Rapid granulation versus functional imbalance of aerobic granular sludge under F-53B and OBS stress: A ROS-mediated metabolic trade-off mechanism.

F-53B and OBS, two major alternatives to perfluorooctane sulfonate (PFOS), are frequently detected in wastewater treatment systems; however, their effects on aerobic granular sludge (AGS) formation and functional stability remain poorly understood. Therefore, this study investigated the impacts of long-term F-53B and OBS exposure on sludge performance and microbial community dynamics during AGS granulation. Compared with the control reactor, both F-53B and OBS significantly stimulated extracellular polymeric substance (EPS) secretion (R1: 103.09 ± 1.73 mg/g VSS; R2: 132.63 ± 4.05 mg/g VSS; R3:

Mechanistic insights into the enhancement of azithromycin biodegradation by extracellular polymeric substances: Roles of metabolic activity, electron transfer, and reactive oxygen species.

White-rot fungi (WRF) have shown potential as an effective alternative for antibiotic removal in wastewater treatment. However, their slow degradation rates remain a major challenge. In this study, we demonstrate that extracellular polymeric substances (EPS) from Brucella intermedia ZL-06 significantly enhance the degradation of azithromycin (AZI) by the WRF Trametes versicolor, reducing the degradation time from 9 to 6 days and achieving > 99.9 % elimination. Mass-balance calculations revealed a 35.33 % increase in the specific biodegradation rate per unit biomass. Mechanistic analysis showed

Investigating the role of extracellular polymeric substances produced by Parachlorella kessleri in Zn(II) bioremediation using atomic force microscopy.

Microalgae, such as Parachlorella kessleri, have significant potential for environmental remediation, especially in removing heavy metals like zinc from water. This study investigates how P. kessleri, isolated from a polluted river in Argentina, can remediate zinc. Using atomic force microscopy (AFM), the research examined the interactions between Zn particles and cells grown with different nitrogen sources-nitrate or ammonium. The results showed that cells grown with nitrate produced extracellular polymeric substances (EPS), while those grown with ammonium did not. Raman spectroscopy revealed

Role of the Yersinia pestis phospholipase D (Ymt) in the initial aggregation step of biofilm formation in the flea.

UNLABELLED: Transmission of Yersinia pestis by fleas depends on the formation of condensed bacterial aggregates embedded within a gel-like matrix that localizes to the proventricular valve in the flea foregut and interferes with normal blood feeding. This is essentially a bacterial biofilm phenomenon, which at its end stage requires the production of a Y. pestis exopolysaccharide that bridges the bacteria together in a cohesive, dense biofilm that completely blocks the proventriculus. However, bacterial aggregates are evident within an hour after a flea ingests Y. pestis, and the bacterial exo

Photodynamic antimicrobial chemotherapy activities of phthalocyanine-antibiotic conjugates against bacterial biofilms and interactions with extracellular polymeric substances.

This study sheds light on how to rationally design efficient photodynamic antimicrobial chemotherapy (PACT) agents by covalently linking phthalocyanines (Pcs) as photosensitizers with an antibiotic: Ciprofloxacin (CIP). Pcs used are zinc (II) 3-(4-((3,17,23-tris(4-(Benzo(d)thiazol-2-yl] thiol) phthalocyanine-9-yl) oxy) phenyl) propanoic acid (1) and zinc (II) 3-(4-(3,17,23-tris(3-(4-(triphenylphosphine) butyl) benzo[d]thiazol-3-ium bromide phthalocyanine-9-yl) oxy) phenyl) propanoic acid (2). High singlet oxygen quantum yields are observed in the presence of CIP. Square wave voltammetry was us

Tetracyclines lead to ammonium accumulation during nitrification process.

The effect of tetracyclines used for swine food-production (tetracycline and oxytetracycline) on enriched nitrifying bacteria cultures over time was investigated in this study. Short-term exposure assays were performed in different concentrations of each antibiotic, using ammonia oxidizing bacteria (AOB) culture and nitrifying bacteria. The results pointed out a higher inhibitory effect of tetracycline on both bacterial communities. The AOB was more sensitive to antibiotic exposure when compared to the nitrifying culture. Although high antibiotic concentrations were applied, the half maximal i

Links between S-adenosylmethionine and Agr-based quorum sensing for biofilm development in Listeria monocytogenes EGD-e.

Listeria monocytogenes is the causative agent of human listeriosis which has high hospitalization and mortality rates for individuals with weakened immune systems. The survival and dissemination of L. monocytogenes in adverse environments can be reinforced by the formation of biofilms. Therefore, this study aimed to understand the mechanisms underlying listerial biofilm development. Given that both nutrient availability and quorum sensing (QS) have been known as the factors influencing biofilm development, we hypothesized that the signal from a sentinel metabolite S-adenosylmethionine (SAM) an

Engineering controlled antimicrobial delivery across biofilm matrix barriers: mechanics, transport and nanocarrier design.

Bacterial biofilms are a major challenge for antimicrobial therapy because they function not only as structured microbial communities, but also as complex biological barriers to drug delivery. Biofilm-associated bacteria are embedded within an extracellular polymeric substance (EPS) matrix that provides mechanical stability, regulates antimicrobial transport and generates heterogeneous microenvironments characterised by nutrient limitation, oxygen gradients and metabolically diverse bacterial subpopulations. As a result, therapeutic failure in mature biofilms often reflects a mismatch between

Quorum sensing molecules enhance cadmium detoxification and cellular removal in the marine diatom Phaeodactylum tricornutum.

Acyl-homoserine lactones (AHLs) are ubiquitous bacteria-derived quorum-sensing molecules in marine environments and play important roles in regulating microbial physiological processes. However, their influence on metal detoxification and handling in microalgae remains poorly understood. Here, we investigated the effects of exogenous AHLs on cell growth, metabolic responses, and cadmium (Cd) removal in the marine diatom Phaeodactylum tricornutum under both Cd-free and Cd-contaminated conditions. It was demonstrated that AHLs slightly inhibited the growth of P. tricornutum and increased extrace

Artificial Phages with Biocatalytic Spikes for Synergistically Eradicating Antibiotic-Resistant Biofilms.

Antibiotic-resistant pathogens have become a global public health crisis, especially biofilm-induced refractory infections. Efficient, safe, and biofilm microenvironment (BME)-adaptive therapeutic strategies are urgently demanded to combat antibiotic-resistant biofilms. Here, inspired by the fascinating biological structures and functions of phages, the de novo design of a spiky Ir@Co3O4 particle is proposed to serve as an artificial phage for synergistically eradicating antibiotic-resistant Staphylococcus aureus biofilms. Benefiting from the abundant nanospikes and highly active Ir sites, the

Chromium(VI) and nitrate removal from groundwater using biochar-assisted zero valent iron autotrophic bioreduction: Enhancing electron transfer efficiency and reducing EPS accumulation.

Current strategies primarily utilize heterotrophic or mixotrophic bioreduction for the simultaneous removal of Cr(VI) and NO3- from groundwater. However, given the oligotrophic nature of groundwater, autotrophic bioreduction could be more appropriate, though it remains notably underdeveloped. Here, an autotrophic bioreduction technology utilizing biochar (BC)-assisted zero valent iron (ZVI) is proposed. The pyrolysis temperature of BC was optimized to enhance electron transfer efficiency and reduce extracellular polymeric substances (EPS) accumulation. BC500, with the superior electron transfe

[Influence of Hydraulic Retention Time on the Treatment of Polluted River Water by an Activated Carbon Rotating Biological Contactor].

The study combined activated carbon with a rotating biological contactor. The activated carbon adsorption experiments were conducted and the biofilm was formed by using the river water directly. The effects of different hydraulic retention times on the removal of NH4+-N, TP, and permanganate index and on the biofilm properties were investigated at the optimum rotational speed of the disc. Experimental results are as follows. The Freundlich isotherm showed that activated carbon had better adsorption properties for NH4+-N, TP, and permanganate index in water. When the disk speed was 3 r·min-1, th

Microbial potential for construction and degradation of the cryoconite biofilm matrix: Implications for carbon and nutrient cycling in Antarctic cryoconite holes.

Cryoconite granule formation underpins the physical structure and biogeochemical functioning of glacier cryoconite holes. Central to this process is the extracellular polymeric substance (EPS) matrix, produced by microorganisms, which binds microbial cells and mineral particles into stable aggregates while retaining organic matter and nutrients. However, the physiological traits governing the EPS matrix construction and degradation remain poorly understood. Here, we investigated microbial capacities for biofilm formation, EPS-related substrate utilization, and extracellular hydrolysis in Antar

Destruction of Staphylococcus aureus biofilms by combining an antibiotic with subtilisin A or calcium gluconate.

In S. aureus biofilms, bacteria are embedded in a matrix of extracellular polymeric substances (EPS) and are highly tolerant to antimicrobial drugs. We thus sought to identify non-antibiotic substances with broad-spectrum activity able to destroy the EPS matrix and enhance the effect of antibiotics on embedded biofilm bacteria. Among eight substances tested, subtilisin A (0.01 U/mL) and calcium gluconate (CaG, Ca2+ 1.25 mmol/L) significantly reduced the biomass of biofilms formed by at least 21/24 S. aureus isolates. Confocal laser scanning microscopy confirmed that they both eliminated nearly

Porcupine-Quill-Inspired Shape-Morphing Biointerfaces Disassemble Biofilms.

Mature biofilms are difficult to eradicate because the extracellular polymeric substance (EPS) matrix shields embedded bacteria from chemical and mechanical attack. Here, we report a porcupine-spine-inspired photo-responsive dynamic offensive bio-interface that dismantles mature biofilms through electrically actuated mechanical penetration and light-driven oxidative stress. A high-aspect-ratio nano-dagger array undergoes reversible nanoscale deformation (∼110 nm) under an alternating electric field, while a photo-responsive heterostructure generates a surface photovoltage (∼364 mV) that enhanc

Characterization of biofilm formation by Borrelia burgdorferi in vitro.

Borrelia burgdorferi, the causative agent of Lyme disease, has long been known to be capable of forming aggregates and colonies. It was recently demonstrated that Borrelia burgdorferi aggregate formation dramatically changes the in vitro response to hostile environments by this pathogen. In this study, we investigated the hypothesis that these aggregates are indeed biofilms, structures whose resistance to unfavorable conditions are well documented. We studied Borrelia burgdorferi for several known hallmark features of biofilm, including structural rearrangements in the aggregates, variations i

Structure-Function Analysis of the Curli Accessory Protein CsgE Defines Surfaces Essential for Coordinating Amyloid Fiber Formation.

Curli amyloid fibers are produced as part of the extracellular biofilm matrix and are composed primarily of the major structural subunit CsgA. The CsgE chaperone facilitates the secretion of CsgA through CsgG by forming a cap at the base of the nonameric CsgG outer membrane pore. We elucidated a series of finely tuned nonpolar and charge-charge interactions that facilitate the oligomerization of CsgE and its ability to transport unfolded CsgA to CsgG for translocation. CsgE oligomerization in vitro is temperature dependent and is disrupted by mutations in the W48 and F79 residues. Using nuclea

Extraction of extracellular polymeric substances from aerobic granule with compact interior structure.

Extracellular polymeric substances (EPS) were extracted from aerobic granules of compact interior structure using seven extraction methods. Ultrasound followed by the chemical reagents formamide and NaOH outperformed other methods in extracting EPS from aerobic granules of compact interior. The collected EPS revealed no contamination by intracellular substances and consisted mainly of proteins, polysaccharides, humic substances and lipids. The quantity of extracted proteins exhibited a weak correlation with quantity of extracted carbohydrates but no correlation with quantity of extracted humic

Treatment of Pseudomonas aeruginosa infectious biofilms: Challenges and strategies.

Pseudomonas aeruginosa, a Gram-negative bacterium, is one of the major pathogens implicated in human opportunistic infection and a common cause of clinically persistent infections such as cystic fibrosis, urinary tract infections, and burn infections. The main reason for the persistence of P. aeruginosa infections is due to the ability of P. aeruginosa to secrete extracellular polymeric substances such as exopolysaccharides, matrix proteins, and extracellular DNA during invasion. These substances adhere to and wrap around bacterial cells to form a biofilm. Biofilm formation leads to multiple a
