The effect of peroral administration of toxic cyanobacteria on laboratory rats (Rattus norvegicus var. alba).

OBJECTIVES: The toxic cyanobacteria are a serious problem for water supply systems, recreation, and agriculture. Cyanobacteria produce numerous bioactive compounds including microcystins - the most studied cyanobacterial hepatotoxins. Only rare studies addressed realistic situation, i.e. impact of MCs accumulated in the fish tissues on the overall physiology. The aim of the present study was to provide a model simulation of the simple food chain for evaluation of impacts of cyanobacteria on the rat physiology under different exposure scenario. METHODS: Experimental rats were fed with food wit…

Combined exposure to hydrogen peroxide and light--selective effects on cyanobacteria, green algae, and diatoms.

The selective toxicity of H2O2 was investigated to develop a potential tool for limiting cyanobacterial blooms and to better understand the occurrence of cyanobacteria and other phytoplankton species in relation to reactive oxygen species in surface waters. The cyanobacterium Microcystis aeruginosa, the green alga Pseudokirchneriella subcapitata, and the diatom Navicula seminulum were tested under pulse exposure to H202 in the dark and at various irradiances. H2O2 was decomposed at rates depending on algal species and was proportional to irradiance. The cyanobacterium was affected by H202 at 1…

Alr2204-Alr2205 form a functional modular thioredoxin system that enhances oxidative stress tolerance in cyanobacteria.

The cysteine-mediated catalytic reactions are essential for redox homeostasis in photosynthetic organisms. In the genome of cyanobacterium Anabaena PCC 7120 (Anabaena), the genes encoding thioredoxin reductase (TR; alr2204) and the atypical thioredoxin (Trx; alr2205) are contiguously located, but whether these proteins function as a redox pair remains unknown. Interestingly, Alr2204 and Alr2205, respectively, show high homology to the TR and Trx domains of the chloroplast NADPH-thioredoxin reductase C (NTRC). To delve into its possible role, Alr2204 was overexpressed in Anabaena. The resulting

Exploring thylakoid emergence: evolution of membrane biogenesis and photosystem II assembly in early-diverging cyanobacteria.

Thylakoid membranes (TM) in cyanobacteria and chloroplasts host the light-dependent reactions of oxygenic photosynthesis. Gloeobacterales, the earliest-diverging cyanobacterial lineage, lack TM and perform photosynthesis in the cytoplasmic membrane (CM), representing an ancestral state relative to other cyanobacteria (Phycobacteria). This study investigates the evolutionary origin of TM. Phylogenomic analyses were performed across a phylogenetically diverse set of cyanobacteria, including extensive representation of basal lineages (Gloeobacterales, Thermostichales, Gloeomargaritales, and Pseud…

Cluster of differentiation 36-engineered photosynthetic cyanobacteria enable immunometabolic reprogramming in triple-negative breast cancer.

Triple-negative breast cancer (TNBC) exhibits profound resistance to immunotherapy, largely due to a tumor microenvironment constrained by d … en availability to reprogram the TNBC microenvironment. By engineering photosynthetic Synechococcus cyanobacteria with surface-displayed anti-CD36 antibodies, we construct a dual-functional system (Syne@aCD36) capable of blocking CD36-mediated fatty acid uptake while generating oxygen in situ under light stimulation. This coordinated regulation alleviates lipid …

Sediment potentially controls in-lake phosphorus cycling and harmful cyanobacteria in shallow, eutrophic Utah Lake.

Lakes worldwide are impacted by eutrophication and harmful algal or cyanobacteria blooms (HABs) due to excessive nutrients, including legacy P released from sediments in shallow lakes. Utah Lake (northern Utah, USA) is a shallow lake with urban development primarily on the east side of the watershed, providing an opportunity to evaluate HABs in relation to a gradient of legacy sediment P. In this study, we investigated sediment composition and P concentrations in sediment, pore water, and the water column in relation to blooms of harmful cyanobacteria species. Sediments on the east side of the…

cKAT acetylation of the CO2 hydration protein ChpX regulates the CO2 concentrating mechanism in cyanobacteria.

Lysine acetylation (Kac) is a critical post-translational modification that regulates photosynthesis and carbon metabolism in cyanobacteria. However, the diversity and functional roles of lysine acetyltransferases (KATs) beyond the well-characterized cGNAT2 remain poorly defined. This study identifies and functionally characterizes the previously unannotated protein A0096, designated here as cKAT, demonstrating its capacity to catalyze Kac both in vivo and in vitro in Synechococcus sp. PCC 7002 (Synechococcus). Deletion of cKAT significantly impaired cellular growth and photosynthetic efficien…

Effect of the Antibiotic Norfloxacin on Volatile Organic Compound Emissions From the Cyanobacteria Microcystis aeruginosa.

Cyanobacterial volatile organic compounds (VOCs) play important roles in aquatic ecosystems and toxicology. To uncover the promoting mechanisms of norfloxacin on cyanobacterial VOC emissions, the cell growth, reactive oxygen species (ROS) content, photosynthetic performance, and VOC emissions in Microcystis aeruginosa were investigated upon exposure to 10, 30, and 60 μM norfloxacin, and related gene expression was analyzed under the concentration (60 μM) with maximum promotion on the emission. Norfloxacin inhibited cell growth by reducing photosynthetic performance, causing ROS accumulation, a…

Classification machine learning to detect de facto reuse and cyanobacteria at a drinking water intake.

Harmful algal blooms (HABs) or higher levels of de facto water reuse (DFR) can increase the levels of certain contaminants at drinking water intakes. Therefore, the goal of this study was to use multi-class supervised machine learning (SML) classification with data collected from six online instruments measuring fourteen total water quality parameters to detect cyanobacteria (corresponding to approximately 950 cells/mL, 2900 cells/mL, and 8600 cells/mL) or DFR (0.5, 1 and 2 % of wastewater effluent) events in the raw water entering an intake. Among 56 screened models from the caret package in …

How Did Thylakoids Emerge in Cyanobacteria, and How Were the Primary Chloroplast and Chromatophore Acquired?

The emergence of thylakoid membranes in cyanobacteria is a key event in the evolution of all oxygenic photosynthetic cells, from prokaryotes to eukaryotes. Recent analyses show that they could originate from a unique lipid phase transition rather than from a supposed vesicular budding mechanism. Emergence of thylakoids coincided with the great oxygenation event, more than two billion years ago. The acquisition of semi-autonomous organelles, such as the mitochondrion, the chloroplast, and, more recently, the chromatophore, is a critical step in the evolution of eukaryotes. They resulted from pr…

Selective production of singlet oxygen for harmful cyanobacteria inactivation and cyanotoxins degradation: Efficiency and mechanisms.

Knowledge about the impact of singlet oxygen (1O2) on the characteristics and inactivation of harmful cyanobacterial organic matter is limited. In this study, the feasibility of using an improved single-iron doped graphite-like phase carbon nitride catalyst (FeCN) to activate peroxymonosulfate (PMS) catalytic production of 1O2 to inactivate four harmful cyanobacteria was investigated. The inactivation efficiencies at 30 min were 92.77%, 66.84%, 91.06%, and 93.45% for Microcystis aeruginosa (M. aeruginosa), Nodularia harveyana, Oscillatoria sp., and Nostoc sp., respectively. An optimized CaO2-functionalized alginate bead for simultaneous and efficient removal of phosphorous and harmful cyanobacteria.

Simultaneous removal of phosphorus (P) and algae is important to mitigate eutrophication, however, it is rather challenging in remediation of harmful algal blooms (HABs)-contaminated water. In this study, a wet alginate bead functionalized by CaO2 particle formed layer by layer was prepared with an in-situ method and optimized to remove phosphorous and inhibit algae growth. The stable H2O2 release with a concentration level of 0.06 mM was observed for a period of 26 d. The content of peroxy groups (-O-O-) in the optimal bead was 0.44 mmol·g-1 through permanganate-based titration study. For sol

Putative antiparasite defensive system involving ribosomal and nonribosomal oligopeptides in cyanobacteria of the genus Planktothrix.

Parasitic chytrid fungi can inflict significant mortality on cyanobacteria but frequently fail to keep cyanobacterial dominance and bloom formation in check. Our study tested whether oligopeptide production, a common feature in many cyanobacteria, can be a defensive mechanism against chytrid parasitism. The study employed the cyanobacterial strain Planktothrix NIVA-CYA126/8 and its mutants with knockout mutations for microcystins, anabaenopeptins, and microviridins, major oligopeptide classes to be found in NIVA-CYA126/8. Four chytrid strains were used as parasite models. They are obligate par…

Different effects of copper (II), cadmium (II) and phosphate on the sorption of phenanthrene on the biomass of cyanobacteria.

Due to the large surface area and high organic carbon content of cyanobacteria, organic contaminants can be readily sorbed on cyanobacteria during algal blooms, and then be transferred to the food web. This process is likely to be affected by the coexisting metals and nutrients, however, the possible impacts remain unclear. Effects of Cu(2+), Cd(2+), and phosphate on the sorption of phenanthrene on cyanobacterial biomass collected from an algal bloom were therefore studied. Continuous decrease in phenanthrene sorption was observed in the presence of low concentrations of Cu(2+), and Cd(2+) (<0…

NanoSIMS 50 elucidation of the natural element composition in structures of cyanobacteria and their exposure to halogen compounds.

AIMS: NanoSIMS (secondary ion mass spectrometry) is a powerful technique for mapping the elemental composition of a variety of small-scale samples (e.g. in Material Research, Cosmochemistry and Geology). However, its analytical features are making it also valuable to address biological questions. We demonstrate the ability of the NanoSIMS 50 to map elements at subcellular lateral resolution (approx. 50 nm) within cyanobacteria (Anabaena sp. and Cylindrospermum alatosporum) and its feasibility to investigate the uptake of bromine-containing substances (NaBr and deltamethrin). Exploitation of genomic sequences in a systematic analysis to access how cyanobacteria sense environmental stress.

The perception and subsequent transduction of environmental signals are primary events in the acclimation of living organisms to changes in their environment. Many of the molecular sensors and transducers of environmental stress cannot be identified by traditional and conventional methods. Therefore, the genomic information has been exploited in a systematic approach to this problem, performing systematic mutagenesis of potential sensors and transducers, namely, histidine kinases and response regulators, respectively, in combination with DNA microarray analysis, to examine the genome-wide expr

Engineered Cyanobacteria-Hydrogel Microecological System for Oxygen Reprogramming Synergizes Immunomodulation With Anabolism in Osteoarthritis Therapy.

Osteoarthritis remains a major clinical challenge due to its complex pathophysiology involving chronic inflammation and progressive cartilage degradation. Current treatments largely fail to address the dual demands of synovial anti-inflammation and cartilage anabolism, particularly under the critical yet often overlooked influence of oxygen zonation within articular tissues. In this study, we propose a novel microorganism oxygen metabolism engineering strategy using an injectable cyanobacteria-laden hydrogel (R-alga) that enables light-dependent dynamic oxygen regulation to mimic native physio…

Cyanobacteria newly isolated from marine volcanic seeps display rapid sinking and robust, high-density growth.

UNLABELLED: Cyanobacteria are photosynthetic organisms that play important roles in carbon cycling and are promising bioproduction chassis. Here, we isolate two novel cyanobacteria with 4.6Mbp genomes, UTEX 3221 and UTEX 3222, from a unique marine environment with naturally elevated CO₂. We describe complete genome sequences for both isolates and, focusing on UTEX 3222 due to its planktonic growth in liquid, characterize biotechnologically relevant growth and biomass characteristics. UTEX 3222 outpaces other fast-growing model strains on a solid medium. It can double every 2.35 hours in a liqu…
