HERC1 Regulates Breast Cancer Cells Migration and Invasion.

Tumor cell migration and invasion into adjacent tissues is one of the hallmarks of cancer and the first step towards secondary tumors formation, which represents the leading cause of cancer-related deaths. This process is considered an unmet clinical need in the treatment of this disease, particularly in breast cancers characterized by high aggressiveness and metastatic potential. To identify and characterize genes with novel functions as regulators of tumor cell migration and invasion, we performed a genetic loss-of-function screen using a shRNA library directed against the Ubiquitin Proteaso

Scorpion Venom Analgesic Peptide, BmK AGAP Inhibits Stemness, and Epithelial-Mesenchymal Transition by Down-Regulating PTX3 in Breast Cancer.

A scorpion peptide reported to exhibit both analgesic and antitumor activity in animal models may present as an alternative therapeutic agent for breast cancer. We aimed to investigate the effect of Buthus martensii Karsch antitumor-analgesic peptide (BmK AGAP) on breast cancer cell stemness and epithelial-mesenchymal transition (EMT). We treated MCF-7 and MDA-MB-231 cells with different concentrations of rBmK AGAP and observed that rBmK AGAP inhibited cancer cell stemness, epithelial-mesenchymal transition (EMT), migration, and invasion. Analysis by qPCR, ELISA, western blot, immunofluorescen

In vitro activity of the αvβ3 integrin antagonist RGDechi-hCit on malignant melanoma cells.

BACKGROUND: In malignant melanoma (MM), overexpression of αvβ3 integrin is linked to a more metastatic phenotype. Development of anti-αvβ3 agents able to counteract melanoma progression would be helpful for disease treatment. A new selective ligand of αvβ3, RGDechi-hCit, has anti-angiogenic properties against endothelial cells in animal angiogenesis models. The aim of this study was to evaluate the in vitro effects of the RGDechi-hCit peptide on MM cell lines. MATERIALS AND METHODS: Cytofluorimetric analysis characterized the cell surface expression of αvβ3 integrin on seven MM cell lines: A3

"Got rats?" Global environmental costs of thirst for milk include acute biodiversity impacts linked to dairy feed production.

Rodents damaging alfalfa crops typically destined for export to booming Eastern markets often cause economical losses to farmers, but management interventions attempting to control rodents (i.e., use of rodenticides) are themselves damaging to biodiversity. These damages resonate beyond dairy feed producing regions through animal migration and are an overlooked part of the transferred environmental burden caused by a growing thirst for milk in China and elsewhere.

Surgical retrieval of a metallic foreign body from the spleen of a dog.

This is the first report of a metallic foreign body in the spleen of a dog. The animal had abdominal discomfort with a tucked-up abdomen and occasional vomition. Radiography and ultrasonography were used to identify a splenic foreign body which was surgically removed through a ventral midline celiotomy. Migration of the foreign body through the stomach wall was suspected as the most likely cause.

Juvenile hormones direct primordial germ cell migration to the embryonic gonad.

Germ cells are essential to sexual reproduction. Across the animal kingdom, extracellular signaling isoprenoids, such as retinoic acids (RAs) in vertebrates and juvenile hormones (JHs) in invertebrates, facilitate multiple processes in reproduction. Here we investigated the role of these potent signaling molecules in embryonic germ cell development, using JHs in Drosophila melanogaster as a model system. In contrast to their established endocrine roles during larval and adult germline development, we found that JH signaling acts locally during embryonic development. Using an in vivo biosensor,

Demonstration of the inflammatory activity of the supernatant of hypersensitive lymph node cells incubated with a high dose of antigen.

When lymphocytes from hypersensitive animals are incubated with antigen, biologically active substances are formed which inhibit the migration of mesenchymal cells from normal animals. These substances were tested by intradermal injection in guinea-pigs and rabbits. The supernatants from incubation of lymphocytes with a high dose of antigen caused immediate pallor which lasted several hours. Later there was a macroscopic inflammation maximal at 24 hours. The histology was characteristic of a delayed hypersensitivity reaction. The injection of the supernatant from hypersensitive lymphocytes inc

Notch signalling governs human enteric nervous system progenitor dynamics.

The enteric nervous system (ENS) is the main branch of the peripheral nervous system that innervates the gastrointestinal tract controlling vital functions. It arises during embryogenesis via migration, proliferation and differentiation of neural crest-derived ENS progenitors. Perturbation of these processes, caused by mutations in key signalling pathway components and transcription factors, prevents progenitor colonisation of the distal gut causing aganglionic phenotypes and enteric neuropathies such as Hirschsprung (HSCR) disease. While animal models implicate Notch signalling in ENS specifi

Genetics and mechanisms leading to human cortical malformations.

Cerebral cortical development involves a complex series of highly regulated steps to generate the laminated structure of the adult neocortex. Neuronal migration is a key part of this process. We provide here a detailed review of cortical malformations thought to be linked to abnormal neuronal migration. We have focused on providing updated views related to perturbed mechanisms based on the wealth of genetic information currently available, as well as the study of mutant genes in animal models. We discuss mainly type 1 lissencephaly, periventricular heterotopia, type II lissencephaly and polymi

Not all butterflies are monarchs: orientation using daytime celestial cues but not magnetic cues in European migratory red admirals (Vanessa atalanta).

Seasonal migration in animals is a widespread and complex phenomenon, yet the mechanisms underlying orientation and navigation remain poorly understood in many taxa. In Lepidoptera, most insights come from monarch butterflies (Danaus plexippus) and Australian bogong moths (Agrotis infusa), which use both visual and geomagnetic cues for orientation. However, these species undertake multigenerational round trips between breeding grounds and specific non-breeding areas and are, therefore, not representative of most other lepidopteran migrants, for which the sensory mechanisms guiding orientation

Lunar cycle and moonlight intensity influence nocturnal migration patterns in a small songbird.

Lunar cycle and moonlight exposure have significant impacts on animal behaviour and physiology. The presence or absence of moonlight, along with predictable changes in brightness throughout the lunar cycle, can shape reproduction, foraging, communication, and other aspects of an animal's world. While it has been shown that invertebrates use the moonlight for orientation, little is known on the effect of the lunar cycle on migratory birds. We found that the lunar cycle affected the nocturnal migration activity of a diurnal songbird species, the Eurasian Skylark Alauda arvensis. The occurrence o

A tiered assessment framework for interregional flows of ecosystem services from migratory species.

The Intergovernmental Science-Policy Platform on Biodiversity and Ecosystem Services has called for assessments explicitly accounting for interregional flows of ecosystem services (ESs) across geographic scales. An important type of interregional ES flow is generated by the long-distance movements of migratory species. Many migratory species provide important benefits to people, and due to migration dynamics, ESs provided in one location may be affected by habitat conservation, or lack thereof, in other locations. The state of the science on interregional flows of ESs from migratory species, h

Uncovering loggerhead (Caretta caretta) navigation strategy in the open ocean through the consideration of their diving behaviour.

While scientists have been monitoring the movements and diving behaviour of sea turtles using Argos platform terminal transmitters for decades, the precise navigational mechanisms used by these animals remain an open question. Until now, active swimming motion has been derived from total motion by subtracting surface or subsurface modelled ocean currents, following the approximation of a quasi-two-dimensional surface layer migration. This study, based on tracking and diving data collected from 25 late-juvenile loggerhead turtles released from Reunion Island during their pre-reproductive migrat

Migratory monarchs that encounter resident monarchs show life-history differences and higher rates of parasite infection.

Environmental change induces some wildlife populations to shift from migratory to resident behaviours. Newly formed resident populations could influence the health and behaviour of remaining migrants. We investigated migrant-resident interactions among monarch butterflies and consequences for life history and parasitism. Eastern North American monarchs migrate annually to Mexico, but some now breed year-round on exotic milkweed in the southern US and experience high infection prevalence of protozoan parasites. Using stable isotopes (δ2 H, δ13 C) and cardenolide profiles to estimate natal origi

Lysophosphatidic acid stimulates cell migration, invasion, and colony formation as well as tumorigenesis/metastasis of mouse ovarian cancer in immunocompetent mice.

We have already established human xenographic models for the effect of lysophosphatidic acid (LPA) on tumor metastasis in vivo. The purpose of this work is to establish a preclinical LPA effect model in immunocompetent mice. We first characterized the mouse epithelial ovarian cancer (EOC) cell line ID8 for its responsiveness to LPA in cell proliferation, migration, and invasion and compared these properties with those of human EOC. The signaling pathways related to cell migration were further investigated using pharmacologic and genetic approaches. The effects of LPA on the tumorigenesis of ID

The migration and attrition of Strongyloides ratti in naive and sensitized rats.

Compressed organ autoradiography has been utilized to study the migration of the homogonic strain of Strongyloides ratti from the site of skin penetration to the gut. This transit is characterized by rapid disappearance of parasites from the cutaneous site of infection, followed by vascular dispersal throughout all organs investigated. For the first time, parasite migration in both naive and previously sensitized hosts is compared; the principal stations of migration are the same in the two groups although there is a quantitative difference in accumulation of parasites with time in these stati

Iliac fixation length and resistance to in-vivo stent-graft displacement.

PURPOSE: Migration of endovascular stent grafts has been related to the security of proximal device fixation to the aortic neck. This study evaluated the importance of iliac fixation in preventing longitudinal in vivo device displacement of a modular, externally supported stent graft. METHODS: Experimental ovine infrarenal aneurysms (n = 8) were treated with a fully supported, modular, bifurcated stent graft (AneuRx, Medtronic, Santa Rosa, Calif). Minimum iliac fixation length (1 cm) was used in four animals and iliac extender modules were used to achieve maximum iliac fixation in four animal

The physiological basis of bird flight.

Flapping flight is energetically more costly than running, although it is less costly to fly a given body mass a given distance per unit time than it is for a similar mass to run the same distance per unit time. This is mainly because birds can fly faster than they can run. Oxygen transfer and transport are enhanced in migrating birds compared with those in non-migrators: at the gas-exchange regions of the lungs the effective area is greater and the diffusion distance smaller. Also, migrating birds have larger hearts and haemoglobin concentrations in the blood, and capillary density in the fli
