The pathology of experimental obstructive hydrocephalus. A scanning electron microscopic study.

Obstructive hydrocephalus was produced in 14-day old rabbits by injection of kaolin into the cisterna magna. The ependymal lining was studied by scanning electron microscopy. Marked hydrocephalus was present 1 or 2 weeks after the kaolin injection. The ependymal lining adapted remarkably well to the rapid expansion by increasing the surface area of the ependymal cells. No breaks or denudement of the ependymal lining was observed except at the sites of ruptured ventricular synechiae. Generally, these findings confirm previous light and electron microscopic observations on the same model (Torvik…

[Deep-seated (corpus callosum, intraventricular, basal ganglia and insula) and brain stem cavernous angiomas. Experience in Brazil].

With a review of the literature, we report our experience with surgical treatment of deep-seated cavernomas (intraventricular, of the corpus … tion, surgery should only concern symptomatic or hemorrhagic lesions close to the pia-matter or the ependyma as well as those covered by a thin layer of parenchyma. Neuronavigation and microsurgical procedures are essential in the treatment of deep-seated cavernomas.

Transnasal endoscopic surgery of sphenoid meningocele associated with pituitary adenoma. A case report.

BACKGROUND: Transsphenoid meningoencephalocele is a congenital anomaly formed by herniation of an ependyma delimited sac through a bony defect into the sphenoid sinus. The sac contains cerebrospinal fluid and neurovascular structures. The prevalence of transsphenoid meningoencephalocele in the adult population is rare. It usually manifests as nasal liquorrhoea. METHODS AND RESULTS: This case report presents an adult male who underwent surgery due to suspected pituitary macroadenoma. The surgery was performed endoscopically via the transnasal approach with a surprising finding of true transsph…

Human neonatal hydrocephalus. An electron microscopic study of the periventricular tissue.

An infant of 43 weeks gestational age with severe congenital hydrocephalus was operated on for removal of a subependymal astrocytoma in the region of the foramen of Monro. A biopsy of periventricular tissue was taken from the lateral ventricle for examination by scanning and transmission electron microscopy. The ependyma was largely denuded, with glial cell processes forming most of the ventricular lining. Many of the attenuated ependymal cells, however, had intact junctional complexes at areas of contact with other ependymal cells. Club-shaped unipolar cells, believed to be a previously undes…

IMMUNOFLUORESCENT STUDIES OF THE HISTOPATHOGENESIS OF LYMPHOCYTIC CHORIOMENINGITIS VIRUS INFECTION.

LCM virus antigen was visualized in infected tissue cultures and mouse tissues by the direct immunofluorescent procedure. In all instances, antigen was localized to the cytoplasm. In intracerebrally infected mice, antigen was almost completely restricted to meninges, choroid plexus, and ependyma; at no time were neurons involved. Mice infected by intraperitoneal inoculation of a viscerotropic strain demonstrated antigen chiefly in liver parenchyma, splenic reticulum, bronchi, and alveolar cells. Congenitally infected mice showed antigen in almost all cell types observed, but generally in only …

Ultrastructural aspects of tumour invasion in the central nervous system.

Tumour invasion in the central nervous system (CNS) was studied using a mouse melanoma cell model. Attention was concentrated on the third ventricle as this site is distant from any implantation artefact. Transmission electron microscopy (TEM) revealed the formation of tumour cell aggregates within the ventricle. Underlying ependymal cells showed loss of cilia and microvilli. Progressive thinning of the ependymal cells took place until continuity of the ependyma was lost through ependymal retraction. Tumour cells were then in direct contact with sub-ependymal neuroglial processes. Further inva…

Ependymal Cells and Hydrocephalus: Recent Advances in Molecular Mechanisms.

Ependymal cells are specialized multiciliated epithelial cells that line the brain ventricles. Their coordinated ciliary beating contributes to cerebrospinal fluid circulation and neural homeostasis. Hydrocephalus, a severe neurological condition affecting ∼1-3 per 1000 births worldwide, can arise from defects in ciliary motility, ependymal cell differentiation, planar cell polarity (PCP), and cytoskeletal organization. Mutations in genes that regulate these processes have been identified in patients with congenital hydrocephalus and related ciliopathies, establishing the clinical relevance of…

[The fine structure of ependymona (author's transl)].

The tumor cells form an unoriented mass rather than a layered structure. Most of the cells have no ependymal specialization, such as cilia, microtubular, etc. Instead, they assume a primitive undifferentiated appearance with large nuclei and abundant free ribosomes. Occasionally, however, certain cells tend to form a ring around a narrow lumen-like space within the mass. Such spaces are virtually filled with microvilli and, occasionally, even cilia arising form the surrounding cells. Elaborate desmosomes decorate the apposing surfaces of such cells and basal bodies are presented within the cyt…

Cerebral artery reverse flow as an indicator of critical intracranial hypertension in fetal intracranial hemorrhage: Case report.

Intracranial hemorrhage (ICH) is a rare serious fetal event. It manifests via non-specific sonographic findings such as: ventriculomegaly, increased periventricular echogenity, hyperechogenic ependyma and presence of avascular intracranial masses. Severe ICH causes an increase in fetal intracranial pressure, which can be quantified in utero by the presence of reverse flow in the middle cerebral artery (MCA-REDF). Retrograde flow in the middle cerebral artery indicates already deteriorating circulatory conditions in the fetal brain with loss of autoregulatory mechanisms and critical damage to t…

Myoclonin1 haploinsufficiency in motile ciliated cells partially recapitulates epileptic features of Efhc1-deficient mice in adult age.

Mutations in the EFHC1 (EF-hand domain containing 1) gene, which encodes myoclonin1, a homolog of the Chlamydomonas axonemal protein Rib72, have been identified in patients with epilepsies including juvenile myoclonic epilepsy (JME). Myoclonin1 is expressed in choroid plexus epithelial cells during fetal development and in motile cilia of ependymal cells lining brain ventricles as well as in tracheal cilia and sperm flagella during postnatal stages. We previously reported that systemic Efhc1 deficiency in mice causes spontaneous myoclonus, increased susceptibility to the chemoconvulsant pentyl…

Methods to study motile ciliated cell types in the zebrafish brain.

Cilia are well conserved hair-like structures that have diverse sensory and motile functions. In the brain, motile ciliated cells, known as ependymal cells, line the cerebrospinal fluid (CSF) filled ventricles, where their beating contribute to fluid movement. Ependymal cells have gathered increasing interest since they are associated with hydrocephalus, a neurological condition with ventricular enlargement. In this article, we highlight methods to identify and characterize motile ciliated ependymal lineage in the brain of zebrafish using histological staining and transgenic reporter lines.

Idiopathic normal pressure hydrocephalus (iNPH) is considered an age-dependent chronic communicating hydrocephalus associated with cerebrosp … phalus were associated with an abnormal structure or dysfunction of motile cilia on the ventricular ependymal cells. Aging, alcohol consumption, sleep apnoea, diabetes and hypertension are candidates for the risk of developing iNPH, although there is no prospective cohort study to investigate the risk factors for iNPH. Alcohol intake may be associated with …

Expression of an antigen associated with basal bodies of human ciliated epithelial cells.

The process and regulation of ciliogenesis in human epithelia is little understood and many components of the cilium and associated structures have not been characterised. We have identified a monoclonal antibody, LhS28, which recognises a 44,000-45,000 M(r) protein specifically associated with human ciliated epithelial cells. Immunoperoxidase labelling of formalin-fixed paraffin wax-embedded human tissues showed that LhS28 was expressed in the sub-apical zone of ciliated epithelial cells of the Fallopian tube and upper respiratory tract, but not ciliated ependyma, non-ciliated epithelia or te…

Distribution of angiotensin II receptor binding in the spinal cord of the sheep.

The distribution of angiotensin II binding sites has been mapped at segmental levels throughout the spinal cord of the sheep using in vitro autoradiographic methods. Binding of 125I-[Sar1.Ile8] Ang II is most prominent in the lateral horns of the thoracolumbar and sacral regions containing the sympathetic and parasympathetic preganglionic neurons respectively. Binding is also present in the dorsal horns of the grey matter, in the central canal region, dorsal root ganglia and associated with non-neuronal elements such as the ependyma surrounding the central canal, and blood vessels. Displacemen…

Epithelial cysts of the neuraxis: presentation of three cases and a review of the origins and classification.

Benign, epithelial-lined cysts of the neuraxis may be asymptomatic or may behave as space-occupying lesions. Presentation of three such cysts, including an intramedullary epidermoid cyst, a lumbosacral subcutaneous enteric cyst that has an epithelium resembling ependyma, and a hypophyseal duct cyst, illustrates typical problems encountered. Review of their histogenesis and possible embryogenesis indicates that intracranial ependymal cysts and cysts of the sella turcica are not normally associated with other anomalies and frequently occur after middle age, whereas dermal and enteric cysts occur…

Multiple neuroepithelial (colloid) cysts: association with other congenital anomalies.

A 4-year-old boy had three neuroepithelial cysts, one in the third ventricle and two in the supratentorial subarachnoid space, and also had stenosis of the cerebral aqueduct, obstructive internal hydrocephalus, frontoethmoidal meningoencephalocele, and right microphthalmia. The occurence of these combined lesions supports the congenitially derived nature of the cysts. The cysts originated from neuroepithelium including ectopic ependyma-lined structures. The exception may be those cysts arising in or near the floor of the third ventricle; these cysts could be derived from the cleft of Rathke's …
