Suppression of motor cortical excitability in anesthetized rats by low frequency repetitive transcranial magnetic stimulation.

Repetitive transcranial magnetic stimulation (rTMS) is a widely-used method for modulating cortical excitability in humans, by mechanisms thought to involve use-dependent synaptic plasticity. For example, when low frequency rTMS (LF rTMS) is applied over the motor cortex, in humans, it predictably leads to a suppression of the motor evoked potential (MEP), presumably reflecting long-term depression (LTD) -like mechanisms. Yet how closely such rTMS effects actually match LTD is unknown. We therefore sought to (1) reproduce cortico-spinal depression by LF rTMS in rats, (2) establish a reliable a…

Visual pattern adaptation in subjects with photoparoxysmal EEG response: evidence for increased visual cortical excitability.

PURPOSE: Photosensitivity, or photoparoxysmal response (PPR), is an abnormal EEG reaction to intermittent photic stimulation (IPS), consisti … reffect (TAE) in the PPR group only. CONCLUSIONS: This study provides further evidence of enhanced cortical excitability in subjects with photosensitivity, which is likely to reflect changes in excitatory neurotransmission.

Assessing cortical excitability in migraine: reliability of magnetic suppression of perceptual accuracy technique over time.

OBJECTIVES: To examine test-retest reliability of magnetic suppression of perceptual accuracy (MSPA) prior to its use as a marker of cortical excitability in a trial of migraine prophylactic agents. BACKGROUND: MSPA is a relatively novel avenue of research in headache, providing an opportunity to study cortical responsiveness objectively and noninvasively. However, little is known about the reliability of magnetic stimulation protocols such as MSPA in longitudinal research designs. METHODS: We tested 10 healthy headache-free volunteers who had no family history of migraine. In 54 trials, the…

Task-specific impairment of motor cortical excitation and inhibition in patients with writer's cramp.

Abnormalities in motor cortical excitation and inhibition have been reported in patients with writer's cramp, at rest and during muscle activation. We were interested in whether such abnormalities might be task-specific and depended on the type of movement task used to activate the dystonic hand. We therefore assessed motor-evoked potentials (facilitation/rest MEP amplitude ratio) and duration of the cortical silent period (CSP) from the right first dorsal interosseus (FDI) muscle to transcranial magnetic stimulation (TMS) in 10 patients with writer's cramp and in 10 healthy volunteers perform

Altered motor cortical excitability to magnetic stimulation in a patient with a lesion in globus pallidus.

A 27-year-old man with Behçet's disease presented with dyskinesia of the right hand and leg, but without paresis. MRI showed a discrete lesi … n excitability of the left motor cortex. We conclude that it is possible to detect changes of motor cortical excitability caused by dysfunction of some inputs from the basal ganglia by using the double cortical stimulation technique here described.

Individual and Sex Differences in Behavior and Cortical Excitability in the Rat Valproate Model of Autism.

The rodent prenatal valproate (VPA) treatment is a widely used animal model of idiopathic autism spectrum disorder (ASD). However, the presence of autistic-like symptoms is highly variable in treated offspring. The disruption of the excitation-inhibition balance of certain brain areas has been proposed as a main feature in both human ASD and the VPA model. The current study presents a detailed analysis of neural development, diverse behaviors, and neocortical excitability in a high number of individually identified VPA-treated rat offspring of both sexes. Neocortical excitability was assessed …

Acute changes in cortical excitability in the cortex contralateral to focal intracerebral hemorrhage in the swine.

Injury to the cerebral cortex results in functional deficits not only within the vicinity of the lesion but also in remote brain regions sharing neuronal connections with the injured site. To understand the electrophysiological basis of this phenomenon, we evaluated the effects of a focal intracerebral hemorrhage (ICH) on cortical excitability in a remote, functionally connected brain region. Cortical excitability was assessed by measuring the somatic evoked potential (SEP) elicited by electrical stimulation of the swine snout, which is somatotopically represented in the rostrum area of the pr…

Abnormalities of the balance between inhibition and excitation in the motor cortex of patients with cortical myoclonus.

Patients with cortical myoclonus may have purely focal or multifocal jerks, or they may have additional bilateral or generalized jerks, suggesting the spread of excitatory myoclonic activity between the cerebral hemispheres and across the sensorimotor cortex. The factors contributing to this spread of activity were investigated in 10 patients with multifocal cortical myoclonus and eight patients with multifocal and bilateral or generalized cortical myoclonus. The two groups were termed 'non- spreaders' and 'spreaders' respectively. Eight of the patients were also epileptic. Motor thresholds to

C9orf72-derived dipeptide repeat proteins poly-PR disrupt membrane excitability and synaptic function in cortical neurons.

Amyotrophic lateral sclerosis (ALS) is one of the most fatal neurodegenerative disease, with the most common genetic form of the ALS is associated with hexanucleotide GGGGCC repeat expansions in the first intron of C9orf72 gene. Cortical hyperexcitability is one of the symptoms reported in several forms of ALS and implicated as a cause of neuronal death, however, the underlying mechanisms are still unclear. The dipeptide repeat (DPR) proteins produced from hexanucleotide repeat expansion have been shown toxic to neurons and induce cellular damages. In this study, we explore relationships betwe

Parietal rTMS Induced Changes in Cortical Excitability in Patients With Minimally Conscious State Using TMS-EEG.

OBJECTIVE: To verify the effectiveness of the parietal repetitive transcranial magnetic stimulation (rTMS) and take advantage of TMS-EEG to assess cortical excitability in patients with minimally conscious states (MCS). METHODS: We enrolled 10 MCS patients who received 10 sessions of 10 Hz rTMS on the parietal cortex for 10 consecutive days and then 10 days of sham stimulation after a 14-day wash-out period. The Coma Recovery Scale-Revised (CRS-R) and TMS-EEG were used to assess the levels of consciousness and cortical excitability before and after active and sham stimulation, respectively. …

American tackle football is associated with high rates of concussion, leading to neurophysiological disturbances and debilitating clinical s … with increased aperiodic exponents in superior frontal cortices, indicating a relative reduction in cortical excitability, which accounted for effects on raw delta and gamma power and was associated with worse cognitive concerns across participants. Subtler effects of non-concussive head-impact exposure on neurophysiology were similar to those of concussion. These find …

Rod-shaped microglia interact with neuronal dendrites to attenuate cortical excitability during TDP-43-related neurodegeneration.

Microglia, the principal immune cells of the central nervous system, have emerged as important players in sensing and regulating neuronal activity. While microglial activation is a hallmark in neurodegeneration, the specific role of microglia in disease-related cortical excitability remains unknown. Utilizing multichannel probe recordings and longitudinal in vivo calcium imaging, we observed neuronal hyperactivity at the initial stage of disease progression in a mouse model of TAR DNA-binding protein 43 (TDP-43) neurodegeneration (rNLS8, regulated nuclear localization sequence-deleted human TD…

A Celsr3 Mutation Linked to Tourette Disorder Disrupts Cortical Dendritic Patterning and Striatal Cholinergic Interneuron Excitability.

Tourette Disorder (TD) is a prevalent neurodevelopmental condition characterized by chronic motor and vocal tics. A mechanistic understanding of both the genetic etiology and brain pathophysiology remains poor. To gain insight into the molecular underpinnings of TD, we have generated a novel mouse model expressing an orthologous human mutation in CELSR3, a high-confidence TD risk gene. This putative damaging de novo variant, R774H, causes an amino acid substitution within the fifth cadherin repeat. Unlike previous Celsr3 TD models and Celsr3 constitutive null mice, mice homozygous for the R774

Relationship between GABA levels and task-dependent cortical excitability in children with attention-deficit/hyperactivity disorder.

OBJECTIVE: Compared to typically developing (TD) peers, children with attention deficit hyperactivity disorder (ADHD) manifest reduced short interval cortical inhibition (SICI) in the dominant motor cortex measured with transcranial magnetic stimulation (TMS). This multimodal study investigates the inhibitory neurophysiology and neurochemistry by evaluating the relationship between SICI and γ-amino butyric acid (GABA+) levels, measured with magnetic resonance spectroscopy (MRS). METHODS: Across two sites, 37 children with ADHD and 45 TD children, ages 8-12 years, participated. Single and pair

Decreased interhemispheric connectivity and increased cortical excitability in unmedicated schizophrenia: A prefrontal interleaved TMS fMRI study.

BACKGROUND: Prefrontal abnormalities in schizophrenia have consistently emerged from resting state and cognitive neuroimaging studies. However, these correlative findings require causal verification via combined imaging/stimulation approaches. To date, no interleaved transcranial magnetic stimulation and functional magnetic resonance imaging study (TMS fMRI) has probed putative prefrontal cortex abnormalities in schizophrenia. OBJECTIVE: /Hypothesis: We hypothesized that subjects with schizophrenia would show significant hyperexcitability at the site of stimulation (BA9) and decreased interhe
