Incidence of pyometra in colony-raised beagle dogs.

Incidence of pyometra observed in our colony-raised beagle dogs over a 12-year period is described. Pyometra was observed in 25 female dogs of more than 4 years old, frequently at 8-11 years, with the average age of onset 9.36 +/- 0.38 years. The incidence of the disease was 15.2% of the female dogs (n = 165) more than 4 years old. Clinical findings useful in the diagnosis of this disease included an excretion of pus from the cervix, rapid increase in leukocyte count, and enlarged uterus as revealed by radiography. The relationships of delivery, estrus cycles to this disease are discussed. Ova

Biology and action of colony--stimulating factor-1.

Colony-stimulating factor-1 (CSF-1), also known as macrophage colony-stimulating factor, controls the survival, proliferation, and differentiation of mononuclear phagocytes and regulates cells of the females reproductive tract. It appears to play an autocrine and/or paracrine role in cancers of the ovary, endometrium, breast, and myeloid and lymphoid tissues. Through alternative mRNA splicing and differential post-translational proteolytic processing, CSF-1 can either be secreted into the circulation as a glycoprotein or chondroitin sulfate-containing proteoglycan or be expressed as a membrane

Granulocyte colony-stimulating factor in amniotic fluid.

OBJECTIVE: The purpose of this study was to determine if granulocyte colony-stimulating factor (G-CSF) is normally present in amniotic fluid and then to determine if amniotic-fluid G-CSF levels are affected by labor and intrauterine infection. METHODS: Amniotic fluid was collected from 35 patients in 4 groups: no labor, early labor, late labor, and labor plus chorioamnionitis. G-CSF levels were measured by enzyme-linked immunosorbent assay (ELISA). RESULTS: The mean amniotic-fluid G-CSF concentrations prior to labor were lower than during labor (0.49 +/- 0.25 ng/ml for prior to labor vs. 1.8

The biology and action of colony stimulating factor-1.

Colony stimulating factor 1 (CSF-1) is a growth factor for mononuclear phagocytic cells. Through alternative mRNA splicing and differential post-translational proteolytic processing, CSF-1 can either be secreted into the circulation as a glycoprotein or chondroitin sulfate-containing proteoglycan or expressed as a membrane-spanning glycoprotein on the surface of synthesizing cells. The discovery that the osteopetrotic (op/op) mutant mouse possesses an inactivating mutation in the CSF-1 gene has greatly contributed to our understanding of CSF-1 biology. CSF-1 directly regulates some non-mononuc

Retrospective study of diseases in a captive Lemming Colony.

Fifty-four ill or nonproductive lemmings (Dicrostonyx spp.) were evaluated for signs, lesions and causes of disease for 5 yr in a domestic colony. Parasitic granulomas caused by Encephalitozoon cuniculi were the most common finding and were seen in 22 lemmings. The disease was characterized by circling and torticollis with granulomas in many tissues, especially the central nervous system. Suppurative otitis occurred in 12 lemmings and was associated with Klebsiella pneumonia infection; circling was the common sign. Hepatic microabscesses were present in seven lemmings but a cause was not ident

Proliferation of haematopoietic colonies in serum-free cultures.

In eight patients with polycythaemia vera and 20 normals, mononuclear cells of the peripheral blood were cultured in methylcellulose with and without exogenously added erythropoietin. The cultures were set up with fetal calf serum and with a serum-free medium of unsaturated fatty acids, lecithin and cholesterol. In patients with polycythemia vera, we found a proliferation of haematopoietic colonies without erythropoietin in both culture systems which could not be seen in normals. The proliferation of endogenous colonies in serum-free media suggests that polycythaemia vera is a clonal disorder

Granulomonocyte colony-forming cells in cord blood.

The number of colony-forming cells (CFC) in cord blood has been found to be widely dispersed (478 +/- 1003/ml or 3-5200). However, a few samples (7 of 37 in this series) contained more than 478 CFC/ml and in four of these cases a history of prenatal hypertension or infection was found. In the other 30 cases the mean number of CFC was 140.6 +/- 116.9/ml blood (3-390), which, however, remains much higher than data found for adult blood. In these 30 cased the mean number of cord blood CFC in newborns of less than 37 weeks gestational age was found significantly higher (194.2 +/- 125.8/ml) than in

Erythroid colony growth in congenital hypoplastic anemia.

Four children with congenital hypoplastic anemia (Diamond-Blackfan syndrome) and 30 control children with normal erythropoiesis were studied by a cell culture method in which human marrow, grown in a plasma clot, responds to added erythropoietin (EPO) with the appearance of discrete colonies of nucleated erythroid cells. The colonies arise from EPO-responsive stem cells and are not related to the number of morphologically identifiable marrow erythroids plated. Results of studies on control marrow indicated that without EPO there was little or no colony formation. Increasing EPO doses or nuclea

Hemopoietic spleen colony studies. II. Erythropoiesis.

The polycythemic repression of erythropoiesis and the restoration of erythropoiesis by specific stimulation were studied in the spleen colony system in irradiated mice. 1. A 5 day period of erythropoietin stimulation (exogenous erythropoietin) or repression (polycythemia) of the bone marrow donor only, does not significantly alter the number or type of colonies formed by the transplanted marrow cells. 2. Erythropoietin stimulation did not alter the number or type of endogenous colonies formed in mice receiving 580 R. Erythropoietin repression (polycythemia) markedly reduced the growth but not

Hapten-specific murine colony-forming B cells. II. Delineation of a tolerogen-sensitive subpopulation of colony-forming B cells.

B cell subpopulations were studied by using B cell cloning procedures and an in vitro tolerance induction model. Fluorescein- (FL) specific B cells from normal spleens were isolated by using FL gelatin plates and were then cultured in semisolid agar in the presence or absence of tolerogen. Hapten-specific cells grew in soft agar to form discrete colonies. Colony growth is dependent on "mitogens" present in agar, sheep red blood cells (SRBC), and lipopolysaccharide (LPS). For example, SRBC plus LPS potentiate the growth of an increased number of colony-forming B cells (CFU-B) compared to either

The role of haematopoietic growth factors granulocyte colony-stimulating factor and granulocyte-macrophage colony-stimulating factor in the management of drug-induced agranulocytosis.

Drug-induced agranulocytosis (neutrophil count <0.5 x 10(9)/l) is a rare haematological complication with an incidence of no more than 10 cases per million inhabitants per year in Europe. Over the past few years there has been a steady decline in mortality rate, (currently at <5%), which can be partly explained by earlier recognition and the improved clinical management of associated intercurrent infections that may lead to severe sepsis if left untreated. The true impact of the use of haematopoietic growth factors, such as granulocyte colony-stimulating factor (G-CSF) and granulocyte-macropha
