Xenobiotic metabolizing enzyme activities in cells used for testing skin sensitization in vitro.

For ethical and regulatory reasons, in vitro tests for scoring potential toxicities of cosmetics are essential. A test strategy for investigating potential skin sensitization using two human keratinocytic and two human dendritic cell lines has been developed (Mehling et al. Arch Toxicol 86:1273–1295, 2012). Since prohaptens may be metabolically activated in the skin, information on xenobiotic metabolizing enzyme (XME) activities in these cell lines is of high interest. In this study, XME activity assays, monitoring metabolite or cofactor, showed the following: all three passages of keratinocyt

Review article: management of cyanide poisoning.

Cyanide poisoning is uncommon, but generates interest because of the presumed utility of an antidote immediately available in those areas with a high risk of cyanide exposure. As part of its regular review of guidelines, the Australian Resuscitation Council conducted a systematic review of the human evidence for the use of various proposed cyanide antidotes, and a narrative review of the relevant pharmacological and animal studies. There have been no relevant comparative or placebo-controlled human trials. Nine case series were identified. Treatment with hydroxocobalamin was reported in a tota

The chemical profile and toxicological impact of heated tobacco products.

INTRODUCTION: Heated Tobacco Products (HTPs) are marketed as less harmful alternatives than cigarettes. While industry-funded studies suggest lower risks associated with HTP use compared with cigarettes, the health impacts of HTP use relative to non-use remain uncertain. METHODS: We reviewed tobacco industry-independent studies published between 2019 and 2024 investigating chemical composition of HTP sticks and emissions, and cardiovascular- and pulmonary health effects associated with their use in human subjects and relevant human in vitro models. Only original research articles were include

[Application of phages in farm animals, food and the environment as part of a One Health approach-potentials and challenges].

In order to ensure the holistic protection of human, animal and environmental health (commonly referred to as One Health), alternatives to the available antimicrobial approaches are required. Although antibiotic drugs have an outstanding broad spectrum of activity, their use is under discussion due to increasingly occurring resistances. Furthermore, the utilisation of antibiotics can result in significant consequences for the affected microbiome, often irreversibly altering its natural composition. The necessity for the implementation of alternative and targeted control measures for bacterial

The critical role of complement in experimental pneumococcal sepsis.

Using a guinea pig model fo bacteremia due to Streptococcus pneumoniae serotype 7, opsonization by the classical and alternative pathways of complement activation was studied in immune and nonimmune animals. Depletion of the alternative complement pathway and complement components C3-C9 resulted in a significant, lethal defect of intravascular clearance in both normal and immune animals. Preopsonization corrected the initial clearance defect in complement-depleted animals. Maximal rates of clearance of bacteremia occurred in immune, normal animals, Immune, C4-deficient animals had clearance cu

Family relationship of female breeders reduce the systematic inter-individual variation in the gut microbiota of inbred laboratory mice.

The gut microbiota (GM) may influence disease expression in several animal models for inflammatory diseases. It may therefore seem reasonable to pursue reduction in the number of animals used for individual studies by reducing the variation in the GM. Previous studies have shown that the composition of the GM is related to genetics to a certain extent. We hypothesized that the GM similarity in a group of mice born by mothers not being sisters would be lower than that in a group born by mothers being sisters. The lower similarity could lead to clustering of the GM of mice born by non-sisters ac

Kidney-on-Chip and Organoid Models: Harnessing Mechanical Forces for Translational Kidney Biology.

The FDA Modernization Act and the subsequent federal policy changes in 2025 have signaled a shift towards the use of non-animal, human-centered models for preclinical drug development and toxicity screening, emphasizing 3D cell-based, organ-on-chip, and organoid platforms as alternatives to animal models. While animal models have been instrumental in improving our understanding of disease mechanisms, they do not allow decoupling of biomechanical and biochemical effects during pathogenesis. Standard in vitro systems offer improved accessibility but often lack physiologically relevant microenvir

Potential Future Pharmacological Treatment of Bladder Dysfunction.

In the last decades, a number of new antimuscarinic drugs have been introduced for treatment of the overactive bladder (OAB), defined symptomatically (OAB syndrome) or urodynamically (detrusor overactivity). Recently, three new drug principles have been approved for clinical use, the β3 -adrenoceptor agonist, mirabegron, the phosphodiesterase-5 inhibitor, tadalafil and the blocker of afferent and efferent nerves, botulinum toxin. However, new alternatives are continuously being explored. OAB is a filling disorder, and ATP is involved in the generation of afferent impulses. One way of blocking

Use and Efficacy of Analgesic Agents in Sheep (Ovis aries) Used in Biomedical Research.

Sheep (Ovis aries) are widely used as large animal models in biomedical research. However, current literature on the use of analgesics in sheep generally focuses on an industry or farm level of use. This structured review evaluates use and efficacy of analgesics administered to sheep in a biomedical research setting. Electronic databases were searched with terms related to analgesia in research sheep. After application of exclusion criteria, 29 peer-reviewed publications were evaluated from 1995 to 2018. Drugs used for analgesia in sheep include opioids, α₂ agonists, NSAID, local anesthetics,

A quantitative in silico model for predicting skin sensitization using a nearest neighbours approach within expert-derived structure-activity alert spaces.

Dermal contact with chemicals may lead to an inflammatory reaction known as allergic contact dermatitis. Consequently, it is important to assess new and existing chemicals for their skin sensitizing potential and to mitigate exposure accordingly. There is an urgent need to develop quantitative non-animal methods to better predict the potency of potential sensitizers, driven largely by European Union (EU) Regulation 1223/2009, which forbids the use of animal tests for cosmetic ingredients sold in the EU. A Nearest Neighbours in silico model was developed using an in-house dataset of 1096 murine

Assessment of diurnal systemic dose of agrochemicals in regulatory toxicity testing--an integrated approach without additional animal use.

Integrated toxicokinetics (TK) data provide information on the rate, extent and duration of systemic exposure across doses, species, strains, gender, and life stages within a toxicology program. While routine for pharmaceuticals, TK assessments of non-pharmaceuticals are still relatively rare, and have never before been included in a full range of guideline studies for a new agrochemical. In order to better understand the relationship between diurnal systemic dose (AUC(24h)) and toxicity of agrochemicals, TK analyses in the study animals is now included in all short- (excluding acute), medium-

Potential benefits of Ginkgo biloba in poultry nutrition: an updated review.

The need for more efficient poultry production to meet the increasing demand for animal protein due to the increase in global human population informs the use of antibiotic growth promoters (AGPs). Rising worries about the possible development of antibiotic-resistant bacteria and residues in chicken products make it imperative to develop alternatives to AGP. Such alternatives must possess qualities that are safe for public health, cost-efficient, environmentally-friendly,possessing antibacterial properties and ensure better growth performance in birds. Examining the Effects of an Anti-Salmonella Bacteriophage Preparation, BAFASAL®, on Ex-Vivo Human Gut Microbiome Composition and Function Using a Multi-Omics Approach.

Salmonella infections (salmonellosis) pose serious health risks to humans, usually via food-chain contamination. This foodborne pathogen causes major food losses and human illnesses, with significant economic impacts. Overuse of antibiotics in the food industry has led to multidrug-resistant strains of bacteria, and governments are now restricting their use, leading the food industry to search for alternatives to secure food chains. Bacteriophages, viruses that infect and kill bacteria, are currently being investigated and used as replacement treatments and prophylactics due to their specifici

Ebselen Inactivates Essential Mycoplasmopsis bovis Proteins.

Mycoplasmopsis bovis contributes to a variety of infectious diseases in cattle that limit meat and milk production, motivating significant antimicrobial use to prevent animal and economic loss. Currently approved antimicrobial treatments for M. bovis are ineffective or becoming so due to adaptive antimicrobial resistance (AMR). Thus, to discover alternatives to these therapies, we screened a drug repurposing library of ∼2800 compounds as antibiotics, resulting in the prioritization of ebselen (Eb) for study. The ability of Eb to prevent M. bovis growth could be ascribed to the covalent modific

Advances in vaccination against tick-borne encephalitis.

Tick-borne encephalitis (TBE) poses a growing health problem in many European countries and parts of Northern Asia. Thus, vaccination has been employed successfully for many years in endemic countries. Long-term experience gained from widespread use, however, prompted the development of improved vaccine formulations of the two licensed European TBE vaccines. Moreover, recent clinical trials also suggested the maintenance of high values of postvaccination neutralizing TBE antibodies for a longer period than expected; thus, also resulting in modifications with regards to immunization regimens. [Postnatal steroid treatment in preterm infants: risk/benefit ratio].

This review examines the risk/benefit ratio of postnatal steroid treatment in preterm infants and correlates epidemiological data with special emphasis on experimental evidence concening the impact of steroid on brain development. With all regimens, steroid treatment consistently reduced the need for assisted ventilation at 28 days of postnatal age or at term. However, neither oxygen at term nor neonatal mortality has been decreased by this treatment. Conversely, respiratory benefits should be weighed against several adverse effects: hyperglycemia, hypertension, gastrointestinal bleeding or pe

Advances in Thyroid Gland Regeneration: The Integrated Approach of Cell Biology and Bioengineering.

The thyroid gland is an endocrine organ responsible for production of triiodothyronine and thyroxine, essential hormones that regulate human metabolism. A wide range of conditions can impair its function, leading to potential life-threatening consequences such as myxedema coma. The standard treatment for hypothyroidism is lifelong levothyroxine supplementation, which, despite being a significant therapeutic breakthrough, has notable limitations and does not fully restore quality of life for many patients. Biomimetic thyroid gland has emerged as a promising alternative treatment strategy for pa

Analysis of the efficacy of Langhans giant cells in controlling cutaneous Mycobacterium marinum infections.

BACKGROUND: Cutaneous Mycobacterium marinum (M. marinum) infection can lead to the formation of infectious granulomas containing Langhans giant cells (LGCs). Due to concerns about prolonged antibiotic use and the development of drug resistance, its treatment poses challenges. As active participants in granulomas, the therapeutic effect of LGCs has not been studied in animal models. OBJECTIVES: We aim to investigate the antibacterial efficacy and immune response after transplanting LGCs into a mouse model of cutaneous M. marinum infection. RESULTS: LGCs effectively reduced the bacterial load

Chitosan 3D scaffolds with resolvin D1 for vertebral arthrodesis: a pilot study.

PURPOSE: Over the last years, the number of vertebral arthrodesis has been steadily increasing. The use of iliac crest bone autograft remains the "gold standard" for bone graft substitute in these procedures. However, this solution has some side effects, such as the problem of donor site morbidity indicating that there is a real need for adequate alternatives. This pilot study aimed to evaluate the usefulness of chitosan (Ch) porous 3D scaffolds incorporated with resolvin D1 (RvD1) as an alternative implant to iliac bone autograft. METHODS: We have performed bilateral posterolateral lumbar ve
