Epidermal basement membrane: its molecular organization and blistering disorders.

The epidermal basement membrane is a specialized structure localized between the epidermis and the dermis. Recent studies have elucidated the biological roles of the basement membrane and its pathophysiological involvement in bullous diseases. To understand the functions of the basement membrane, it is essential to have clear and precise information regarding the ultrastructural molecular organization of the epidermal basement membrane. Immunoelectron microscopy is a powerful technique and the only method available for the clarification of the ultrastructural localization or orientation of mol…

Thin basement membrane nephropathy associated with other glomerular diseases.

Many reports confirm that thin basement membrane nephropathy (TBMN) commonly occurs together with other glomerular diseases such as minimal change glomerulonephritis, diabetes, membranous nephropathy, immunoglobulin (Ig)A glomerulonephritis, and focal segmental glomerulosclerosis. We postulate 3 explanations for these observations. The association of minimal change glomerulonephritis with TBMN probably is artifactual whereas the association with diabetes and membranous glomerulonephritis probably is coincidental. However, the link between TBMN and IgA disease and focal segmental glomeruloscler…

Basement membrane and beta amyloid fibrillogenesis in Alzheimer's disease.

High-resolution ultrastructural and immunohistochemical studies revealed that in situ beta amyloid fibrils of Alzheimer's disease were made up of a core consisting of a solid column of amyloid P component (AP) and associated chondroitin sulfate proteoglycan, and a heparan sulfate proteoglycan surface layer with externally associated fine filaments of beta protein. The main body of beta amyloid fibrils closely resembled that of microfibrils. Abundant microfibrils were reported to be present at the basement membrane of capillaries with "leaky" blood-urine or blood-air barriers. Similarly, abunda…

Histological differentiation and basement membrane distribution in gastric adenocarcinomas.

To analyse the histological distribution of basement membranes (BM) in gastric adenocarcinomas, we produced a monoclonal antibody, BM909 (IgG 2b, kappa), which has been found useful for identifying the BM in routinely processed specimens. The BM 909 antibody was shown by ELISA to be negative against the three major BM components (type IV collagen, laminin and fibronectin). Gastric carcinomas from 78 patients including both differentiated and undifferentiated adenocarcinomas were classified into three types (tubular, trabecular, and isolated) based on the histological arrangement of the cancer …

[Immunohistochemical alterations in basement membrane components in skin cancer].

To investigate alterations in the basement membrane (BM) components around tumor nests, Bowen's disease (BD), actinic keratosis (AK), basal cell epithelioma (BCE), squamous cell carcinoma (SCC) were studied by double immunofluorescent staining with antibodies to laminin (LN), type IV collagen (CIV), heparan sulfate proteoglycan (HSPG), and chondroitin 6-sulfate glycosaminoglycan (C6S). In BD, all BM components were continuous on the dermo-epidermal junction. In AK, C6S was partially disrupted, but the other components were continuous. In BCE, LN and CIV were continuous around the tumor nests, …

Electron microscopic findings in corneal epithelial basement membrane degeneration.

In an attempt to correlate basement membrane changes with clinical symptoms we examined 29 specimens from 28 patients who had undergone corneal epithelial basement membranectomy for corneal epithelial basement membrane degeneration (CEBMD). Twenty-two of the 28 patients were women. Recurrent erosion was the most frequent symptom and occurred in 24 of the 29 eyes. The most common electron microscopic finding was reduplication of the basement membrane with loculated connective tissue. The electron microscopic finding that predicted clinical symptoms was absence of the basement membrane and hemid…

Abnormally thin glomerular basement membrane and the Goodpasture epitope.

Using indirect immunofluorescence, cryostat sections from renal biopsy specimens of 14 adult patients showing marked diffuse thinning of the glomerular basement membrane (GBM) on ultrastructural analysis were examined for the presence of the Goodpasture (GP) epitope M2 using anti-M2 antiserum. In no case was a total absence of M2 noted. The fluorescence pattern was fine but homogeneously linear along the GBM in 12 cases, intensity varying from +-++, as compared with for the control specimen GBM. A faint, broken line of stain, intensity+, was observed in biopsy specimens of two patients, one of…

Hereditary nephritis: immunoblotting studies of the glomerular basement membrane.

Hereditary nephritis (HN) is associated with antigenically abnormal glomerular basement membrane (GBM) manifest by reduced or absent binding of MCA-P1, a mouse monoclonal antibody which recognizes Goodpasture antigen. In the present studies, immunoblotting has been used to analyze antigenic and biochemical composition of renal tissue from patients with HN in whom binding of MCA-P1 could not be demonstrated by indirect immunofluorescence (IIF). Pooled normal collagenase-solubilized GBM (CS-GBM) and CS-GBM from three patients with either end-stage renal failure (ESK1-3) or HN (HNK1-3), were exam…

Glomerular basement membrane thickness in children. A morphometric study.

Measurements of glomerular basement membrane (GBM) thickness in children with minimal change nephrotic syndrome were made on electron micrographs of 43 biopsy specimens obtained from 35 children aged 1-13 years, of whom 13 were in remission at the time of biopsy. Measurements were also made in 2 children with non-glomerular disorders. A mean of 189 measurements per biopsy were made, using prints of x6,400-12,000 magnification. Quadratic curves were fitted by regression analysis to the mean GBM thickness for each of the 35 initial nephrotic biopsies, plotted against age for the whole group and …

Coexistent Wegener's granulomatosis and anti-glomerular basement membrane disease.

Wegener's granulomatosis and Goodpasture's syndrome represent two major causes of a pulmonary-renal syndrome. We describe the clinical course and morphologic features of a patient in whom pulmonary manifestations of Wegener's granulomatosis developed and were followed six months later by anti-glomerular basement membrane disease. Although we regard this as a unique and probably fortuitous association, a genetic predisposition or a secondary form of anti-GBM disease cannot be excluded.

Specific changes in the basement membrane of the proximal tubules in the murine polycystic kidney detected by the novel anti-basement membrane monoclonal antibody D28.

In order to analyze renal abnormalities in mice with polycystic kidney disease (PKD), we produced a series of monoclonal antibodies reactive with the murine kidney by hybridizing P3U1 myeloma cells with spleen cells from DBA/2 mice immunized with the kidney of adult-type PKD mice, DBA/2FG-pcy. One clone, D28, reacted specifically with the basement membrane of the proximal tubules of DBA/2 mice and DBA/2FG-pcy mice. It did not react with other parts of the murine kidney nor with other tissues such as the skin, ovary, fallopian tube, testis, lung and small intestine. While other components such …

[Effect of dithranol therapy on membrane, basement membrane and nuclear markers in psoriasis lesions].

We investigated the effects of anti-psoriatic therapy with dithranol (1/20-1%) in salicylic acid (0.5%) in white petrolatum on lesional skin. FITC-labeled lectins and pemphigus vulgaris antibodies (PV) served as analytical means to study the glycocalyx. Antibodies of bullous pemphigoid (BP) were used as basal membrane markers. Nuclear antigens were recorded according to the binding of speckled, anti-nuclear antibodies (ANA) as well as antibodies to dsDNA. With some lectins, dithranol therapy resulted in pronounced fluorescence of the lower parts of the basal cells. ConA was fixed by the basal

Fabry Disease in a Patient With Thin Basement Membrane Nephropathy.

Diagnosing Fabry disease can be challenging due to its broad spectrum of clinical presentations, highlighting the need for a high index of suspicion. A 59-year-old male patient with personal and family history of persistent hematuria was referred to our renal clinic for workup. Genetic workup confirmed a heterozygous inheritance pattern of alpha-4 chain of type IV collagen. A renal biopsy revealed characteristic electron microscopic findings typical of a lipid storage disease within the glomerular podocytes, reminiscent of Fabry disease. This case in an older adult patient underscores the vari

AMACO is a component of the basement membrane-associated Fraser complex.

Fraser syndrome (FS) is a phenotypically variable, autosomal recessive disorder characterized by cryptophthalmus, cutaneous syndactyly, and other malformations resulting from mutations in FRAS1, FREM2, and GRIP1. Transient embryonic epidermal blistering causes the characteristic defects of the disorder. Fras1, Frem1, and Frem2 form the extracellular Fraser complex, which is believed to stabilize the basement membrane. However, several cases of FS could not be attributed to mutations in FRAS1, FREM2, or GRIP1, and FS displays high clinical variability, suggesting that there is an additional gen…

Evidence of integrity of the follicular basement membrane in Hashimoto's thyroiditis.

The basement membrane as an antigenic structure in autoimmune diseases has been a matter of controversy. The purpose of our study was to determine possible structural changes in the follicular basement membrane (FBM) in thyroid autoimmune and non-autoimmune diseases. Immunohistochemical staining for collagen IV and laminin showed that the continuity of the FBM was preserved in toxic adenoma (three cases), atoxic multinodular goiter (nine cases) as well as in autoimmune disease. Integrity of the FBM was observed in all 11 cases of Graves' disease and Hashimoto's thyroiditis (seven cases) studie…

Perlecan maintains the integrity of cartilage and some basement membranes.

Perlecan is a heparan sulfate proteoglycan that is expressed in all basement membranes (BMs), in cartilage, and several other mesenchymal tissues during development. Perlecan binds growth factors and interacts with various extracellular matrix proteins and cell adhesion molecules. Homozygous mice with a null mutation in the perlecan gene exhibit normal formation of BMs. However, BMs deteriorate in regions with increased mechanical stress such as the contracting myocardium and the expanding brain vesicles showing that perlecan is crucial for maintaining BM integrity. As a consequence, small cle…

The basement membrane zone in lichen sclerosus: an immunohistochemical study.

The alteration in expression of basement membrane zone (BMZ) components in lichen sclerosus was investigated by immunohistochemical staining of skin biopsies from seven patients with histologically confirmed disease compared with controls. Monoclonal antibodies and polyclonal sera directed against proteins of the hemidesmosomes, anchoring fibrils, lamina lucida, lamina densa and BMZ collagens were used. Characteristic histological appearances at the dermo-epidermal junction were reflected in widespread alterations in antigen expression in the epidermal basement membrane and the papillary dermi…

Localization of a corneal basement membrane glycoconjugate in bovine eye.

Lectin histochemistry was used to analyze the ocular surface basement membrane in order to identify novel, tissue-specific glycoconjugates. Soybean agglutinin (SBA), a lectin marker for N-acetylgalactosamine residues, recognized a 130 kDa glycoconjugate in corneal but not conjunctival basement membrane. Following corneal epithelial cell wounding in vitro, the glycoconjugate was not expressed at the epithelial cell-stromal interface until 72-96 h in culture, much later than the expression of other basement membrane molecules. In epithelial cells maintained on various extracellular matrix substr…
