Inhibition of pentagastrin-induced pressor response in conscious rats by the CCK-B receptor antagonist CI-988 and chlordiazepoxide.

Elevation of blood pressure is a characteristic feature of spontaneous and pharmacologically induced panic attacks in man. We examined wheth … ependent increases in arterial blood pressure of up to 41 mmHg. The benzodiazepine receptor agonist chlordiazepoxide (10 mg/kg i.v, 15 min previously) attenuated the pressor response induced by either pentagastrin (1 microgram/kg i.v.) or FG-7142 (0.3 mg/kg i.v.). In contrast, the CCK-B receptor antagonist CI-988 (3 mg/kg i.v., 15 min previously) inhibited the pre …

Ethanol, pentobarbital, and chlordiazepoxide effects in squirrel monkeys responding under fixed-ratio food presentation and stimulus-shock termination schedules.

Ethanol, pentobarbital, and chlordiazepoxide were administered to squirrel monkeys responding under a multiple schedule comprised of two fixed-ratio 100-response schedules (FR 100). Under one FR schedule, the one-hundredth response produced food. Under the other schedule, the one-hundredth response terminated the prevailing stimuli and accompanying schedule of electric shock presentation (stimulus-shock termination). Comparable high response rates were maintained under the two schedules. Each drug produced dose-related decreases in response rates. With all three drugs the effects on all aspect…

Desensitization of a gamma-aminobutyric acid type A receptor in rat is increased by chronic treatment with chlordiazepoxide: a molecular mechanism of dependence.

When rats were made tolerant to the benzodiazepine tranquilizer chlordiazepoxide (CDPX) by its steady administration, a particular gamma-aminobutyric acid type A (GABAA) receptor in cerebral cortex was modified. Its rate of desensitization in the absence of CDPX was enhanced (3-fold with 10 microM GABA) below saturation with GABA, and the dependence of this rate on GABA concentration was changed from sigmoid to hyperbolic. This mimicked the effect of the presence of CDPX on desensitization of the naive receptor. This receptor has been characterized by its rapid desensitization (t1/2 = 30 msec …

Learnt tolerance to sedative effects of chlordiazepoxide on self-stimulation performance, but no tolerance to facilitatory effects after 80 days.

Sedative and facilitatory effects on variable-interval hypothalamic self-stimulation were monitored during chronic treatment with chlordiazepoxide (CDP; 7.5 mg/kg IP), given at 48-h intervals in two groups of rats. Group 1 was injected immediately before each of 40 1-h self-stimulation sessions ("drugged responding"); Group 2 was injected after self-stimulation for the first 20 sessions ("undrugged responding"), and before self-stimulation for a further 20 sessions ("drugged responding"). Significant sedation occurred in both groups in initial sessions of drugged responding, even though Group …

Behavioral studies with anxiolytic drugs. I. Interactions of the benzodiazepine antagonist Ro 15-1788 with chlordiazepoxide, pentobarbital and ethanol.

Lever pressing by squirrel monkeys was maintained under two behavioral procedures known to be sensitive to anxiolytic drugs. Under one procedure, responding maintained by food was suppressed by electric shock (punishment). Under a second procedure, responding was maintained under a multiple schedule in which the first response after 5 min produced either food or shock depending on the stimulus that was present throughout the interval (fixed-interval schedule). Under the punishment schedule, chlordiazepoxide (1.0-100 mg/kg), pentobarbital (1.0-17.0 mg/kg) and ethanol (0.5-2.5 g/kg) increased re…

The anxiogenic action of FG 7142 in the social interaction test is reversed by chlordiazepoxide and Ro 15-1788 but not by CGS 8216.

The effects of FG 7142 were examined in the social interaction test, alone and in combination with chlordiazepoxide, Ro 15-1788 and CGS 8216. The anxiogenic action of FG 7142 (5 mg/kg) was reversed by chlordiazepoxide (5 mg/kg) and by Ro 15-1788 (10 mg/kg), but not by CGS 8216 (10 mg/kg). The profile of FG 7142 in this test and the pattern of its interactions with other compounds is similar to that of beta-CCE, but can be distinguished from that of Ro 15-1788 and CGS 8216. It is concluded that FG 7142, Ro 15-1788 and CGS 8216 may act via different sites to produce their anxiogenic effects.

Histamine as a punisher in squirrel monkeys: effects of pentobarbital, chlordiazepoxide and H1- and H2-receptor antagonists on behavior and cardiovascular responses.

Squirrel monkeys pressed a key under a two-component, 30-response fixed-ratio schedule of food presentation. In both nonpunishment and punis … tamine could be markedly increased by presession i.m. treatment with pentobarbital (3-5.6 mg/kg) or chlordiazepoxide (10-30 mg/kg). Presession i.m. treatment with 1 to 3 mg/kg of the H1-receptor antagonist, diphenhydramine, reversed the punishment effects of histamine but only enhanced the cardiovascular effects of histamine. In contrast, 10 to 30 mg/kg of the H2- …

Differential interactions between chlordiazepoxide, pentobarbital and benzodiazepine antagonists Ro 15-1788 and CGS 8216 in a drug discrimination procedure.

Rats were trained to discriminate either chlordiazepoxide (CDP, 4 mg/kg, IP, N = 8) or pentobarbital (PB, 10 mg/kg, IP, N = 8) from saline in a two-lever food-reinforced procedure. CDP and PB dose-dependently substituted for each other (greater than or equal to 90% drug lever responses); indicating that their discriminative stimulus properties were closely similar. However, discriminative stimulus control induced by CDP and PB differentially was affected by the proposed benzodiazepine (BDZ) antagonists Ro 15-1788 (0.08-20 mg/kg, IP) and CGS 8216 (2.5-20 mg/kg, IP) in each experimental group; s…

Effects of 8-OH-DPAT, buspirone and ICS 205-930 on feeding in a novel environment: comparisons with chlordiazepoxide and FG 7142.

Previously, the 5-hydroxytryptamine (5-HT)1A receptor agonists, 8-hydroxy-2-(di-n-propylamino)tetralin (8-OH-DPAT) and buspirone and the 5-HT3 receptor antagonist ICS 205-930 have been shown to exert anxiolytic-like effects in several animal models. In the experiments reported here the effects of these compounds on feeding behaviour and food preference in a novel environment were examined, and compared with the effects of the anxiolytic drug chlordiazepoxide and the anxiogenic compound FG 7142. Chlordiazepoxide significantly reduced the latency to begin eating and prolonged the total time spen…

Maternal aggression towards different sized male opponents: effect of chlordiazepoxide treatment of the mothers and d-amphetamine treatment of the intruders.

Lactating female rats vigorously attack equally sized conspecific males introduced into their home cage. Under conditions of such high aggression, the previously reported pro-aggressive action of a low (5 mg/kg) dosage of chlordiazepoxide (CDP) is hardly detectable. When opponents are large, the intensity of the aggression is less than what is seen with small ones. In this situation treatment of the females with CDP increases aggression levels substantially. The importance of intruders evoking aggression was further investigated by treating different sized opponents with d-amphetamine. d-Amphe…

A novel LC-MS/MS method for simultaneous estimation of chlordiazepoxide and clidinium in human plasma and its application to pharmacokinetic assessment.

A highly sensitive and selective LC-MS/MS assay was developed and validated for the simultaneous quantification of Chlordiazepoxide and Clidinium in human plasma for the first time, employing solid-phase extraction. Chromatographic separation of the analytes and their deuterated internal standards was performed on a reversed-phase Kinetex XB-C18 (150 × 4.6 mm, 5 μm) column with a gradient mobile phase. Mass spectrometric detection was achieved using electrospray ionization in positive ion mode, employing the ion transitions: m/z 300.0 → 227.1 for Chlordiazepoxide, m/z 352.1 → 142.1 for Clidini…

Effects of plus-maze experience and chlordiazepoxide on anxiety-like behavior and serotonin neural activity in the dorsal raphe nucleus in rats.

The extent to which rats express anxiety-like behavior on the elevated plus-maze (EPM) depends on their previous maze experience. Open-arm avoidance develops in maze-experienced rats, and is often accompanied by a diminished anxiolytic response to benzodiazepines. Regions of the dorsal raphe nucleus (DRN) were examined in male Sprague-Dawley rats using c-Fos and serotonin immunohistochemistry following a single exposure, a second exposure or no exposure to the EPM. We then examined the effect of the benzodiazepine anxiolytic chlordiazepoxide (CDP, 5 mg/kg) on EPM behavior and DRN neural activi…

Optimization of dispersive liquid-liquid microextraction with central composite design for preconcentration of chlordiazepoxide drug and its determination by HPLC-UV.

A simple, rapid, and sensitive method based on dispersive liquid-liquid microextraction combined with HPLC-UV detection applied for the quantification of chlordiazepoxide in some real samples. The effect of different extraction conditions on the extraction efficiency of the chlordiazepoxide drug was investigated and optimized using central composite design as a conventional efficient tool. Optimum extraction condition values of variables were set as 210 μL chloroform, 1.8 mL methanol, 1.0 min extraction time, 5.0 min centrifugation at 5000 rpm min(-1), neutral pH, 7.0% w/v NaCl. Pindolol antagonizes the effects on hippocampal rhythmical slow activity of clonidine, baclofen and 8-OH-DPAT, but not chlordiazepoxide and sodium amylobarbitone.

Effects of chlordiazepoxide, chlorpromazine, diazepam, diphenylhydantoin, flunitrazepam and haloperidol on the voltage-dependent sodium current of isolated mammalian brain neurons.

The effects of chlordiazepoxide, chlorpromazine, diazepam, diphenylhydantoin, flunitrazepam and haloperidol on the voltage-dependent sodium current (INa) were studied on the hippocampal pyramidal neurons, isolated acutely from rats, using a concentration clamp technique. The drugs used here reduced dose-dependently the peak amplitude of INa without affecting its current-voltage relationship. Chlorpromazine was most potent drug inhibiting the INa among them. Chlorpromazine and diphenylhydantoin at the concentration of half inhibition (IC50; 4 x 10(-6) M and 2 x 10(-4) M, respectively) shifted t…

Antagonism of the anticonflict effects of chlordiazepoxide by beta-carboline carboxylic acid ethyl ester, Ro 15-1788 and ACTH(4--10).

The antagonism of the anticonflict effect of chlordiazepoxide (CDP) by beta-carboline carboxylic acid ethyl ester (BCCE), Ro 15-1788 and ACTH(4--10) has been evaluated in the Geller-Seifter rat conflict test in which CDP increases punished (conflict), but not unpunished responding. BCCE (0.5--10 micrograms ICV) produced a dose-dependent reduction in the anticonflict activity of CDP. This was also significantly reduced by Ro 15-1788 (25 mg/kg IP) and a high dose of ACTH(4--10) (5 micrograms ICV). None of these test compounds had a marked direct effect on punished or unpunished responding in the…

Thiopental, phenobarbital, and chlordiazepoxide induce the same differences in narcotic reaction as ethanol in long-sleep and short-sleep selectively-bred mice.

Hypnotic effects following administration of thiopental, phenobarbital or chlordiazepoxide were evaluated in mice selectively-bred for differential hypnotic sensitivity to ethanol. For every dose employed, except one which had no effect, all three agents induced greater sedation in the ethanol-sensitive Long-Sleep (LS) line than in the ethanol-insensitive Short-Sleep (SS) line. Such findings with regard to the LS and SS lines suggest that the differences in sedative response to ethanol, as well as some barbiturates and benzodiazepines, may be mediated, in part, by a common mechanism. The secon…
