Age of onset and death in inherited prion disease are heritable.

The common polymorphism at codon 129 of the prion protein gene (PRNP) is known to affect prion disease susceptibility, incubation period and phenotype. Mouse quantitative trait locus (QTL) studies demonstrate multiple modifiers of incubation time unlinked to Prnp, suggesting the existence of homologous human prion disease modifiers, but direct evidence of these has been lacking. We investigated the correlation of age at onset and death, expressed as a composite Z score, between parents and offspring in three large UK inherited prion disease kindreds. Our analysis suggests that overall heritabi

An epidemiological model for direct and indirect transmission of typhoid fever.

The modified SIS epidemiological model considers the usual direct transmission (short cycle) and indirect transmission (long cycle) of typhoid fever. Thresholds are determined, and the equilibrium points are shown to be globally stable. Local stability of the equilibrium points is shown in the corresponding model with vaccines. After estimating parameters using current statistical data for typhoid fever in Chile, computer simulations are used to obtain the numerical behavior of this disease and to estimate the effect of several control policies.

Health, wealth, and air pollution: advancing theory and methods.

The effects of both ambient air pollution and socioeconomic position (SEP) on health are well documented. A limited number of recent studies suggest that SEP may itself play a role in the epidemiology of disease and death associated with exposure to air pollution. Together with evidence that poor and working-class communities are often more exposed to air pollution, these studies have stimulated discussion among scientists, policy makers, and the public about the differential distribution of the health impacts from air pollution. Science and public policy would benefit from additional research

Pyrethroid pesticide exposure and hematological cancer: epidemiological, biological and molecular evidence.

Pyrethroid insecticides are commonly used worldwide. The chronic effects of these compounds are of concern given that epidemiological studies have suggested an association with hematological cancer, particularly in children. However, the biological evidence at molecular and cellular levels is limited. A review on the molecular and cellular effects of pyrethroids is helpful to guide the study of the biological plausibility of the association of pyrethroids with hematological cancer. We reviewed studies suggesting that pyrethroids are genotoxic, induce genetic rearrangements, alter gene expressi

Drug repositioning for Alzheimer's disease.

Existing drugs for Alzheimer's disease provide symptomatic benefit for up to 12 months, but there are no approved disease-modifying therapies. Given the recent failures of various novel disease-modifying therapies in clinical trials, a complementary strategy based on repositioning drugs that are approved for other indications could be attractive. Indeed, a substantial body of preclinical work indicates that several classes of such drugs have potentially beneficial effects on Alzheimer's-like brain pathology, and for some drugs the evidence is also supported by epidemiological data or prelimina

Dementia treatment versus prevention.

Alzheimer disease (AD) and dementia are becoming increasingly prevalent due to the aging of the global populations. Currently available treatment options, including acetylcholinesterase inhibitors and memantine, only have symptomatic effects and no drugs with disease-modifying properties are available. Research on the amyloid cascade indicates that amyloid-β (Aβ) clearance from the brain may be the main pathophysiological change in late-onset AD and the key driver of neurodegeneration, which ultimately results in progressive cognitive deterioration and dementia. Most new AD drug candidates tar

Prognostic Value of Tumor-Infiltrating Lymphocytes in Head and Neck Squamous Cell Carcinoma.

IMPORTANCE: Biomarkers that reflect prognosis and cellular immunity in patients with head and neck squamous cell carcinoma (HNSCC) are a prerequisite for improving individualized treatment that limits the intensity and morbidity of conventional treatment and may be useful in the introduction of new immunotherapy regimens. OBJECTIVE: To determine if specific classes of tumor-infiltrating lymphocytes (TILs) in pretreatment biopsy specimens have prognostic value for outcomes in a large training and validation cohort of patients with HNSCC. DESIGN, SETTING, AND PARTICIPANTS: In this prospective,

[Developmental origins of health and disease in adults: role of maternal environment].

Many epidemiological studies indicate that environmental conditions during embryonic and fetal development can have an impact on health at adulthood. Animal studies clearly demonstrate that maternal, and even paternal undernutrition or nutritional excess durably modify some epigenetic marks in their offspring, affecting gene expression and physiological adaptations to the environment. It is crucial to better define the effects of early environment on adult phenotype and epigenetic marks in humans and to develop, with the help of animal models, new preventive strategies and treatments.

An Aspirin a Day: New Pharmacological Developments and Cancer Chemoprevention.

Chemoprevention refers to the use of natural or synthetic agents to reverse, suppress, or prevent the progression or recurrence of cancer. A large body of preclinical and clinical data suggest the ability of aspirin to prevent precursor lesions and cancers, but much of the clinical data are inferential and based on descriptive epidemiology, case control, and cohort studies or studies designed to answer other questions (e.g., cardiovascular mortality). Multiple pharmacological, clinical, and epidemiologic studies suggest that aspirin can prevent certain cancers but may also cause other effects

Integrating Knowledge of Modifiable and Non-modifiable Risk Factors: A Comprehensive Framework for Breast Cancer Prevention Practices in Women.

Breast cancer is the leading cause of death among women worldwide, with mortality rates disproportionately high in less-developed regions. Effective prevention strategies, including educational initiatives, are crucial. This study explores a comprehensive framework for breast cancer prevention by examining the relationship between awareness of modifiable and non-modifiable risk factors and knowledge of preventive practices among women over 20 years of age. A cross-sectional, observational study was conducted from January to June 2022 at a Mexican first-contact center on the Mexico-USA border.

ERS International Congress 2021: highlights from the Epidemiology and Environment Assembly.

In this article, early career members of the Epidemiology and Environment Assembly of the European Respiratory Society (ERS) summarise a selection of four sessions from the Society's 2021 virtual congress. The topics covered focus on chronic respiratory disease epidemiology, the health effects of tobacco and nicotine, and the respiratory health impact of environmental exposures and climate change. While the burden of chronic respiratory diseases such as COPD is expected to increase in the next decades, research on modifiable risk factors remains key. The tobacco and nicotine research presented

Foundations of hand-transmitted vibration standards.

Standards for hand-transmitted vibration predict dependent variables (e.g. finger blanching) from measurements of a few independent variables (e.g. vibration magnitude, vibration frequency, exposure duration). This paper illustrates the assumptions in the current International Standard guidelines for the evaluation of hand-transmitted vibration and compares research methods which may provide information to improve the guidance. Subjective assessments of vibration discomfort have influenced the frequency weighting used in current standards, but the data have been modified greatly for this purpo

Single nucleotide polymorphisms of the DNA repair gene XPD/ERCC2 alter mRNA expression.

OBJECTIVES: Epidemiological studies documented associations between single nucleotide polymorphisms (SNPs) in the nucleotide excision repair gene XPD/ERCC2 and cancer risk. Little is known, however, about the underlying mechanisms for these associations. We explored a novel mechanism that could further explain the reported risk-modifying effect of these SNPs on disease susceptibility. METHODS: Using quantitative real-time polymerase chain reaction, we examined the relationship between three SNPs in the XPD gene (R156R in exon 6, D312N in exon 10 and K751Q in exon 23) and mRNA levels as a pote

Synthesis, characterization and evaluation of hyaluronic acid-based polymers for nasal delivery.

Epidemiological research has found that between 5 and 12 percent of the population suffers from chronic rhinosinusitis. Patients are dealing with local side effects such as nasal dryness, sporadic sneezing, and nasal pain in addition to the inflammation. The aim of this study was to synthesize a polymer based on hyaluronic acid in order to provide lubrication combined with a ligand leading to a covalent binding on the nasal mucosa. Hyaluronic acid (HA) was modified with L-cysteine ethyl ester hydrochloride (CYS) via amid bond formation. Ellman's assay, together with spectroscopic techniques li

What was lost in translation in the DHA trial is whom you should intend to treat.

The results of a randomized double-blind placebocontrolled trial with docosahexaenoic acid (DHA) supplementation in mild to moderate Alzheimer's disease (AD) published by Quinn and colleagues in JAMA argues against overall efficacy of DHA in slowing progression. However, certain caveats in the results caution against discarding DHA altogether, raising questions about oxidation, dosage, pharmacogenomics and stage of intervention. One potential misconception is that what works for prevention will slow progression in AD subjects.Preclinical studies with DHA supported the rationale for early stage

Aluminum in the diet and Alzheimer's disease: from current epidemiology to possible disease-modifying treatment.

In recent years, interest in the potential role of metals in the pathogenesis of Alzheimer's disease (AD) has grown considerably. In particular, aluminum (Al) neurotoxicity was suggested after its discovery in the senile plaques and neurofibrillary tangles that represent the principal neuropathological hallmarks of AD. Al is omnipresent in everyday life and can enter the human body from several sources, most notably from drinking water and food consumption. The evidence supporting association from ingestion of Al from drinking water is somewhat stronger than for its ingestion from food. Howeve

Health effects from stratospheric ozone depletion and interactions with climate change.

The potential health effects of elevated levels of ambient UV-B radiation are diverse, and it is difficult to quantify the risks, especially as they are likely to be considerably modified by human behaviour. Nevertheless epidemiological and experimental studies have confirmed that UV radiation is a definite risk factor for certain types of cataract, with peak efficacy in the UV-B waveband. The causal link between squamous cell carcinoma and cumulative solar UV exposure has been well established. New findings regarding the genetic basis of skin cancer, including studies on genetically modified

The community epidemiology of trachoma.

Effective eradication of avoidable blindness due to trachoma requires an understanding of the epidemiologic determinants that promote the disease. These determinants include both host factors, such as age, sex, and race, and environmental determinants, including climate, water availability, and general socioeconomic conditions. All of these factors may drastically modify the incidence, prevalence, and severity of trachoma within a community. Longitudinal seroepidemiologic studies on the transmission of the infection have helped to elucidate the more important risk factors in trachomatous commu

Instruments to assess and measure personal and environmental radiofrequency-electromagnetic field exposures.

Radiofrequency-electromagnetic field (RF-EMF) exposure of human populations is increasing due to the widespread use of mobile phones and other telecommunication and broadcasting technologies. There are ongoing concerns about potential short- and long-term public health consequences from RF-EMF exposures. To elucidate the RF-EMF exposure-effect relationships, an objective evaluation of the exposures with robust assessment tools is necessary. This review discusses and compares currently available RF-EMF exposure assessment instruments, which can be used in human epidemiological studies. Quantita
