Antithrombin prevents apoptosis by regulating inflammation in the liver in a model of cold ischemia/warm reperfusion injury.

BACKGROUND/AIMS: Hepatic ischemia-reperfusion injury is a major problem in liver surgery. To modulate the complex process of inflammation, additional drugs to add to well-defined organ preserving solutions have been sought. The aim of the current study was to investigate the additive potential of antithrombin (AT) in liver preservation. METHODOLOGY: Female Wistar rats were randomized into four groups: sham (Group I), experiment model (Group II), and treatment groups with AT (250U/kg) administration systematically (Group III) or locally (Group IV) before hepatectomy. UW solution was used for l

Military cold injury during the war in the Falkland Islands 1982: an evaluation of possible risk factors.

Throughout the history of war, there have been many instances when the cold has ravaged armies more effectively than their enemies. Delineated risk factors are restricted to negro origins, previous cod injury, moderate but not heavy cigarette smoking and the possession of blood group O. No attention has been directed to the possibility that abnormal blood constituents could feasibly predispose to the development of local cold injury. This study considers this possibility and investigates the potential contribution of certain components of the circulating blood which might do so. Three groups o…

Lumbar sympathectomy attenuates cold allodynia but not mechanical allodynia and hyperalgesia in rats with spared nerve injury.

UNLABELLED: In certain patients with neuropathic pain, the pain is dependent on activity in the sympathetic nervous system. To investigate whether the spared nerve injury model (SNI) produced by injury to the tibial and common peroneal nerves and leaving the sural nerve intact is a model for sympathetically maintained pain, we measured the effects of surgical sympathectomy on the resulting mechanical allodynia, mechanical hyperalgesia, and cold allodynia. Decreases of paw withdrawal thresholds to von Frey filament stimuli and increases in duration of paw withdrawal to pinprick or acetone stimu

Studies on the mechanism of shock. The effect of cocaine and theophylline on the temperature response to injury in the cold-acclimated rat.

The effects of cocaine and theophylline on the changes in core temperature, O2 consumption and the rate of heat loss from the tail were studied in 20° and 3° acclimated rats during and after a 4-hr. period of bilateral hindlimb ischaemia at an environmental temperature of 20°. Cocaine (7 mg. base per kg. body wt. intravenously) had no effect on injured 20° acclimated rats but increased the core temperature in both injured and uninjured 3° acclimated ones. The changes in O2 consumption were unaffected and heat loss from the tail decreased. Cocaine acted mainly by increasing peripheral vasoconst

Machine perfusion modulates cold-preservation injury in kidney transplantation: IDEAL stage 4 United States population cohort study.

Hypothermic machine perfusion (HMP) has surged in popularity for donor kidney preservation. Continuous HMP (cHMP) has shown clear benefits over static cold storage (SCS), whereas randomized trials on short-duration end-ischemic-HMP (eiHMP) have not. We assessed whether HMP modulates injury from increasing cold-preservation time (CPT) and analyzed the impact of HMP on short- and long-term outcomes, using Organ Procurement and Transplantation Network data (OPTN; 2010-2024) on single-kidney-transplant recipients (n = 137 835). Multivariable nonlinear (restricted cubic spline) regression with inte

A cold-inducible RNA-binding protein (CIRP)-derived peptide attenuates inflammation and organ injury in septic mice.

Cold-inducible RNA-binding protein (CIRP) is a novel sepsis inflammatory mediator and C23 is a putative CIRP competitive inhibitor. Therefore, we hypothesized that C23 can ameliorate sepsis-associated injury to the lungs and kidneys. First, we confirmed that C23 dose-dependently inhibited TNF-α release, IκBα degradation, and NF-κB nuclear translocation in macrophages stimulated with CIRP. Next, we observed that male C57BL/6 mice treated with C23 (8 mg/kg BW) at 2 h after cecal ligation and puncture (CLP) had lower serum levels of LDH, ALT, IL-6, TNF-α, and IL-1β (reduced by ≥39%) at 20 h after

The mitochondria-targeted antioxidant mitoquinone protects against cold storage injury of renal tubular cells and rat kidneys.

The majority of kidneys used for transplantation are obtained from deceased donors. These kidneys must undergo cold preservation/storage before transplantation to preserve tissue quality and allow time for recipient selection and transport. However, cold storage (CS) can result in tissue injury, kidney discardment, or long-term renal dysfunction after transplantation. We have previously determined mitochondrial superoxide and other downstream oxidants to be important signaling molecules that contribute to CS plus rewarming (RW) injury of rat renal proximal tubular cells. Thus, this study's pur

Severe Intravascular Hemolysis and Acute Kidney Injury Triggered by Mycoplasma-Associated Cold Agglutinin Disease: A Case Report.

INTRODUCTION: Mycoplasma pneumoniae infection commonly causes mild respiratory illness, but cold agglutinin syndrome (CAS) is an uncommon extrapulmonary complication. CAS typically results in mild, predominantly extravascular hemolysis; however, severe intravascular hemolysis and acute kidney injury (AKI) are rare. CASE PRESENTATION: A 59-year-old woman presented with profound weakness, jaundice, dark urine, and anuria following a mild upper respiratory illness. Laboratory evaluation revealed severe intravascular hemolysis, markedly elevated LDH activity, undetectable hemoglobin (Hb) level, h

Ex vivo pretreatment of donor organ with siRNA nanoparticles attenuates cold ischemia-reperfusion injury in cardiac transplantation.

Heart transplantation is an optimal therapeutic regimen for terminal-stage cardiac failure. However, cold ischemia-reperfusion injury (CIRI) remains an unavoidable and outstanding challenge, which is a significant obstacle to early graft dysfunction and long-term survival. Blockage of complement, apoptosis, and inflammation by small interfering RNA is considered a strategy for attenuating CIRI and protecting cardiac function. However, their delivery to the donor organ is still a serious challenge due to the polyanionic nature and high molecular weight properties. Here, we have designed a novel

Protective Effects of Quercetin and Sucrose on Cold Preservation Injury in Porcine Kidneys Donated After Cardiac Death.

BACKGROUND The University of Wisconsin (UW) solution is the gold standard for kidney preservation. Quercetin (QE), a widely used flavonoid, possesses strong antioxidant, anti-inflammatory, and anti-apoptotic properties. Sucrose (Suc) has shown beneficial effects as a supplement. The aim of this study was to investigate the protective effects of QE and Suc against cold injury in porcine kidneys and to evaluate whether their use improves ischemia-reperfusion (I/R) injury after simple CS. MATERIAL AND METHODS Kidney grafts were procured after 30 min of warm ischemia, followed by preservation unde…

Splenic tissue injury and physiological response mechanisms in juvenile yellowfin tuna (Thunnus albacares) under acute cold stress.

Abnormal seawater temperatures driven by global climate change are profoundly disrupting the physiological homeostasis and immune regulation of marine fish. As a warm-blooded pelagic species, yellowfin tuna (Thunnus albacares) possesses partial thermoregulatory capability but still experiences significant physiological stress under abrupt cold exposure. The spleen, a key immune and metabolic organ, is highly sensitive to temperature fluctuations and serves as a critical indicator of cold stress effects. In this study, juvenile yellowfin tuna were subjected to cold stress at 24 °C (LT group) an

Cold-stored whole blood and platelet counts in severe acute injury: A comparison of four retrospective cohorts.

BACKGROUND: Low-titer group O whole blood (LTOWB) is attractive for acute trauma care as it delivers concentrated and balanced hemostatic resuscitation in single large bags. Whether cold-stored LTOWB can sustain platelet counts is unclear. STUDY DESIGN AND METHODS: Four cohorts of trauma patients-three historic, one retrospective-were identified by their urgency of blood use. Admission and all subsequent platelet counts over the first 24 h of care were compared with t-tests. The cohorts were as follows: 1292 patients at Maryland Shock Trauma as described by Stansbury and colleagues in 2013; 3

Cold-induced injury to lung epithelial cells can be inhibited by iron chelators - implications for lung preservation.

OBJECTIVES: Hypothermia has been shown to lead to cell death via an iron-dependent formation of reactive oxygen species (ROS) in diverse cell types. The susceptibility of lung cells to this injury is unknown - but is of great interest as oxygen is used in lung preservation. Therefore, we studied whether preservation injury to lung epithelial cells can be influenced by iron chelators. METHODS: A549 lung epithelial cells were preserved at 4°C for varying periods in either cell culture medium (Dulbecco's Modified Eagle Medium, DMEM), Bretschneider histidine-tryptophan-ketoglutarate (HTK), modifi

Causes and Consequences of Freezing Cold Injuries in the Norwegian Armed Forces from the Soldier's Perspective-A Qualitative Study.

Freezing cold injury (FCI) is a localized injury caused by prolonged exposure to sub-zero temperatures, posing a significant health risk to military personnel operating in extreme climates. This study aimed to explore how FCI affects the health and lives of soldiers in the Norwegian Armed Forces (NAF), addressing the broader context of its impact on military readiness and the personnel's well-being. Sixteen participants with a history of grade 2 FCI were recruited through the Norwegian Armed Forces Health Registry and interviewed. Systematic content analysis was used to identify key themes rel…

Cold burns as a result of cosmetic cryolipolysis: An emerging concern from the NSW Statewide Burn Injury Service.

Cold burns are an uncommon mechanism of burn injury in Australia, where environmental exposure to freezing temperatures is rare. Aetiological research shows that most likely causes are related to intentional injury or industrial accidents1 Cryolipolysis, a cosmetic technique advertised as 'fat freezing', has recently emerged as a method of fat reduction and body contouring. Whilst perceived as safe4,5, this study presents the world's first case series on cold burns sustained from cryolipolysis. A retrospective review was undertaken of the New South Wales Statewide Burn Injury Service (SBIS) Re
