Biodegradable Cationic and Ionizable Cationic Lipids: A Roadmap for Safer Pharmaceutical Excipients.

Cationic and ionizable cationic lipids are broadly applied as auxiliary agents, but their use is associated with adverse effects. If these excipients are rapidly degraded to endogenously occurring metabolites such as amino acids and fatty acids, their toxic potential can be minimized. So far, synthesized and evaluated biodegradable cationic and ionizable cationic lipids already showed promising results in terms of functionality and safety. Within this review, an overview about the different types of such biodegradable lipids, the available building blocks, their synthesis and cleavage by endog…

Novel trehalose-based excipients for stabilizing nebulized anti-SARS-CoV-2 antibody.

COVID-19 is caused by the infection of the lungs by SARS-CoV-2. Monoclonal antibodies, such as sotrovimab, showed great efficiency in neutra … eatment, and can increase its immunogenicity. To address those issues, we developed a series of new excipients composed of a trehalose core, a succinyl side chain and a hydrophobic carbon chain (from 8 to 16 carbons). Succinylation increased the solubility of the excipients, allowing their use at relevant concentrations for protein stabilization. In particul …

Pharmaceutical excipients with potential to cause adverse effects in paediatric nasal medicines.

Some pharmaceutical excipients may cause adverse reactions, excipient-related interactions and/or contraindications. Due to the unique characteristics of the paediatric population, adverse effects may occur to substances generally thought safe. The proportion of topical nasal medicines approved for paediatric use and the prevalence and labelling of excipients with known effect (EKE) in these products were compared in Serbia as a non-EU country and Croatia and Slovenia as EU countries. The study was designed as a post-authorization safety study and safety of excipients was considered in accorda…

Acute cell stress screen with supervised machine learning predicts cytotoxicity of excipients.

Excipients serve as vehicles, preservatives, solubilizers, and colorants for drugs, food, and cosmetics. They are considered to be inert at biological targets; however, several reports suggest that some could interact with human targets and cause unwanted effects. We investigated 40 commonly used drug excipients for cellular stress in the AsedaSciences® SYSTEMETRIC® Cell Health Screen, which was developed to estimate toxicity risk of small molecular entities (SMEs). The screen uses supervised machine learning (ML) to classify test compound cell stress phenotypes relative to a training set of o…

Intestinal absorption-modifying excipients: A current update on preclinical in vivo evaluations.

Pharmaceutical excipients in drug products are defined as pharmacologically inactive and are integral constituents of all types of oral dosage forms. However, some excipients may increase drug absorption by interacting with the mucosal membrane. If the strategy is to use an excipient with a potential to affect the processes determining the rate and/or extent of the intestinal drug absorption, it is defined as an absorption-modifying excipients (AME). These pharmaceutical excipients may act as AMEs, depending on the amounts applied, and accordingly influence bioequivalence assessment of innovat…

Mechanistic Insight into Caffeine-Oxalic Cocrystal Dissociation in Formulations: Role of Excipients.

Caffeine-oxalic acid cocrystal, widely reported to be stable under high humidity, dissociated in the presence of numerous pharmaceutical excipients. In cocrystal-excipient binary systems, the water mediated dissociation reaction occurred under pharmaceutically relevant storage conditions. Powder X-ray diffractometry was used to identify the dissociated products obtained as a consequence of coformer-excipient interaction. The proposed cocrystal dissociation mechanism involved water sorption, dissolution of cocrystal and excipient in the sorbed water, proton transfer from oxalic acid to the exci…

Surface engineered excipients: I. improved functional properties of fine grade microcrystalline cellulose.

Excipients with good flowability, bulk density as well as compaction properties are desired for use in tableting since they play important roles in formulation development and processing, including, handling, mixing, feeding and compaction. The objective of this paper is to examine the feasibility of using dry coating based surface modification of microcrystalline cellulose, Avicel PH-105, to produce an engineered fine grade (<30 μm) excipient that has all three desired properties. Using a material sparing high-intensity vibrational mixer, Avciel PH-105 is dry coated with 1 wt% Aerosil 200, se…

Analysis of structural variability in pharmaceutical excipients using solid-state NMR spectroscopy.

Polysaccharide-based excipients comprise the majority of most solid dosage forms and can vary dramatically in terms of structural and functionally related properties. Analytical methods for characterizing these important formulation components are crucial. Solid-state NMR spectroscopy (SSNMR) can provide a wealth of information on these materials while offering the advantages of non-destructive sample preparation and selectivity. The overall objective of this work is to identify SSNMR parameters that can be used to detect differences among these excipients. Excipients were obtained from a wide…

Translational Barriers to Pharmaceutical Excipient Readiness in Marine-Derived Polymers.

Marine polymers have attracted a lot of attention as potential alternatives to the traditional animal-derived polymers in pharmaceutical formulation since they are abundant, biocompatible, and versatile in functionality. However, the presence of these materials in dosage-form studies, often in support of proof-of-concept trials, does not mean they are ready to apply as excipients routinely. This review critically evaluates the reasons why three of the most highly researched marine-derived polymers, chitosan, alginate, and carrageenan, continue to encounter significant translational barriers in…

Effects of anion excipients on the viscosity of high-concentration mAb solution.

High protein concentrations often lead to high viscosity, necessitating reduced solution viscosity. We investigated the effects of eight anionic excipients commonly used in biopharmaceutical formulations on the solution viscosity of high-concentration monoclonal antibodies using three antibodies with different isoelectric points. The three antibodies exhibited attractive or repulsive protein-protein interaction tendencies in histidine buffer without anionic excipients. The effect of adding anionic excipients on solution viscosity differed by interaction tendencies. Preferential interaction coe…

Review of Applications of Cyclodextrins as Taste-Masking Excipients for Pharmaceutical Purposes.

It is widely recognized that many active pharmaceutical ingredients (APIs) have a disagreeable taste that affects patient acceptability, particularly in children. Consequently, developing dosage forms with a masked taste has attracted a lot of interest. The application of cyclodextrins as pharmaceutical excipients is highly appreciated and well established, including their roles as drug delivery systems, solubilizers and absorption promoters, agents that improve drug stability, or even APIs. The first work describing the application of the taste-masking properties of CDs as pharmaceutical exci…

Excipient Lung Disease in a Patient with Preexisting Chronic Thromboembolic Disease.

Excipient lung disease is an uncommon condition that occurs when crushed oral tablets are injected intravenously and cause pulmonary hypertension. Diagnosis requires a high degree of clinical suspicion and recognition of characteristic imaging findings. The authors present a previously unreported combination of findings in a patient with excipient lung disease and preexisting chronic thromboembolic disease. A pattern of diffuse centrilobular nodules with sparing of lung segments with chronically occluded pulmonary arteries was highly suggestive of the diagnosis and indicated a vascular cause.

Formulation Excipients and Their Role in Insulin Stability and Association State in Formulation.

While  excipients are often overlooked as the "inactive" ingredients in pharmaceutical formulations, they often play a critical role in protein stability and absorption kinetics. Recent work has identified an ultrafast absorbing insulin formulation that is the result of excipient modifications. Specifically, the insulin monomer can be isolated by replacing zinc and the phenolic preservative metacresol with phenoxyethanol as an antimicrobial agent and an amphiphilic acrylamide copolymer excipient for stability. A greater understanding is needed of the interplay between excipients, insulin assoc…

Analyzing excipient-related adverse events in antiseizure drug formulations.

There have been several reports that switching formulations of antiseizure medications (ASMs) has been associated with a deterioration of seizure control, seizure relapse or increased adverse effects. Considering recent findings that excipients, namely purported inactive ingredients, may nevertheless exert biological effects, it is possible that the variation in adverse event profile of antiseizure drugs may be related to differences in excipients. To test our hypothesis, we constructed a new research tool to connect FDA-compiled adverse event reports to the excipient in the medicine. Analysis…
