An experimental colitis in rabbits is described, following the intravenous injection of preformed immune complexes of human serum albumin (HSA) and anti-HSA into non-sensitised rabbits. Tissue damage was localised to the colon by the Auer technique of inducing local non-specific inflammation, by the rectal instillation of dilute formalin. We have developed a rapid purification method for DNA topoisomerase I from Raji cells, a human Burkitt lymphoma cell line, using ammonium sulfate fractionation followed by chromatography on a Mono S column (FPLC, Pharmacia). The removal of all debris and background, as well as antifertility factors present in seminal fluid, may increase the fertilizing ability of spermatozoa. A number of semen manipulative techniques are currently available to remove the undesirable spermatozoa, debris and other factors and to increase sperm quality before performance of the various assisted reproductive technologies. We compared the impact of the wash and concentration method, swim-up method, 80% Percoll method, migration-gravity sedimentation method and SpermPrep method on postmanipulation sperm quality.
