Boosting Piezoelectricity and Mechanical Properties of Silk Fibroin Films via Polyethylene Glycol Phase Manipulation.

Piezoelectric silk fibroin (SF) shows great potential in fabricating sustainable wearable electronics toward health monitoring and human-machine interfacing, yet the processing difficulty and brittleness restrict its practical applications severely. This study proposes a simple strategy to achieve the boosted piezoelectricity and mechanical properties in electrospun regenerated silk fibroin (RSF) nanofiber mats by adding polyethylene glycol (PEG) with different molecular weights to induce the formation of β-sheets via hydrogen bonding and the regulation of PEG phase morphology. Specifically, R

Development of a compartmentalized silk fibroin-reinforced GelMA hydrogel platform for kidney-inspired tissue engineering.

Chronic kidney disease (CKD) and the limited availability of donor organs continue to drive the need for advanced biomaterial systems for renal tissue engineering and in vitro kidney modeling. However, reproducing the spatial complexity and compartmentalized organization of native renal tissue remains a significant challenge. In this study, a compartmentalized silk fibroin (SF)-reinforced gelatin methacrylate (GelMA) hydrogel platform with spatially defined architectures inspired by renal tissue organization was developed. Initially, SF-reinforced GelMA hydrogels with tunable stiffness were fa

Fabrication of Mechanically Adjustable Silk Fibroin-Based Hydrogels with Excellent Biocompatibility for Skin Wound Repair.

Silk fibroin (SF) hydrogels have attracted widespread attention as promising wound dressings owing to their excellent biocompatibility and tunable structure. However, constructing SF hydrogels with easily adjustable mechanical properties and clarified structure-function relationships remains challenging. Herein, we fabricated a series of mechanically tunable SF hydrogels using ethylene glycol diglycidyl ether (EGDE) as a biocompatible crosslinker. The epoxy groups of EGDE readily react with the nucleophilic amino and hydroxyl groups on SF chains via ring-opening reactions, forming stable coval

Nitrogen Plasma-Modified Silk Fibroin Films Promote Wettability and Airway Epithelial Cell Growth.

The regulation of cell behavior by surface properties is a central principle for bioengineering tissue scaffolds. The effects of surface wettability on airway epithelial cells are particularly instrumental to functional epithelium formation but are not well understood, thus restricting bioengineered airway graft efficacy. Here, we demonstrate that nitrogen low-temperature plasma (N2 LTP) treatment enhances the surface wettability of two types of 3D-printed silk fibroin films (opaque and transparent) and promotes the growth of primary human bronchial epithelial cells (HBECs) in submerged cultur

Silk proteins for 3D bioprinting: Integrating fibroin structural support with sericin cell-supportive capability.

Three-dimensional (3D) bioprinting requires inks and bioinks that combine appropriate processing and mechanical properties with a biologically supportive environment capable of sustaining cell viability and proliferation. Silk-based biomaterials have attracted considerable attention in biofabrication due to their tunable properties, processability, and structural similarity to extracellular matrix components. While most silk-based inks/bioinks rely primarily on silk fibroin (SF) for structural integrity, the use of silk sericin (SS) as a cell-supportive component remains comparatively underexp

Famotidine-loaded chitosan hybridized fibroin nanoparticles exhibit outstanding efficacy in ameliorating peptic ulcer.

Famotidine (FMD) is an H₂-receptor antagonist with limited oral bioavailability and a short plasma half-life (2.5-4 h). Silk fibroin-chitosan nanoparticles (FBN-CS-NPs) represent a novel nanocarrier approach for treating peptic ulcers, combining biocompatibility, mucoadhesiveness, and pH-sensitive release. These nanoparticles were developed by electrostatic interactions between fibroin and chitosan to improve FMD encapsulation and mucosal retention and potentiate its therapeutic efficacy. The optimized formulation demonstrated a particle size of 94.4 ± 3.6 nm, a zeta potential of +65.3 ± 4.6 m

Biomimetic Silk Fibroin Scaffolds Functionalized with Hydroxyapatite and Platelet Growth Factors for Bone Tissue Engineering.

Non-union fractures represent a significant clinical challenge requiring innovative therapeutic approaches. Silk fibroin (SF) scaffolds have gained recognition as advantageous biomaterials for bone tissue engineering due to their biocompatibility and mechanical characteristics. This study investigated the biocompatibility and osteoinductive potential of SF scaffolds functionalized with hydroxyapatite (HA) and loaded with platelet growth factors (PGFs) using hematopoietic stem cells (HSCs). SF scaffolds were prepared and functionalized with HA through methanol impregnation, while PGFs were obta

A Triple-Layered Composite Scaffold of Silk Fibroin and Decellularized Amniotic Membrane for Bladder Tissue Engineering.

Augmentation cystoplasty has different side effects in urinary bladder reconstruction. Accordingly, it is necessary to develop substitutes using natural and synthetic biomaterials to address current problems. This study evaluates the potential of a triple-layered composite scaffold for bladder regeneration. The triple-layered scaffold consists of a silk fibroin (SF) film blended with polyethylene oxide (PEO), a decellularized human amniotic membrane (DHAM), and a lyophilized SF sponge, which is seeded with adipose tissue-derived stem cells (ADSCs) encapsulated in collagen hydrogel. The mechani

In vitro mineralization and osteogenesis effects of the silk fibroin hydrogels mineralized with SBF and CaCl2/Na2HPO4 methods.

In vitro mineralization has been proposed as promising approach for fabricating mineralized scaffolds and repairing bone defects. In this study, various mineralized glycidyl methacrylate-modified silk fibroin (GMA-SF) hydrogels were developed using two mineralization techniques: calcium chloride/sodium phosphate (CaCl2/Na2HPO4) solution immersion and simulated body fluid (SBF) solution immersion. Compared to the SBF immersion method, the CaCl2/Na2HPO4 method produced a greater quantity of minerals on the surface of GMA-SF hydrogels within the same mineralization duration and cycle numbers. High-performance silk fibroin/hyaluronic acid interpenetrating network hydrogel microneedles for diabetes management.

Hydrogel microneedles (MNs) gained more attentions for diabetes treatments owing to their biocompatibility and versatility. However, the inherent fragility and instability of hydrogels pose limitations on their efficacy in biomedical applications. To overcome this limitation, we developed interpenetrating network hydrogels (IPNs) by incorporating silk fibroin (SF) and methacrylated hyaluronic acid (HAMA). These hydrogels exhibit rapid formation, structural stability, mechanical robustness, and sustainability through photo-crosslinking without the need for crosslinking agents. The hydrogels dem

Dexamethasone-loaded fibroin nanoparticles promote retinal reattachment in rats by regulating the Th17/Treg balance.

Retinal detachment (RD) is a common acute blinding eye disease, and dexamethasone (DEX), an adrenocorticosteroid, shows protective effects against RD. However, its poor water solubility and low bioavailability limit its effectiveness. To address this, we developed SF@DEX nanomaterials and investigated their therapeutic potential and mechanisms in RD. The nanomaterials were successfully synthesized and characterized, achieving 90% encapsulation efficiency and releasing 60% of DEX within 12 h.In vitro, phagocytosis was measured by flow cytometry, and enzyme-linked immunosorbent assay determined

Layer-by-Layer Deposition of Regenerated Silk Fibroin─An Approach to the Surface Coating of Biomedical Implant Materials.

Biomaterials and coating techniques unlock major benefits for advanced medical therapies. Here, we explored layer-by-layer (LbL) deposition of silk fibroin (SF) by dip coating to deploy homogeneous films on different materials (titanium, magnesium, and polymers) frequently used for orthopedic and other bone-related implants. Titanium and magnesium specimens underwent preceding plasma electrolytic oxidation (PEO) to increase hydrophilicity. This was determined as surface properties were visualized by scanning electron microscopy and contact angle measurements as well as Fourier transform infrar

Structural-Functional Pluralistic Modification of Silk Fibroin via MOF Bridging for Advanced Wound Care.

Silk fibroin (SF) is widely used to fabricate biomaterials for skin related wound caring or monitoring, and its hydrogel state are preferred for their adaptability and easy to use. However, in-depth development of SF hydrogel is restricted by their limited mechanical strength, increased risk of infection, and inability to accelerate tissue healing. Therefore, a structure-function pluralistic modification strategy using composite system of metal organic framework (MOF) as bridge expanding SF's biomedical application is proposed. After developing the photocuring and bonding SF hydrogel, a MOF dr

Birefringent Silk Fibroin Hydrogel Constructed via Binary Solvent-Exchange-Induced Self-Assembly.

Birefringent hydrogels have a strong potential for applications in biomedicine and optics as they can modulate the optical and mechanical anisotropy in confined two-dimensional geometries. However, production of birefringent hydrogels with hierarchical structures, mechanical properties, and biorelated behavior that are analogous to biological tissues is still challenging. Starting from the silk fibroin (SF)-ionic liquid solution system, this study aimed to rationally design a "binary solvent-exchange-induced self-assembly (BSEISA)" strategy to produce birefringent SF hydrogels (SFHs). In this

Controlled radical polymerization of hydrophilic and zwitterionic brush-like polymers from silk fibroin surfaces.

Bombyx mori silk fibroin is a fibrous protein whose tunable properties and biocompatibility have resulted in its utility in a wide-variety of applications, including as drug delivery vehicles, wound dressings, and tissue engineering scaffolds. Control of protein and cell attachment is vital to the performance of biomaterials, but silk fibroin is mostly hydrophobic and interacts nonspecifically with cells and proteins. Silk functionalised with hydrophilic polymers reduces attachment, but the low number of reactive sites makes achieving a uniform conjugation a persistent challenge. This work pre

Novel fabrication method of the peritoneal dialysis filter using silk fibroin with urease fixation system.

During the last decade, there has been a great advance in the kidney dialysis system by wearable artificial kidney (WAK) system for end-stage renal disease patients. Uremic solute removal and water regeneration system are the most prerequisite for WAK to work properly. In this study, we designed a filtering membrane system by using immobilized urease silk fibroin filter and evaluated its comparative effectiveness with a PVDF filtering system in peritoneal dialysate regeneration system by urea removal efficacy. We evaluated this membrane's characteristic and performances by conducting SEM-EDX a

Rapid and repeatable fabrication of high A/R silk fibroin microneedles using thermally-drawn micromolds.

Thermal drawing is a versatile rapid prototyping method that can freely form microneedle (MN) structures with ultra-high aspect ratio without relying on any complex and expensive process. However, it is still challenging to repeatedly produce MNs with identical shapes using this thermal drawing due to small fluctuations in processing conditions such as temperatures, drawing speeds, drawing heights, or parallelism in the drawing setup. In addition, thermal drawing is only applicable to thermoplastic materials and most natural biomaterials are incompatible with this method. Thus, we propose use

Functional recovery guided by an electrospun silk fibroin conduit after sciatic nerve injury in rats.

The aim of this study was to evaluate the regenerative capacity of a newly developed nerve guidance conduit using electrospun silk fibroin (SFNC) implanted in a 10-mm defect of the sciatic nerve in rats. After evaluating the physical properties and cytocompatibility of SFNC in vitro, rats were randomly allocated into three groups: defect only, autograft and SFNC. To compare motor function and abnormal sensation among groups, ankle stance angle (ASA) and severity of autotomy were observed for 10 weeks after injury. Immunostaining with axonal neurofilament (NF) and myelin basic protein (MBP) ant

Process-Driven Modulation of Structure and Cytocompatibility in Electrospun Silk Fibroin Membranes through Controlled Degumming.

The degumming process technique critically determines the structural and biological performance of silk fibroin-based biomaterials. In this study, silk fibroin obtained from Bombyx mori cocoons was subjected to a controlled experimental design that combined sodium carbonate (Na2CO3), sodium lauryl ether sulfate (SLES), and reaction time to modulate the efficiency of sericin removal. Degumming effectiveness was quantified by mass loss and chemical analysis (FTIR-ATR, solid-state 13C NMR), while structural modifications were evaluated by X-ray diffraction. Alkaline-based degumming treatments (C4

Evaluation of Colombian silk fibroin hydrogels functionalized with recombinant LSECtin for intervertebral disc tissue engineering.

This study evaluated the potential of Colombian silk fibroin hydrogels functionalized with recombinant LSECtin (rLSECtin) for intervertebral disc tissue engineering. In silico analysis indicated that rLSECtin may interact with glycans similar to those found in human adipose-derived stem cells (ADSCs) through residues Asn105, Asp106, Glu93, Asn87, and Glu85, which coordinate calcium to form hydrogen bonds with GlcNAc hydroxyl groups. rLSECtin was successfully produced, purified, and incorporated into silk fibroin hydrogels. Thermal analysis showed that the hydrogels were resistant to sterilizat
