Intra-erythrocyte chromium as an indicator of exposure to hexavalent chromium: An in vivo evaluation in intravenous administered rat.

Hexavalent chromium (Cr(VI)) compounds are classified as carcinogenic to humans. Whereas chromium measurements in urine and plasma attest to the last few hours of total chromium exposure (all oxidation states of chromium), chromium in red blood cells (RBC) is attributable specifically to Cr(VI) exposure over the last few days. Before recommending Cr in RBC (CrIE) as a biological indicator of Cr(VI) exposure, in vivo studies must be undertaken to assess its reliability. The present study examines the kinetics of Cr(VI) in rat after a single intravenous dose of ammonium dichromate. Chromium leve…

Speciation of chromium in tanned leather gloves and relapse of chromium allergy from tanned leather samples.

There are concerns that wearing chromium tanned leather gloves can provoke a lapse of hand dermatitis in chromium sensitive patients. Various forms of chromium have different biological effects and the determination of total chromium (both soluble and insoluble), which often is used in clinical studies, may therefore not be an adequate measure of the adverse exposure. However, investigations, where dissolution of hexavalent chromium from leather gloves has been experimentally studied, could not been found in the literature. The aim of this pilot study was therefore to determine the total amoun…

Reduction of chromium(VI) by ascorbate leads to chromium-DNA binding and DNA strand breaks in vitro.

Chromium(VI) is a known human carcinogen which requires intracellular reduction for activation. Ascorbate (vitamin C) has been reported to function as a major reductant of Cr(VI) in animals and cell culture systems. The reaction of Cr(VI) with varying concentrations of ascorbate was studied under physiological conditions in vitro in order to determine the types of reactive intermediates produced and to evaluate the reactivity of these intermediates with DNA. Reactions of 1.8 mM Cr(VI) with 0-18 mM ascorbate at pH 7.0 in N-(2-hydroxyethyl)piperazine-N'-2-ethanesulfonic acid (HEPES; 0.10 M) and …

Metabolic fate of chromium compounds. I. Comparative behavior of chromium in rat administered with Na251CrO4 and 51CrCl3.

Comparative metabolic fate of labelled chromium chloride and sodium chromate and interaction of these compounds in the rat liver and blood were investigated after their oral and intravenous administration. Gastrointestinal absorption of both compounds was below 1% of the oral dose, but trivalent chromium showed higher radioactivity than the hexavalent form in rats (biological half-life: CrCl3 91.79 days, Na2CrO4 22.24 days). The higher residual activity of the trivalent chromium was also observed after intravenous administration. Both forms of chromium were excreted more in the urine via the k…

Toxic chromium treatment induce amino-assisted electrostatic adsorption for the synthesis of highly dispersed chromium catalyst.

Removal of toxic Cr (VI) from aqueous solutions using silicon-based adsorbents has been widely investigated. Meanwhile, contradictory between highly dispersed active Cr species and high Cr loading over commercial Cr-based catalyst was inevitable. In this work, amino-assisted electrostatic adsorption from toxic Cr (VI) treatment was developed to prepare highly dispersed Cr oxides catalysts supported on MCM-41. The Cr loading was as high as 15 wt%, and structure characters of the catalysts were well-reserved. As a result, electrostatic adsorption and subsequent complexation from negatively charg

Chromium(II) and chromium(III) complexes supported by tris(2-pyridylmethyl)amine: synthesis, structures, and reactivity.

This report describes the synthesis, structural characterization, and polymerization behavior of a series of chromium(II) and chromium(III) complexes ligated by tris(2-pyridylmethyl)amine (TPA), including chromium(III) organometallic derivatives. For instance, the combination of TPA with CrCl(2) yields monomeric (TPA)CrCl(2) (1). A similar reaction of CrCl(2) with TPA, followed by chloride abstraction with NaBPh(4) or NaBAr(F)(4) (Ar(F) = 3,5-(CF(3))(2)C(6)H(3)), provides the weakly associated cationic dimers [(TPA)CrCl](2)[BPh(4)](2) (2A) and [(TPA)CrCl](2)[BAr(F)(4)](2) (2B), respectively. X…

The serum level was 1.4 ng/ml (normal 0.05-0.4). The urine chromium excretion was 1.27 micrograms/day (normal 0.22). The TPN regimen (unsupplemented with Cr) provided 4 micrograms/day. Normal Cr intake is about 60 micrograms/day with 0.4% absorption (net 0.24 microgram/day). We conclude that Cr contamination of stan …

Characteristics of chromium-allergic dermatitis patients prior to regulatory intervention for chromium in leather: a questionnaire study.

BACKGROUND: Chromium-tanned leather articles currently constitute the most important cause of contact allergy to chromium in Denmark. A regulation on the content of hexavalent chromium in leather was adopted in November 2013 by the EU member states. OBJECTIVES: To characterize patients with chromium allergy and their disease, to serve as a baseline for future studies on the potential effect of the new regulation on chromium in leather. METHODS: A questionnaire case-control study was performed on 155 dermatitis patients with positive patch test reactions to potassium dichromate and a matched …

Efficacy of chromium(III) supplementation on growth, body composition, serum parameters, and tissue chromium in rats.

Chromium(III) is often claimed to have a positive effect on body composition, while the responses in researches with supplementation of different chemical form of chromium are various and inconsistent. We have studied the effects of 6 weeks of treatment with three different forms of chromium (300 mug/kg) as chromium chloride, chromium tripicolinate, and chromium nanocomposite (CrNano) on growth, body composition, serum parameters, and tissue chromium in rats. The supplementation of CrNano significantly increased average daily gain, food efficiency, and lean body mass and decreased fat mass and…

The effect of remediation of chromium waste sites on chromium levels in urine of children living in the surrounding neighborhood.

Exposure to chromium was assessed for 40 children living near chromium waste sites. Sampling was conducted in one Jersey City, New Jersey neighborhood during the summer and fall of 1991. Household dust samples from residences and urine samples from children living near chromium waste sites were collected and analyzed for chromium. During the summer and fall visits when the samples were collected, the children were also interviewed about lifestyle/activity patterns. Comparisons were made with similar samples collected from children and homes in other areas of New Jersey outside of Hudson County…

Chromium bond detection in isolated erythrocytes: a new principle of biological monitoring of exposure to hexavalent chromium.

Internal stress to chromium is only relevant in occupational medicine if it is due to the handling of hexavalent chromium. Cr(VI) ions, after uptake by inhalation or percutaneously are carried in the blood plasma and penetrate--depending on the concentration--into the erythrocytes. Due to the intracellular reduction to Cr(III) and the concurrent intracellular protein binding, the erythrocytes represent an easily accessible target organ for quantitative chromium determination after occupational exposure to Cr(VI) compounds. The results of an earlier experimental study indicate that human plasma…

The reduction of chromium (VI) to chromium (III) by glutathione: an intracellular redox pathway in the metabolism of the carcinogen chromate.

The capacity of glutathione (GSH) to reduce Cr(VI) to Cr(III) in vitro was investigated. The reaction was determined spectrophotometrically … ubations of prepared lysates (as opposed to intact cells) with 10 mM Na2 51CrO4 markedly raised the chromium content of low-molecular fractions (probably GSH-Cr-complexes), probably indicative of a role of GSH in the intra-cellular reduction of Cr(VI) to Cr(III), the latter being regarded as the ultimately toxic species of this metal.

Dialysis treatment of acute chromium intoxication and comparative efficacy of peritoneal versus hemodialysis in chromium removal.

External burns with chromic acid have been fatal even in cases with corrosions covering less than 10% of the body surface area. We observed a 19-year-old man with third-degree burns and chromium intoxication following accidental contact of both legs with chromium acid solution. The initial course was characterized by serum chromium levels known to be usually lethal (220 micrograms/100 ml), complete anuria, hepatic damage and progressive anemia. Aggressive peritoneal dialysis with a total duration of 252 h between the 4th and 22nd day after exposure caused a progressive decrease in serum chromi…

Tricarbon-yl(η-flavone)chromium(0).

In the title compound, [Cr(C(15)H(10)O(2))(CO)(3)], the Cr(CO)(3) unit exhibits a three-legged piano-stool conformation. The chromium metal centre is coordinated by the phenyl ring of the flavone ligand [Cr-(phenyl centroid) distance = 1.709 (1) Å]. The ligand is approximately planar, the dihedral angles between the γ-pyrone ring and the phenyl ring and between the γ-pyrone and the phenyl-ene ring being 2.91 (5) and 3.90 (5)°, respectively. The mol-ecular packing shows π-π stacking between the flavone ligands of neighbouring mol-ecules.

Oxidative stress and chromium(VI) carcinogenesis.

Cr(VI)-containing compounds are known carcinogens that are present in industrial settings and in the environment. The biological mechanisms responsible for the initiation and pregression of diseases resulting from exposure to Cr(VI) are not fully understood. In the last two decades, there has been increasing evidence of the correlation between Cr(VI)-induced generation of reactive oxygen species and carcinogenic actions. This article summarizes the current literature on Cr(VI)-induced generation in various chemical and biological systems. This article also covers the relationship between Cr(VI

Crystallographic evidence for chromium-platinum interaction.

Heteronuclear Pt-Cr paddlewheel complexes with significant Pt-Cr interaction have been made. They can be interconverted. Upon oxidation, the Pt-Cr distance shortens significantly while other bond lengths remain unchanged. By taking into account the strong axial coordination from the chloride ligand in the oxidized compound, we suspect that the removed electron upon oxidation is probably from a Pt-Cr orbital that is significantly antibonding in nature.

Blood protein interactions with chromium surfaces.

Protein adsorption, contact activation, and complement activation were studied on thin evaporated films of chromium (Cr) in vitro. The surfaces were, prior to the experiments, cleaned in either ethanol and water, or in a basic peroxide solution (RCA standard clean 1, SC-1). Surface spectroscopic studies of the outermost oxides showed a significant reduction of carbon contaminants after washing in SC-1 but also suggested an increase in the oxidation state as compared with the ethanol-washed surfaces. In situ ellipsometry combined with antibody techniques was used to determine protein deposition…

[Molecular effects of chromium compound activity].

Chromium(VI) compounds produce a variety of DNA damage such as DNA single strand breaks, alkali-labile sites and DNA-protein cross-links in vivo and in cultured cells. Chromium(III) compounds bound to isolated DNA and proteins. Cr(VI) readily entered cell through general anion channels. In contrast, Cr(III) did not readily cross cell membranes. Inside cells Cr(VI) is reduced through intermediates to Cr(III) by cellular reductants. Reactive intermediates formed during intracellular Cr(VI) reduction might be responsible for some chromate genotoxicity. Ascorbic acid decreases chromate induced alk…
