Effects of nitric oxide synthase inhibition on glutamine action in a bacterial translocation model.

Glutamine may be a precursor for NO synthesis, which may play a crucial role in bacterial translocation (BT). The goal of the present study was to investigate the potential effects of glutamine on BT and the immunological response in an experimental model of NO synthase inhibition by NG-nitro-L-arginine methyl ester (l-NAME). Mice were randomly assigned to four groups: sham; intestinal obstruction (IO); IO+500 mg/kg per d glutamine (GLN); IO+GLN plus 10 mg/kg per d l-NAME (GLN/LN). The groups were pretreated for 7 d. BT was induced by ileal ligation and was assessed 18 h later by measuring the…

Effects of glucagon-like peptide-2 on bacterial translocation in rat models of colitis.

BACKGROUND/AIMS: The aim of this trial was to study the role of glucagon-like peptide-2 in reducing bacterial translocation by virtue of its anti-inflammatory effects and ability to decrease intestinal permeability in rat models of inflammatory bowel diseases. On the basis of our results and those of other recent studies, we suggest a new treatment modality for colitis. To our knowledge, this is the first study of the effectiveness of glucagon-like peptide-2 on bacterial translocation, in treating an experimental colitis model. METHODS: Rats were randomized into 3 groups of 7 rats each-the co…

Intestinal barrier dysfunction in developing liver cirrhosis: An in vivo analysis of bacterial translocation.

AIM: Transmucosal passage of bacteria across the intestine, the essential step for bacterial translocation, has been identified as a key event in the pathogenesis of life-threatening infections in cirrhosis. Existing animal models of liver cirrhosis only provide indirect information about the pathogenesis of such infections. The aim of this study has been to assess transmucosal passage and bacterial translocation directly in vivo using a rat model of developing liver cirrhosis. METHODS: Male Sprague Dawley rats were randomly assigned to two groups: controls and animals with developing liver c…

Effect of CO(2) insufflation on bacteremia and bacterial translocation in an animal model of peritonitis.

BACKGROUND: The widespread adoption of the laparoscopic approach has created some concern over the potential for increased risk of bacteremia and sepsis due to increased intraabdominal pressure in patients with intraabdominal infection and peritonitis. This study examines the effect of the CO(2) pneumoperitoneum on bacteremia and bacterial translocation. METHODS: New Zealand white rabbits were assigned into three groups of 10 animals. In group 1, 100 ml of sterile saline was infused into the peritoneal cavity under 10 mmHg CO(2) insufflation for 1 h. Group 2 received 100 ml of saline containi…

IgA defects in CVID lead to bacterial translocation, increased serum γ-interferon, and BAFF.

Common variable immunodeficiency (CVID) is a primary antibody defect that leads to frequent infections, but inflammatory complications appear in 30-50%, leading to increased morbidity and mortality. We have previously shown that circulating bacterial 16S ribosomal DNA (rDNA), originating from gut commensals, is significantly increased in the serum of patients with CVID with inflammatory conditions (P = 0.0007). Here we examined the relationships between serum 16S ribosomal DNA (rDNA), isotype-switched memory (SM) B cells, serum IgA, IgA+ SM B cells, serum IFN-γ, and serum B cell-activating fac

Post-flexible Bronchoscopy Escherichia coli Empyema: A Rare Complication or an Inflammatory Bacterial Translocation?

Empyema is an unusual complication of diagnostic bronchoscopy. We report a case of a male patient in his late 70s with bronchiectasis who developed a right lower lobe empyema 10 days after undergoing flexible bronchoscopy for incidentally detected lung nodules. Bronchial washings grew Staphylococcus aureus (S. aureus) and the atypical Gram-negative bacillus Pantoea septica (P. septica), while subsequent pleural fluid culture yielded Escherichia coli (E. coli). This case highlights the risk of post-bronchoscopy infectious complications in patients with structural lung disease and illustrates ho

Risk factors for bacterial translocation after loop ileostomy closure in patients with colorectal cancer.

PURPOSE: Although rare, septic shock can develop following the closure of an ileostomy created during colorectal cancer surgery. In such cases, bacterial translocation (BT) is considered the primary diagnosis, and appropriate treatment is provided. Herein, we investigated the risk factors of BT following ileostomy closure after colorectal cancer surgery. METHODS: A retrospective analysis was conducted using the colorectal cancer database of Nagasaki University, focusing on 91 patients who received ileostomy closure after colorectal cancer surgery. The patients were divided into two groups bas…

Gut microbial translocation corrupts myeloid cell function to control bacterial infection during liver cirrhosis.

OBJECTIVE: Patients with liver cirrhosis suffer from increased susceptibility to life-threatening bacterial infections that cause substantial morbidity. METHODS: Experimental liver fibrosis in mice induced by bile duct ligation or CCl4 application was used to characterise the mechanisms determining failure of innate immunity to control bacterial infections. RESULTS: In murine liver fibrosis, translocation of gut microbiota induced tonic type I interferon (IFN) expression in the liver. Such tonic IFN expression conditioned liver myeloid cells to produce high concentrations of IFN upon intrace

Inhibition of nitric oxide with aminoguanidine reduces bacterial translocation after endotoxin challenge in vivo.

BACKGROUND: Administration of lipopolysaccharide (LPS) has been shown to increase bacterial translocation (BT) in vivo and in vitro. In addition, LPS upregulates inducible nitric oxide synthase expression in the intestinal epithelium-a phenomenon that can either enhance microbial killing, or alternatively, promote BT by impairing the gut barrier. OBJECTIVE: To determine the effect, if any, of an inhibitor of nitric oxide synthase, namely, aminoguanidine (AG), on BT after LPS challenge. DESIGN: Sprague-Dawley rats were randomized to receive either AG or normal saline solution via subcutaneous…

Gut bacterial translocation/dissemination explains the increased mortality produced by parenteral nutrition following methotrexate.

Mortality is reported to be higher in methotrexate (MTX)-treated animals receiving total parenteral nutrition (TPN), compared to animals receiving enteral nutrition. The increased mortality is felt to be related to gut atrophy and bacterial translocation. In this study, we examined the effect of MTX (30 mg/kg) on survival, body weight loss, gut mass, and gut bacterial translocation in rats randomized to TPN or enteral nutrition. Twenty-four male Sprague-Dawley rats (n = 8 per group) were randomized to TPN, a peptide-based enteral diet (PEP), or CHOW. Animals were weighed daily and followed for…

[Bacterial translocation from the gastrointestinal tract and endogenous infection induced by immunosuppression after burn].

This study described the endogenous infections and bacterial translocation from GI tract caused by immunosuppression after burn. In the group of burned plus injected dexamethasone (DXM) (BIS, n = 31), the rate of enteric bacteria translocation was 67.4%, the rate of visceral abscess was 65.5%, much more higher than in the group of only DXM (IS, n = 15). The translocation of intestinal bacteria also was found in the group of only burned (BU 3/15) and control (ck, 1/23), but endogenous infection did not occur in both group. The bacteria cultured from the rat organs are mainly enteric bacilli and…

Bruton tyrosine kinase modulates systemic immune activation to bacterial translocation in primary antibody deficiencies.

BACKGROUND: Bacterial translocation is a shared phenomenon in common variable immunodeficiency (CVID) and X-linked agammaglobulinemia (XLA). In CVID, bacterial translocation is linked to systemic immune activation and chronic inflammatory manifestations. OBJECTIVE: We investigated whether the absence of functional Bruton tyrosine kinase (BTK) in XLA is associated with protection against systemic inflammation driven by bacterial translocation, and to assess whether BTK inhibition could modulate these inflammatory responses in CVID. METHODS: Clinical data from the US national registry were ana…
