Effect of psychotropic drugs on caudate spindle in cats.

To ascertain whether neuroleptics act on the caudate nucleus itself, the effects of these compounds as well as other centrally acting drugs were examined in relation to caudate spindle and EEG arousal responses (sciatic nerve stimulation) in gallamine-immobilized cats. Haloperidol and chlorpromazine enhanced the caudate spindle at a dose which had no effect on the EEG arousal response. On the other hand, clozapine and a higher dose of chlorpromazine enhanced the caudate spindle, but depressed the arousal response. High frequency stimulation of the sciatic nerve suppressed the caudate spindle. …

Shape analysis of subcortical nuclei in Huntington's disease, global versus local atrophy--results from the TRACK-HD study.

Huntington's disease (HD) is characterized by brain atrophy. Localized atrophy of a specific structure could potentially be a more sensitive biomarker reflecting neuropathologic changes rather than global volume variation. We examined 90 TRACK-HD participants of which 30 were premanifest HD, 30 were manifest HD and 30 were controls. Using FMRIB's Integrated Registration and Segmentation Tool, segmentations were obtained for the pallidum, caudate nucleus, putamen, thalamus, accumbens nucleus, amygdala, and hippocampus and overall volumes were calculated. A point distribution model of each struc…

Increased proteins in post-mortem brain in a case of Pick's disease and in Huntington's disease.

Soluble proteins from temporal cortex and caudate nucleus from a case of Pick's disease, 5 cases of Huntington's disease and 5 controls were analysed by SDS-polyacrylamide gel electrophoresis. Acidic proteins of molecular weight 39 000-42 000, which showed glial fibrillary acidic protein immunoreactivity, were increased in temporal cortex of the Pick's case. Proteins of these molecular weights were also increased in caudate nucleus of the Huntington's cases. Our results show that the astrocytic gliosis observed in temporal cortex in Pick's disease and in caudate nucleus in Huntington's disease…

Thalamostriatal projections from the ventral anterior nucleus in the dog.

Thalamostriatal projections from the ventral anterior nucleus (VA) were mapped by using autoradiographic and horseradish peroxidase techniques in the dog. Injections of tritiated leucine and proline into the lateral, central, and medial parts of VA resulted in anterograde label over the dorsolateral, midlateral, and dorsal parts of the head of the caudate nucleus, respectively. The dorsolateral and midlateral parts of the caudate contained the heaviest label. No silver grains were located over the medial or ventral parts of the caudate. Light to moderate label was located over the most dorsal …

Characteristics of hypersomnia due to inflammatory demyelinating diseases of the central nervous system.

BACKGROUND: Neuromyelitis optica spectrum disorder (NMOSD) diagnostic criteria for inflammatory demyelinating central nervous system diseases included symptomatic narcolepsy; however, no relevant case-control studies exist. We aimed to examine the relationship among cerebrospinal fluid orexin-A (CSF-OX) levels, cataplexy and diencephalic syndrome; determine risk factors for low-and-intermediate CSF-OX levels (≤200 pg/mL) and quantify hypothalamic intensity using MRI. METHODS: This ancillary retrospective case-control study included 50 patients with hypersomnia and 68 controls (among 3000 pati

[Effect of nembutal anesthesia, electric stimulation of the paws and elaboration of a defensive reflex on acetylcholinesterase activity and protein content in different regions of rat brain].

Pentobarbitone sodium anaesthesia was found to produce an increase in protein content in some regions of the rat brain, i. e. posterior cortex, caudate nucleus, and a decrease in protein content in the ventral cortex. Acetylcholinesterase expressed in terms of wet weight was found to increase in the cerebellum, medulla, and to decrease in the medial cortex, hippocampus, thalamus and caudate nucleus. The changes in activity were not explicable in terms of a direct effect of the anaesthetic on the enzyme. A decrease in protein content of rat brain was observed in the frontal cortex, ventral cort…

Early atrophy of pallidum and accumbens nucleus in Huntington's disease.

In Huntington's disease (HD) atrophy of the caudate nucleus and putamen has been described many years before clinical manifestation. Volume changes of the pallidum, thalamus, brainstem, accumbens nucleus, hippocampus, and amygdala are less well investigated, or reported with contradicting results. The aim of our study is to provide a more precise view of the specific atrophy of the subcortical grey matter structures in different stages of Huntington's disease, and secondly to investigate how this influences the clinical manifestations. All TRACK-HD subjects underwent standardised T1-weighted 3…

Cortical activity in Parkinson's disease during executive processing depends on striatal involvement.

Patients with idiopathic Parkinson's disease exhibit impairments in executive processes, including planning and set-shifting, even at the early stages of the disease. We have recently developed a new card-sorting task to study the specific role of the caudate nucleus in such executive processes and have shown, using functional magnetic resonance imaging (fMRI) in young healthy adults, that the caudate nucleus is specifically required when a set-shift must be planned. Here the same fMRI protocol was used to compare the patterns of activation in a group of early-stage Parkinson's disease patient…

Caudate volumes in childhood predict symptom severity in adults with Tourette syndrome.

BACKGROUND: Most children with Tourette syndrome (TS) experience a marked decline in the severity of tic symptoms during adolescence. Currently no clinical measures can predict whose tic symptoms will persist into adulthood. Previous cross-sectional imaging studies have identified reduced caudate nucleus volumes in subjects with TS. OBJECTIVE: To evaluate whether caudate nucleus volumes in childhood can predict the severity of tic or obsessive-compulsive symptoms at follow-up in early adulthood. METHODS: In a prospective longitudinal study, clinical status and basal ganglia volumes of 43 chi…

Repeated amphetamine administration outside the home cage enhances drug-induced Fos expression in rat nucleus accumbens.

Induction of the immediate early gene protein product Fos has been used extensively to assess neural activation in the striatum after repeated amphetamine administration to rats in their home cages. However, this technique has not been used to examine striatal activation after repeated administration outside the home cage, an environment where repeated drug administration produces more robust psychomotor sensitization. We determined the dose-response relationship for amphetamine-induced psychomotor activity and Fos expression in nucleus accumbens and caudate-putamen 1 week after repeated admin

An fMRI study of reward-related probability learning.

The human striatum has been implicated in processing reward-related information. More recently, activity in the striatum, particularly the caudate nucleus, has been observed when a contingency between behavior and reward exists, suggesting a role for the caudate in reinforcement-based learning. Using a gambling paradigm, in which affective feedback (reward and punishment) followed simple, random guesses on a trial by trial basis, we sought to investigate the role of the caudate nucleus as reward-related learning progressed. Participants were instructed to make a guess regarding the value of a …

Forebrain damage following prenatal exposure to low-dose X-irradiation.

Exposure of fetal rats to X-irradiation on gestational day 15 resulted postnatally in dose-related effects on body weight, growth of forebrain structures, and branching of dendrites of caudate neurons. Rats were followed for 4 months postnatally after 125, 75, 50, or 25 R whole-body irradiation to the dam. Significant decreases in body weight were present at birth after the three high doses and continued as long as 4 months after 125 or 75 R. Decreased thickness of the cerebral cortex and decreased area of the caudate nucleus were also seen. Cortical thickness was reduced by 125 R to half the …
