight in humans. Experimental studies consistently show that AFB1 induces fetal abnormalities, with skeletal malformations and ossification defects being the most common. However, the specific impact of AFB1 on fetal osteogenesis remains unclear. Given this knowledge gap, this study aimed to review the existing literature concerning the pathogenesis of AFB1 and its potential influence on bone development. The primary mechanisms implicated in AFB1's impact on bone include…

Toxicological assessment of cyclopiazonic acid and aflatoxin B1: EFFECTS on zebrafish development and cardiac physiology.

Mycotoxins such as Aflatoxin B1 (AFB1) and Cyclopiazonic Acid (CPA) pose serious health risks, yet their combined toxicity remains poorly understood. This study examined, for the first time, the individual and joint effects of AFB1 and CPA on zebrafish (Danio rerio) embryos, focusing on developmental and cardiac outcomes. Embryos were exposed from 6 h to 4 days post-fertilization to varying concentrations of each toxin and their mixtures. AFB1 caused fin fold wrinkling, underdeveloped pectoral fins, body curvature, growth retardation, and high lethality at elevated doses. CPA induced dose-depe…

Exposure of pregnant and lactating parental mice to aflatoxin B1 promotes hepatotoxicity in offspring mice.

Aflatoxin B1 (AFB1) taints feeds stuffs, endangering livestock's health and resulting in the liver and breast damage. At the same time, while breastfeeding, AFB1 crosses the mammary glands and enters the milk, harming the offspring. This study investigated the liver damaging effects of maternal AFB1 exposure during pregnancy and lactation in offspring mice. The livers of 8-day-old offspring mice were obtained from female mice who were administered AFB1 (2 mg/kg) 1 week prior to and 1 week following birth. The results showed that AFB1 increased the levels of malondialdehyde (MDA), alanine amino…

Differential Hepatic Expression of miRNA in Response to Aflatoxin B1 Challenge in Domestic and Wild Turkeys.

Aflatoxin B1 (AFB1) is a major foodborne mycotoxin that poses a significant economic risk to poultry due to a greater degree of susceptibility compared to other agricultural species. Domesticated turkeys (Meleagris gallopavo) are especially sensitive to AFB1; however, wild turkeys (M. g. silvestris) are more resistant. A lack of functional isoforms of hepatic glutathione S-transferases (GSTs), an enzyme that plays a role in the detoxification of aflatoxin, is suspected as the reason for the increased sensitivity. Previous studies comparing the gene expression of domesticated and wild turkeys e…

Research progress on the protection and detoxification of phytochemicals against aflatoxin B1-Induced liver toxicity.

Aflatoxin B1 (AFB1) is a potent hepatotoxic toxin, which can cause hepatitis, cirrhosis, and liver immunological damage. It has been involved in the etiology of human hepatocellular carcinoma. AFB1 can cause oxidative stress in the body's metabolism process, and then cause cytotoxicity, such as apoptosis and DNA damage. Scientific research has discovered that phytochemicals can induce the detoxification pathway of AFB1 through its biotransformation, thereby reducing the damage of AFB1 to the human body. In clinical treatment, certain phytochemicals have been effectively used in the treatment o…

Effects of multiple risk factors for hepatocellular carcinoma on formation of aflatoxin B1-DNA adducts.

Covalent binding of aflatoxin B1 (AFB1) with hepatic DNA may be a critical step in hepatocarcinogenesis. The extent of the AFB1 binding to DNA may depend on various endogenous factors and concurrent exposure to other environmental agents. This study was performed to determine whether any individual characteristics correlated with the formation of AFB1-DNA adducts. The major AFB1-DNA adduct, AFB1-N7-guanine, was measured using a high performance liquid chromatographic assay in urine samples from 43 asymptomatic hepatitis B virus surface antigen carriers and 43 noncarriers randomly selected from…

Contribution of aflatoxin B1 and hepatitis B virus infection in the induction of liver tumors in ducks.

The study of two major risk factors in the development of hepatocellular carcinoma, namely persistent hepatitis virus infection and exposure to dietary aflatoxins, has been hampered by lack of an experimental system. To this end we have used a Pekin duck model to examine the effect of congenital duck hepatitis B virus (DHBV) infection and aflatoxin B1 (AFB1) exposure in the induction and development of liver cancer. AFB1 was administered to DHBV infected or noninfected ducks at two doses (0.08 and 0.02 mg/kg) by i.p. injection once a week from the third month posthatch until they were sacrific…

Pollen-stigma biomimetic framework: a CuCo2O4 nanozyme-based electrochemical aptasensor for ultrasensitive aflatoxin B1 detection.

Inspired by the highly specific recognition and signal amplification of pollen germination on a stigma, we propose a biomimetic electrochemical sensing strategy for ultrasensitive aflatoxin B1 (AFB1) detection. In this pollen-mimetic system, CuCo2O4 nanoflowers serve as "pollen-mimetic grains", with their bimetallic Cu/Co centers functioning as an internal "energy conversion system" for robust signal output. AFB1 acts as the "stigmatic secretion", triggering the release of CuCo2O4 nanoprobes from the aptamer complex via target-induced strand displacement. The released nanoprobes are subsequent…
