Persistence of injectable collagen in human urethra: case report.

A polypoid lesion was found near the bladder neck during cystoscopy in a woman with urinary incontinence who had undergone periurethral collagen (Contigen) injections 3 years before. She had previously received radiation therapy in addition to a radical vulvectomy for vaginal cancer. On transurethral resection of the lesion, particles of unresorbed collagen material extruded from the capsulated suburothelial space. Histologic evaluation verified the material as the foreign collagen. The persistence of glutaraldehyde cross-linked collagen in our patient was much longer than previously reported

Trapping of transition metal-nucleotide complexes in myosin subfragment 1 by cross-linking thiols; divalent transition metal probes of the active site.

Recent experiments [Wells, J., & Yount, R. (1979) Proc. Natl. Acad. Sci. U.S.A. 76, 4966] have shown it is possible to trap MgADP and other nucleotides stably at the active site of myosin by cross-linking two thiol groups. A variety of cross-linking reagents including chelation of the two thiols by cobalt (III) phenanthroline or covalent reaction with N,N'-p-phenylenedimaleimide (pPDM) are effective trapping agents. No trapping of nucleotides occurs in the absence of divalent metals. Thus far Mg2+, Mn2+, Co2+, Ni2+, and Ca2+ but not Zn2+ all function to promote trapping of the 1:1 divalent met…

Tethered benzophenone reagents for the synthesis of photoactivatable ligands.

A new radiolabeled, bifunctional photoaffinity cross-linking reagent, N-succinimidyl p-benzoyl-[2,3-3H2]dihydrocinnamate, has been synthesized in high yield and with high specific activity. This reagent can be used to append the benzophenone photophore to amino groups of small molecules, such as O-aminoalkylinositol polyphosphates and polypeptides. The resulting tritiated photoaffinity labels can be purified and manipulated in ambient light and can be activated at 360 nm.

Influence of pH on radical reactions between kynurenic acid and amino acids tryptophan and tyrosine. Part II. Amino acids within the protein globule of lysozyme.

An acidosis, a decrease of pH within a living tissue, may alter yields of radical reactions if participating radicals undergo partial or complete protonation. One of photosensitizers found in the human eye lens, kynurenic acid (KNA-), possesses pKa 5.5 for its radical form that is close to physiological pH 6.89 for a healthy lens. In this work we studied the influence of pH on mechanisms and products of photoinduced radical reactions between KNA- and amino acids tryptophan (Trp) and tyrosine (Tyr) within a globule of model protein, Hen White Egg Lysozyme (HEWL). Our results show that the rate

Comparison of epithelium-off versus iontophoresis-assisted transepithelial corneal collagen cross-linking in keratoconus: a systematic review and meta-analysis.

BACKGROUND / PURPOSE: Keratoconus is a progressive corneal ectasia characterized by stromal thinning and protrusion, leading to irregular astigmatism and visual impairment. To mitigate these issues, iontophoresis-assisted transepithelial corneal collagen cross-linking (I-CXL) was developed to enhance riboflavin penetration while preserving the epithelium, aiming for comparable biomechanical effects with fewer adverse events. This systematic review and meta-analysis aim to compare the efficacy and safety of iontophoresis-assisted transepithelial corneal collagen cross-linking (I-CXL) and standa

Refractive Index Engineering for Human Corneal Tissue by Femtosecond Laser Two-Photon Cross-Linking.

Clinical refractive surgeries for myopia correction generally entail stromal tissue removal with risks of postoperative complications. NIR femtosecond laser-induced corneal two-photon collagen cross-linking (two-photon CXL) through photochemical effects can lead to localized refractive index (RI) changes in the cornea, promising for non-ablative and non-incisional visual correction. However, previous studies mainly focused on hydrogels and animal corneal tissues, adopting a diffraction grating method that is difficult for biological tissue. Here we systematically investigate femtosecond laser-

[Cross-linking treatment of ultrathin corneas: the ELZA-sub400 protocol : Individualized adaptation of fluence as paradigm shift in CXL treatment of thin corneal conditions].

BACKGROUND: The Dresden corneal cross-linking (CXL) protocol requires a minimum corneal thickness of 400 µm to protect the endothelium from UV (ultraviolet) damage; however, many patients with advanced corneal ectatic disease have thinner corneas and are excluded from treatment. Earlier approaches to circumvent this limitation, hypo-osmolar swelling, contact lens-assisted CXL, epithelial island techniques, showed relevant shortcomings. The ELZA-sub400 protocol takes a fundamentally different approach: the UV fluence is individually adapted to pachymetry. METHODS: This narrative review is base

Use of corneal cross-linking beyond keratoconus: a systemic literature review.

PURPOSE: The success of corneal collagen cross-linking in altering keratoconus' clinical course has driven a search for further uses of this procedure. This literature review aims to analyze the scientific evidence available for the benefit of cross-linking in the management of ophthalmic diseases other than progressive keratoconus or ectasia induced by corneal refractive procedures. METHODS: A systemic literature review. RESULTS: We reviewed 97 studies. We found that collagen cross-linking can limit the progression of several other corneal ectasias, thus reducing and limiting the need for k

Photomodulation of the nucleating activity of a photocleavable crosslinked actin dimer.

The ability to generate substrate concentration jumps through photo-deprotection of amine, carboxyl and phosphate groups has been an important development for investigations of protein activity in complex systems. To broaden the versatility and applications of photo-deprotection techniques for the photomodulation of protein activity we describe the synthesis and characterisation of a reagent for generating free thiol from thioether groups and a related photocleavable, heterobifunctional crosslinking reagent. Chemical and spectroscopic studies of a model thiol protected derivative were used to

Nearest neighbor relationships of the polypeptides in ubiquinone cytochrome c reductase (complex III).

Ubiquinone cytochrome c reductase (complex III) in detergent dispersion has been cross-linked with two reversible cross-linking agents dithiobissuccinimidylpropionate and dimethyl-3.3'-dithiobispropionimidate and the cross-linked products formed have been analyzed by two-dimensional gel electrophoresis. Under mild reaction conditions, polypeptides I and II, II and VI, I and V, and VI and VII were the most prominent subunit pairs seen. With higher levels of reagent, larger aggregates were produced until an aggregate of apparent molecular weight 310 000 was the dominant band on gels. This is the

Substrate Channeling in Compartmentalized Nanoreactors.

Thermo- and photoresponsive nanoreactors based on shell cross-linked micelles (SCMs) for the rhodium-catalyzed asymmetric transfer hydrogenation (ATH) of ketones have been developed from poly(2-oxazoline) triblock terpolymers. The nanoreactors incorporate thermoresponsive poly(2-isopropyl-2-oxazoline) as the hydrophilic corona and are covalently cross-linked with a photoswitchable spiropyran molecule. UV irradiation or changes in temperature trigger morphology switching of the polymer-based nanoreactors that alters the hydrophobicity in separate layers of the SCMs, resulting in dynamic substra

Mass spectrometry reveals the chemistry of formaldehyde cross-linking in structured proteins.

Whole-cell cross-linking coupled to mass spectrometry is one of the few tools that can probe protein-protein interactions in intact cells. A very attractive reagent for this purpose is formaldehyde, a small molecule which is known to rapidly penetrate into all cellular compartments and to preserve the protein structure. In light of these benefits, it is surprising that identification of formaldehyde cross-links by mass spectrometry has so far been unsuccessful. Here we report mass spectrometry data that reveal formaldehyde cross-links to be the dimerization product of two formaldehyde-induced

Cross-linking of human T cell receptor proteins: association between the T cell idiotype beta subunit and the T3 glycoprotein heavy subunit.

Bifunctional cross-linking reagents DSP, DSS, and BSOCOES were used to cross-link 125I-surface-labeled viable T lymphocytes. The cross-linked cells were solubilized in Nonidet-P40, immunoprecipitated with anti-Ti (monoclonal antibody T40/25) or anti-T3 (monoclonal antibodies UCHT-1 or OKT3), and analyzed by SDS-PAGE. With all three cross-linkers, the intact cross-linked products obtained with monoclonal antibody T40/25 from HPB-ALL cells were 20-30 kd heavier than the Ti dimer (Mr 80,000). When the DSP cross-linked product was isolated using either anti-Ti or anti-T3 monoclonal antibodies and …

Mechanism of site-directed protein cross-linking. protein-directed selectivity in reactions of hemoglobin with aryl trimesates.

Site-directed cross-linking of hemoglobin has become an efficient way to produce a structurally defined altered protein with desirable functional properties. The reagent trimesoyl tris(3, 5-dibromosalicylate) (1) introduces a bis amide cross-link derived from the epsilon-amino groups of the side chains of the two beta-Lys-82 residues in human hemoglobin. The basis of its specificity was investigated using a set of analogues of 1 (2-12). There are marked differences in the reaction patterns of these compounds with amino groups in hemoglobin compared to reactions with n-propylamine. The compound

Proteoform-Resolved Cross-Linking Reveals Environment-Dependent Structural Effects of α-Synuclein S129 Phosphorylation.

Intrinsically disordered proteins (IDPs) drive many neurodegenerative disorders, but their structures remain difficult to define because they populate dynamic ensembles that change with the chemical environment. This problem is central for α-synuclein (aSyn), a Parkinson's disease-linked IDP in which S129 phosphorylation is highly enriched in disease-associated aggregates. Although S129 phosphorylation stabilizes a more compact aSyn ensemble in dilute solution, whether this structural effect persists across other biochemical environments has remained unknown. Here, we developed a quantitative

Design of polymer boron crosslinking agents based on scale effects and study of the crosslinking mechanism of guar gum at ultra-low concentrations.

Research on guar gum-based fracturing fluids reveals a scale disparity of approximately ten thousandfold between their molecular chains and conventional boron crosslinkers, leading to pronounced scale and structure effects. Traditional crosslinking theories focus solely on group interactions, lacking a holistic molecular structural perspective. To address this, this study proposes synergistically optimising scale and structural effects to overcome theoretical limitations. Targeting the core issue of low crosslinking efficiency at low concentrations, a novel polymeric boron crosslinking agent,

Endoskeleton-Enhanced Virus-Like Particles (VLPs): Internal Cross-Linking of Physalis Mottle Virus and Stability Tests.

Virus-like particles (VLPs) often exhibit favorable chemical, thermal, and mechanical stability compared to other biologic materials. Nonetheless, enhancing their stability remains crucial for advancing VLP-based materials toward practical and commercial use. To address this, we introduced an "endoskeleton-armoring" approach aimed at reinforcing VLP stability. Using the VLP from physalis mottle virus (PhMV) as a model system, we created an internal polymer backbone by covalently linking the coat proteins within the capsid using a maleimide-PEG15-maleimide crosslinker. This contrasts with nativ
