Evolution of jasmonate biosynthesis and signaling mechanisms.

Jasmonates are phytohormones that modulate a wide spectrum of plant physiological processes, especially defense against herbivores and necrotrophs. The molecular mechanisms of jasmonate biosynthesis and signaling have been well characterized in model plants. In this review, we provide an in-depth analysis and overview of the origin and evolution of the jasmonate biosynthesis and signaling pathways. Furthermore, we discuss the striking parallels between jasmonate and auxin signaling mechanisms, which reveals a common ancestry of these signaling mechanisms. Finally, we highlight the importance o

Control of anthracnose disease via increased activity of defence related enzymes in 'Hass' avocado fruit treated with methyl jasmonate and methyl salicylate.

Development of anthracnose disease caused by Colletotrichum gloeosporioides Penz. is one of the major issues within the avocado supply chain. Exposure to methyl jasmonate (MeJA) and methyl salicylate (MeSA) vapours at 10 and 100µmoll-1 was investigated as an alternative solution to commercial fungicide - prochloraz® that is currently being used by the industry. The incidence of anthracnose disease was found to be significantly reduced in 'Hass' avocado fruit treated with MeJA or MeSA vapours, especially at 100μmoll-1. The mechanism involved enhanced activity of defence related enzymes, i.e. ch

Allosteric modulation of dextromethorphan binding sites.

The nonopioid antitussives dextromethorphan (DM), carbetapentane and caramiphen are efficacious anticonvulsant agents in the rat MES test. The findings presented strongly suggest the existence of a novel allosteric mechanism by which drugs acting at two different but interacting sites, exert their effects. This mechanism has marked similarities with the gamma-aminobutyric acid (GABA)-benzodiazepine interactions, even though their binding sites are different. The allosteric interactions of dextromethorphan and phenytoin in the binding assay and the potentiation of the anticonvulsant effects of

Tripartite Interaction Between Penicillium pinophilum-Host Plants and CMV-Y: A Model for Endophyte-Mediated Viral Biocontrol.

Plant-fungus-virus tripartite interactions represent complex ecological systems in which mutualistic endophytes can influence host physiology, yet the molecular basis of endophyte-mediated defence remains poorly understood. Here, we demonstrate that the endophytic fungus Penicillium pinophilum EU0013 suppresses the yellow strain of cucumber mosaic virus (CMV-Y) in Solanum lycopersicum and Nicotiana benthamiana. CMV-Y infection induced pronounced oxidative and hormonal perturbations, which were mitigated by P. pinophilum colonisation. Endophyte-associated plants exhibited early accumulation of

Neddylation blockade protects endothelial cells against inflammation and ischemia-reperfusion injuries.

BACKGROUND: Long-term allograft survival is limited by the occurrence of chronic allograft dysfunction, mainly due to antibody mediated rejection and aspecific fibrosis. The latter is favored by ischemia-reperfusion injury (IRI), particularly in organs from extended criteria donors and from donors after cardiocirculatory arrest. The graft endothelium is highly sensitive to IRI that causes chronic damages and increases its immunogenicity. Thus, improvement of preservation methods and the treatment of organs prior to implantation (so called "graft preconditioning") constitute a major clinical ch

Enhancing the Phenolic Value of Polyphenol-Rich Extra Virgin Olive Oils Through Optimized Water Extraction, Macroporous Resin Recovery and Spray Drying.

Olive oil polyphenols are well-established for their health-promoting properties. A recently emerged polyphenol-rich extra virgin olive oil (EVOO) was subjected to a water extraction process, optimized using response surface methodology (RSM). The effects of salt content and the water-to-oil ratio on the total phenolics were initially evaluated. Maximal phenolic yield was obtained by a water/EVOO ratio of 9.6:1 (v:v) and 0% added salt, although the addition of 3% salt provided an oil-free extract with a satisfactory phenolic yield compared to the optimized extract. Further, the phenolics of th

Spirocyclopentane as a Novel Scaffold for Potent Ghrelin Receptor Full Agonists.

Muscle atrophy and cachexia are common comorbidities among patients suffering from cancer, chronic obstructive pulmonary disease, acquired immunodeficiency syndrome, and several other chronic diseases. The peptide hormone ghrelin exerts pleiotropic effects; it acts by stimulating growth hormone secretion and subsequent increase of insulin-like growth factor-1 levels, an important mediator of muscle growth and repair. Ghrelin also acts on inflammation, appetite, and adipogenesis and, therefore, has been considered a promising therapeutic target for catabolic conditions. We previously reported o

Effector enrichment by Candidatus Liberibacter promotes Diaphorina citri feeding via Jasmonic acid pathway suppression.

BACKGROUND: Citrus huanglongbing (HLB) is a devastating disease caused by Candidatus Liberibacter asiaticus (CLas) that affects the citrus industry. In nature, CLas relies primarily on Diaphorina citri Kuwayama as its vector for dissemination. After D. citri ingests CLas-infected citrus, the pathogen infiltrates the insect's body, where it thrives, reproduces, and exerts regulatory control over the growth and metabolism of D. citri. Previous studies have shown that CLas alters the composition of proteins in the saliva of D. citri, but the functions of these proteins remain largely unknown. RE

Sensing nature's alarm: SnO2/MXene gas sensor unveils methyl jasmonate signatures of plant insect stress.

The incorporation of artificial intelligence into agriculture presents challenges, particularly due to hardware limitations, especially in sensors. Currently, pest detection relies heavily on manual scouting by humans. Therefore, the objective of this study is to create a chemoresistive sensor that enables early identification of the characteristic volatile compound, viz., methyl jasmonate, released during pest infestations. Given the lower reactivity of esters, we have fine-tuned a composite consisting of SnO2 nanoparticles and 2D-MXene sheets to enhance adsorption and selective oxidation, re

Phytotoxic and Antimicrobial Terrein Derivatives and Butenolides Isolated from the Endophytic Fungus Aspergillus terreus HT5.

Two new terrein derivatives, aspergilethers A and B (1 and 2), two known analogues (3 and 4), and three known butenolides (5-7) were isolated from the endophyte Aspergillus terreus HT5. Their structures were determined by spectroscopic analysis and ECD and NMR calculations. Interestingly, 1 and 2 had unpresented medium aliphatic side chains in terrein derivatives, with different absolute configurations at C-7, which was very scarce. (+)-Terrein (3) exhibited potent postemergence phytotoxicity toward Amaranthaceae, Portulacaceae, and Fabaceae, with MIC values of 250-1000 μg/mL. Transcriptome an

Plant jasmonic acid mediated contrasting effects of two citrus aphid species on Diaphorina citri Kuwayama.

BACKGROUND: Herbivores may influence each other directly and through plant mediated inter-specific interactions. The Asian citrus psyllid (Diaphorina citri Kuwayama) and citrus aphids are key pests that can co-exist on citrus, but their plant-mediated interaction between them is unknown. Here we investigated plant-mediated effect of two citrus aphid species, the polyphagous Aphis spiraecola Patch and the oligophagous Aphis (Toxoptera) citricidus (Kirkaldy) on the feeding behavior and reproduction of Diaphorina citri, and explored the underlying mechanisms. RESULTS: In comparison with those on

Exogenous methyl jasmonate enhances phytochemicals and delays senescence in harvested strawberries by modulating GABA shunt pathway.

In order to illustrate the relationship between methyl jasmonate (MeJA) and gamma aminobutyric acid (GABA) in maintaining the quality and improving the postharvest life of strawberry fruit, the harvested fruit were treated with MeJA for 16 h at 20 °C and stored at 3 ± 0.5 °C for 12 days. MeJA enhanced the expression levels of GABA shunt pathway-related genes, including glutamate decarboxylase, GABA transaminase, and succinic semialdehyde dehydrogenase, leading to an increase in GABA accumulation. Treated fruit showed higher levels of total acids, anthocyanins, total phenolics, antioxidants, an

Cyclopentadiene as a Multifunctional Reagent for Normal- and Inverse-Electron Demand Diels-Alder Bioconjugation.

Optimizing the Diels-Alder (DA) reaction for aqueous coupling has resulted in practical methods to link molecules such as drugs and diagnostic agents to proteins. Both normal electron demand (NED) and inverse electron demand (IED) DA coupling schemes have been employed, but neither mechanism entails a common multipurpose reactive group. This report focuses on expanding the bioconjugation toolbox for cyclopentadiene through the identification of reactive groups that couple through NED or IED mechanisms in aqueous solution. Dienophiles and tetrazine derivatives were screened for reactivity and s

Methyl jasmonate-induced resistance to Delia platura (Diptera: Anthomyiidae) in Lupinus mutabilis.

BACKGROUND: Andean lupin (Lupinus mutabilis Sweet) is an important leguminous crop from South America with a high protein content. In Ecuador, lupin yields are severely affected by the infestation of Delia platura larvae on germinating seeds. The application of elicitor molecules with activity against herbivorous insects to control D. platura infestation constitutes an unexplored and promising alternative for chemical insecticides. In this study, methyl jasmonate (MeJA), hexanoic acid, menadione sodium bisulfite, and DL-β-aminobutyric acid were evaluated for their ability to induce resistance

Long-distance signalling in plant defence.

Plants use inducible defence mechanisms to fend off harmful organisms. Resistance that is induced in response to local attack is often expressed systemically, that is, in organs that are not yet damaged. In the search for translocated defence signals, biochemical studies follow the physical movement of putative signals, and grafting experiments use mutants that are impaired in the production or perception of these signals. Long-distance signals can directly activate defence or can prime for the stronger and faster induction of defence. Historically, research has focused on the vascular transpo

Compounds from gum ammoniacum with acetylcholinesterase inhibitory activity.

The use of herbal medicinal preparations in dementia therapy has been studied based on experience from traditional medicine. A dichloromethane extract of gum ammoniacum, the gum-resin from Dorema ammoniacum D. Don had shown acetylcholinesterase (AChE) inhibitory activity in a previous study. The aim of this study was the isolation and characterization of the active compounds from this resin. The extract was investigated by a respective colorimetric microplate assay and the active zones were identified via TLC bioautography and isolated using several chromatographic techniques. The structures o
