Transforming DNA integrates at multiple sites in the dimorphic fungal pathogen Blastomyces dermatitidis.

Blastomyces dermatitidis is a primary fungal pathogen of man and other mammals, but like many other human fungal pathogens, relatively little is known about the factors that account for its virulence and pathogenicity. We developed a transformation system to facilitate molecular genetic studies of putative virulence factors from B. dermatitidis. Transformation of the multinucleate yeasts was achieved by electroporation of DNAs containing a dominant selectable marker, hygromycin B (HygB) resistance. Southern analysis showed that transforming DNA invariably integrated ectopically into the chromo…

Induction and detection of cell-mediated reactions with different Blastomyces dermatitidis antigenic preparations.

Blastomyces dermatitidis yeast-phase antigens (killed cell, KWY; lysate, Lys; and filtrate, Fil) from canine isolate T-58 were compared with respect to the induction and detection of cellular immune responses in mice. The antigens exhibited good sensitivity and specificity when used to detect a delayed-type hypersensitivity (DTH) response in mice previously immunized with T-58 or Histoplasma capsulatum antigens. Greater reactivity was observed with the KWY and Lys antigens as DTH-inducing agents (immunogens) than with the Fil antigen. The antigens were also compared with regard to the inductio…

Enzyme immunoassay detection of antibodies in canine blastomycosis using Blastomyces dermatitidis lysate antigens.

A Blastomyces dermatitidis yeast phase lysate antigen (T-58) prepared in our laboratory was used in an enzyme-linked immunosorbent assay (ELISA) for the detection of antibodies in 201 serum specimens from dogs with blastomycosis. In addition 36 sera from non-infected dogs from an endemic area for blastomycosis and 36 sera from non-infected dogs residing in a blastomycosis non-endemic area were assayed. Sensitivity values ranged from 96.2% (53 pre-treatment sera) to 88.5% (148 post-treatment sera) with the specimens from dogs with confirmed blastomycosis. Minimal reactivity was experienced with…

В реальном времени Blastomyces dermatitidis yeast-induced responses of immune and nonimmune human mononuclear cells.

Peripheral blood mononuclear cells from humans with treated blastomycosis or from normal persons were cultured with live Blastomyces dermatitidis yeast. There was no inhibition of growth of the fungus in this suspension culture technique but morphologic and functional differences of the human cells were great between the two groups. Lymphocyte stimulation by live Blastomyces yeast was found in the patient group but not in the normal donors. These events add to the observations that cellular immunity is expressed in blastomycosis.

Immunoadjuvant effects of Blastomyces dermatitidis against EL 4 lymphoma in C57BL/6J mice.

The ability of thimerosal-killed Blastomyces dermatitidis yeast cells, which greatly enhance the cell-mediated immune response in C57BL/6J mice, to act as an immunopotentiator against EL 4 lymphoma was investigated. Mice treated with yeast cells were protected from as many as 10(4) tumor cells. Complete suppression of tumor growth was observed in treated animals that received ip injections of 10(2) or 10(3) tumor cells. The mice, however, were not immune to further EL 4 lymphoma cell challenge. The lack of tumor-specific immunity indicated nonspecific suppression of tumor growth probably by ma…

Isolation of skin test-active preparations from yeast-phase cells of Blastomyces dermatitidis.

Cell wall and cytoplasmic fractions were isolated from mechanically disrupted yeast-phase cells of Blastomyces dermatitidis in an effort to obtain a reliable skin-test antigen. The biological activities of these fractions were compared with those of two blastomycin preparations. The cytoplasmic antigens exhibited a low index of specificity yet exceeded the specificities of the blastomycins. The skin test-active component(s) of the cytoplasmic material had a molecular weight between 10,000 and 30,000 and could easily be concentrated by ultrafiltration on a PM-10 membrane. Unlike the cytoplasmic…

Blastomyces Urine Antigen Testing for Active Case Identification During a Blastomycosis Outbreak.

Blastomyces urine antigen testing is a sensitive blastomycosis diagnostic method, but its utility for active case identification during outbreaks is unknown. We evaluated urine antigen testing for identifying blastomycosis cases during a 2023 outbreak at a Michigan, USA, paper mill and assessed demographic and clinical factors associated with test positivity. Approximately 2 months after the outbreak was recognized, we collected work and health information for 603 employees; 95% (n = 578) underwent urine antigen testing and 9% (n = 52) tested positive, including 25 previously undetected cases.…

Blastomyces dermatitidis Yeast Lysate Antigen Combinations: Antibody Detection in Dogs with Blastomycosis.

The systemic fungal infection, blastomycosis, which infects both humans and animals has presented a diagnostic challenge for clinicians for many years. The aim of this study was to evaluate the diagnostic sensitivity of Blastomyces dermatitidis yeast lysate antigens with respect to antibody detection in dogs with blastomycosis. Lysate antigens were prepared from B. dermatitidis isolates T-58 and T-66 (dogs, Tennessee) and WI-R and WI-J (dogs, Wisconsin). Based on results obtained from a preliminary comparative study, five combinations of these isolates and one individual isolate were tested ag…

The Dynamic Genome and Transcriptome of the Human Fungal Pathogen Blastomyces and Close Relative Emmonsia.

Three closely related thermally dimorphic pathogens are causal agents of major fungal diseases affecting humans in the Americas: blastomycosis, histoplasmosis and paracoccidioidomycosis. Here we report the genome sequence and analysis of four strains of the etiological agent of blastomycosis, Blastomyces, and two species of the related genus Emmonsia, typically pathogens of small mammals. Compared to related species, Blastomyces genomes are highly expanded, with long, often sharply demarcated tracts of low GC-content sequence. These GC-poor isochore-like regions are enriched for gypsy elements…

Identification of the mating-type (MAT) locus that controls sexual reproduction of Blastomyces dermatitidis.

Blastomyces dermatitidis is a dimorphic fungal pathogen that primarily causes blastomycosis in the midwestern and northern United States and Canada. While the genes controlling sexual development have been known for a long time, the genes controlling sexual reproduction of B. dermatitidis (teleomorph, Ajellomyces dermatitidis) are unknown. We identified the mating-type (MAT) locus in the B. dermatitidis genome by comparative genomic approaches. The B. dermatitidis MAT locus resembles those of other dimorphic fungi, containing either an alpha-box (MAT1-1) or an HMG domain (MAT1-2) gene linked t…

Genetic diversity in Blastomyces dermatitidis: implications for PCR detection in clinical and environmental samples.

Blastomycosis is a serious and potentially fatal infection caused by the thermally dimorphic fungus Blastomyces dermatitidis. Polymerase chain reaction (PCR) assays targeting the BAD-1 virulence gene promoter have been developed to aid in the detection of the pathogen in clinical and environmental samples. However, little is known regarding the genetic diversity of B. dermatitidis and how this might affect the performance characteristics of these assays. We explored the genetic relatedness of 106 clinical and environmental isolates of B. dermatitidis using a previously described rDNA PCR restr…

Growth inhibition of Blastomyces dermatitidis in alveolar and peripheral macrophages from patients with blastomycosis.

Blastomyces dermatitidis evokes a pyogranulomatous disorder with organisms frequently found inside giant cells. Macrophages from bronchoalveolar lavage fluid and peripheral blood in monolayer cell cultures were challenged with live yeast organisms to examine phagocytosis and intracellular growth. A greater number of macrophages from patients recovering from blastomycosis had phagocytized Blastomyces compared with macrophages from healthy control donors. No differences were detected within the groups between alveolar and peripheral macrophages. Intracellular growth of the fungus was reduced in …
