BACKGROUND: Alzheimer's disease (AD) is a form of dementia that strikes elderly people more frequently than it does younger people. BACKGROUND: Non-ergot dopamine agonists (NEDAs) have been used as monotherapy or as an adjunctive therapy to levodopa for many years. Novel long-acting formulations of NEDAs including pramipexole extended-release (ER), ropinirole prolonged-release (PR), and rotigotine transdermal patch have been developed. However, there is no strong evidence that a given NEDA is more potent than another. Twenty-two hyperprolactinaemic women were randomly allocated to two groups and treated with bromocriptine or the new, non-ergot, long-acting dopamine agonist, CV 205-502. The study was double-blind for 6 months. Four patients in the bromocriptine group, but none in the CV 205-502 group, discontinued the study because of adverse reactions. Rotigotine (RTG) is a non-ergot dopamine agonist developed as a new transdermal formulation, indicated for use in early and advanced Parkinson's disease (PD). The potential advantages of the RTG patch include immediacy of effect onset, constant drug delivery, better tolerability avoiding drug peaks and easy of use, helping patient's compliance. Ergot alkaloids (EAs) are mycotoxins produced by Claviceps and are present in cereals and their products; their residues pose significant threats to human health through food consumption, resulting in ergotism and sickness. Herein, a sensitive and rapid method for the determination of 11 EAs in cereals and their products using ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) were developed. Eleven EAs were extracted with 20 mL acetonitrile-200 mg/L ammonium acetate solution (80∶20, v/v) for 15 min using the vortex shock method followed by 15 min of ultrasonication.

Ergotism is an uncommon condition that affects patients with exposure to ergot alkaloids causing ischemia of extremities. We report the case of lower extremities ischemia caused by ergot toxicity in a human immunodeficiency virus (HIV) positive individual due to the interaction between ergot alkaloid and Cobicistat. In addition, we present a brief review of medical, and pharmacological aspects of this condition. To our knowledge, this is the second reported case describing this interaction.

Ergot poisoning is an uncommon but recognized cause of peripheral vascular ischemia and lower extremity gangrene. The traditional management of ergot-related vasospasm includes withdrawal of the inciting compound and supportive care. The authors report a rare case of ergotism related to lysergic acid diethylamide (LSD) ingestion and describe unconventional treatment of associated extensive lower extremity vasospasm with use of balloon percutaneous transluminal angioplasty.

BACKGROUND: Postpartum cerebral angiopathy is associated with the use of of ergot alkaloids.
