First confirmed autochthonous case of human Babesia microti infection in Europe.

A 42-year-old female patient with acute myeloid leukemia presented with fever and heavy chest pain after her first cycle of specific chemotherapy. Acute myocardial infarction was excluded, but surprisingly, parasitic inclusions in erythrocytes became obvious in Pappenheim and Giemsa-stained peripheral blood smears. The patient did not remember a tick bite but acknowledged having received several blood transfusions in her recent medical history. Suspicion of malaria was ruled out by use of a dip-stick test. The diagnosis of Babesia microti infection was finally established by specific polymeras…

Fay and Rausch 1969 revisited: Babesia microti in Alaskan small mammals.

The Holarctic distribution of Babesia microti within small rodents implies an ancient association. A seminal report of piroplasms in Alaskan voles suggested to us the possibility that B. microti entered North America within Eurasian microtine rodents dispersing through Beringian corridors. To test this hypothesis, we analyzed samples from Alaskan rodents by polymerase chain reaction for evidence of infection with B. microti; one-third of the rodents were found to be infected. Sequence analysis of the 18S rDNA gene demonstrates that Alaskan B. microti comprises a clade that infects microtines i…

Demonstrable parasitemia among Connecticut blood donors with antibodies to Babesia microti.

BACKGROUND: Reports of transfusion-transmitted Babesia microti have risen steadily during the past several years, reflecting a concurrent increase in US cases of human babesiosis. Although several studies have measured B. microti antibodies in blood donors, little is known about associated parasitemia and the inherent risk of transmitting the parasite by transfusion. STUDY DESIGN AND METHODS: Donations from blood donors located in Babesia-endemic and nonendemic areas of Connecticut were tested for B. microti antibodies from July through September. Subsequently, an additional blood sample was …

Macrophages are critical for cross-protective immunity conferred by Babesia microti against Babesia rodhaini infection in mice.

Although primary infection of mice with Babesia microti has been shown to protect mice against subsequent lethal infection by Babesia rodhaini, the mechanism behind the cross-protection is unknown. To unravel this mechanism, we investigated the influence of primary infection of mice with nonlethal B. microti using different time courses on the outcome of subsequent lethal B. rodhaini infection. Simultaneous infections of mice with these parasites resulted in rapid increases in parasitemia, with 100% mortality in BALB/c mice, as observed with control mice infected with B. rodhaini alone. In con…

Effects on in vitro growth of Babesia microti by cells and serum from B. microti and Schistosoma mansoni infected mice.

The effect of peritoneal macrophages and serum from mice infected with Babesia microti, Schistosoma mansoni and B. microti plus S. mansoni on the growth of B. microti was assessed in short term in vitro cultures using the criterium of rate of incorporation of (3H)Hypoxan-thine. In the absence of serum, macrophages and supernatants from macrophage cultures failed to affect the in vitro growth of B. microti. In contrast, in the absen-de of macrophages, serum from mice infected with B. microti and with B. microti plus S. mansoni induced a marked inhibition of the in vitro growth of B. Microti. Th…

Detections of Babesia odocoilei in Ixodes pacificus and Babesia microti in Ixodes angustus Ticks in British Columbia, Canada.

BACKGROUND: Surveillance of tick-borne pathogens is important to inform risks to human and animal health. Through active surveillance, we have detected the continued presence of both Babesia odocoilei since its first detection in 2019 and Babesia microti in British Columbia (BC). MATERIALS AND METHODS: Tick drag sampling was conducted across four regions of the province using a protocol developed by the Canadian Lyme Sentinel Network. Captured ticks were identified morphologically, and pathogens were tested for Borrelia species, Anaplasma phagocytophilum, Babesia species, and Powassan virus. …

Effect of protease inhibitors on the intraerythrocytic development of Babesia microti and Babesia duncani, the causative agents of human babesiosis.

Human babesiosis is a malaria-like, tick-borne infectious disease with a global distribution. Babesiosis is caused by intraerythrocytic, apicomplexan parasites of the genus Babesia. In the United States, human babesiosis is caused by Babesia microti and Babesia duncani. Current treatment for babesiosis includes either the combination of atovaquone and azithromycin or the combination of clindamycin and quinine. However, the side effects of these agents and the resistance posed by these parasites call for alternative approaches for treating human babesiosis. Proteases play several roles in the c…

Isoenzyme patterns of glucose-6-phosphate dehydrogenase, malate dehydrogenase and lactate dehydrogenase in Babesia rodhaini and Babesia microti.

Glucose-6-phosphate dehydrogenase (G6PD), malate dehydrogenase (MDH), and lactate dehydrogenase (LDH) activities and their isoenzyme patterns in B. rodhaini (BR) and B. microti (BM), the two major causative species of murine babesiosis, were examined. G6PD and LDH activities were higher in BR than those in BM, whereas MDH activity was lower in BR than that in BM. No differences were observed between BR and BM in the mobility of isoenzyme bands of G6PD and MDH. On LDH isoenzyme pattern, at least 5 bands were detected in BR, while only one band in BM. Since each subunit of LDH is known to be cod

In vitro monitoring of Babesia microti infection dynamics in whole blood microenvironments.

Babesiosis - a globally emerging tick-borne infectious disease primarily caused by the intraerythrocytic piroplasm parasite, Babesia microti - has traditionally been studied using animal models such as mice. Compared to animal models, microfluidic-based models offer advantages, including direct analysis of human samples (such as patient blood), enhanced assay capacity (including physical/optical access, consistency, and throughput), low costs, and easy adoption. Here, we report an open microfluidic platform named "μ-Blood" for monitoring B. microti infection dynamics in vitro . Compared to oth…

Histopathological Aspects of the Influence of Babesia microti on the Placentas of Infected Female Rats.

Babesiosis is perceived mainly an animal disease; however, awareness that Babesia spp. parasites that can cause diseases in humans is increasing significantly. Babesiosis is spread by the bite of an infected tick (Ixodes spp.), but it can also be transmitted by transfusion of infected blood and from an infected mother to her child during pregnancy or childbirth. The parasites multiply in the bloodstream and destroy red blood cells. This study aimed to assess the influence of Babesia microti on the histological structure of the placenta. Histopathological material collected from pregnant rats i…

Protective efficacy and correlates of immunity of immunodominant recombinant Babesia microti antigens.

Babesia microti, an intraerythrocytic apicomplexan parasite, is the primary causative agent of human babesiosis and an emerging threat to public health in the United States and elsewhere. An effective vaccine against B. microti would reduce disease severity in acute babesiosis patients and shorten the parasitemic period in asymptomatic individuals, thereby minimizing the risk of transfusion-transmitted babesiosis. Here we report on immunogenicity, protective efficacy, and correlates of immunity following immunization with four immunodominant recombinantly produced B. microti antigens-Serine Re…

A Novel Thioredoxin-Like Protein of Babesia microti Involved in Parasite Pathogenicity.

Babesiosis poses a serious threat to immunocompromised individuals and the major etiological species of Babesia for human babesiosis is Babesia microti. Merozoites are a critical stage in the life cycle of Babesia microti. Several merozoite proteins have been demonstrated to play important roles in this process; however, most of the merozoite proteins of B. microti remain unknown. In the present study, we identified a novel merozoite protein of B. microti with similar structure to the thioredoxin (Trx)-like domain of the Trx family, which was named as B. microti Trx-like protein (BmTLP). Weste…

Detection of antibodies to Anaplasma phagocytophilum and Babesia microti using linear peptides.

Anaplasma phagocytophilum and Babesia microti are emerging tick-borne pathogens in the United States. Although active infection is typically diagnosed by direct diagnostic tests, such as blood smear or polymerase chain reaction assay, serologic assays can be helpful to identify past infections, and the use of acute plus convalescent testing can potentially identify recent infections. We employed a peptide array to select sets of linear peptides for serologic diagnosis of infections with A. phagocytophilum and B. microti. Three optimal peptides were selected for each agent based on their perfor…

In Silico Survey and Characterization of Babesia microti Functional and Non-Functional Proteases.

Human babesiosis caused by the intraerythrocytic apicomplexan Babesia microti is an expanding tick-borne zoonotic disease that may cause severe symptoms and death in elderly or immunocompromised individuals. In light of an increasing resistance of B. microti to drugs, there is a lack of therapeutic alternatives. Species-specific proteases are essential for parasite survival and possible chemotherapeutic targets. However, the repertoire of proteases in B. microti remains poorly investigated. Herein, we employed several combined bioinformatics tools and strategies to organize and identify genes …

Molecular Survey of Babesia microti (Aconoidasida: Piroplasmida) in Wild Rodents in Turkey.

Babesia microti (Aconoidasida: Piroplasmida) (Franca, 1910) is an important tick-borne zoonotic parasite with rodents serving as reservoir hosts. In the present study, 536 rodents were captured from Burdur, Bartin, Giresun, and Yozgat provinces of Turkey between the years 2010 and 2012, and blood samples were examined for the presence of Babesia spp. using conventional PCR which targeted the 18S rRNA gene. The sequence analysis of PCR amplicons was tested for B. microti as well as for Hepatozoon spp., and Sarcocystis spp. Overall, 5.8% of the rodents were positive for B. microti: 41% in Myodes…

Evidence for vesicle-mediated antigen export by the human pathogen Babesia microti.

The apicomplexan parasite Babesia microti is the primary agent of human babesiosis, a malaria-like illness and potentially fatal tick-borne disease. Unlike its close relatives, the agents of human malaria, B. microti develops within human and mouse red blood cells in the absence of a parasitophorous vacuole, and its secreted antigens lack trafficking motifs found in malarial secreted antigens. Here, we show that after invasion of erythrocytes, B. microti undergoes a major morphogenic change during which it produces an interlacement of vesicles (IOV); the IOV system extends from the plasma memb…
