Determining the optimal fluoride concentration in drinking water for fluoride endemic regions in South India.

Fluoride ion in drinking water is known for both beneficial and detrimental effects on health. The prevalence of fluorosis is mainly due to the intake of large quantities of fluoride through drinking water owing to more than 90% bioavailability. The objective of this study is to predict optimal fluoride level in drinking water for fluoride endemic regions by comprising the levels of fluoride and other water quality parameters in drinking water, prevalence of fluorosis, fluoride intake through water, food and beverages such as tea and coffee and also considering the progressive accumulation of

Fluoride-induced oxidative stress of osteoblasts and protective effects of baicalein against fluoride toxicity.

The key role of osteoblasts in skeletal fluorosis makes the exploration of the possible mechanisms of the fluoride-induced oxidative stress of osteoblasts of great importance. In this article, the in vitro effects of fluoride on the oxidative stress of osteoblasts are presented. To study the inhibitory effect of baicalein on the oxidative stress of osteoblasts, the antioxidant activity of baicalein was evaluated for osteoblasts exposed to fluoride. Calvarial osteoblasts were prepared and respectively treated with alpha-MEM (5% calf serum) containing 0.5, 1.0, 2.0, 4.0, 8.0, 12.0, and 20.0 mg/L

Slow-release fluoride glass devices: in vivo fluoride release and retention of the devices in children.

AIMS: To study the fluoride release and retention of new-shaped glass slow-release F devices (SRFD) in vivo in children. METHODS: A group of 15 children had a SRFD with 17.4% or 14.8 mgm F with a relative solubility of 294, attached intra-orally on the buccal surface of one of their maxillary permanent molars. The devices were monitored for six months. RESULTS: The retention rate of the devices was 93% and the mean salivary F levels in the child volunteers was 01.17+/-0.1 ppm F compared with 0.025 ppm F baseline levels. The difference was statistically significant (p<0.01). There were no sys

Nucleophilic identity substitution reactions. The reaction between hydrogen fluoride and protonated alkyl fluorides.

The gas phase reactions between HF and the protonated alkyl fluorides MeFH+, EtFH+, Pr(i)FH+, and Bu(t)FH+ have been studied using ab initio methods. The potential energy profiles for both nucleophilic substitution (S(N)2) and elimination (E2) pathways have been investigated. Both backside Walden inversion and frontside nucleophilic substitution reaction profiles have been generated. Backside substitution is very favourable, but shows relatively little variation with the alkyl group. Frontside substitution reaction barriers are only slightly higher than the barrier for backside substitution fo

Binding of phosphate, aluminum fluoride, or beryllium fluoride to F-actin inhibits severing by gelsolin.

Actin exhibits ATPase activity of unknown function that increases when monomers polymerize into filaments. Differences in the kinetics of ATP hydrolysis and the release of the hydrolysis products ADP and inorganic phosphate suggest that phosphate-rich domains exist in newly polymerized filaments. We examined whether the enrichment of phosphate on filamentous ADP-actin might modulate the severing activity of gelsolin, a protein previously shown to bind differently to ATP and ADP actin monomers. Binding of phosphate, or the phosphate analogs aluminum fluoride and beryllium fluoride, to actin fil

Fluoride levels in human parotid saliva following ingestion of fluoride compounds of varying solubility.

Healthy dental students received oral doses of 10.0 mg F daily in the form of compounds of widely varying solubilities. Parotid saliva F concentration and urinary F excretion were highest with NaF, Na2PO3F, and SnF2, lowest with CaF2, and intermediate with AlF3. Solubility of the ingested fluoride compound was a controlling factor in the amount of F excreted in the urine, the peak F concentration reached in parotid saliva, and the duration of a significant elevation in salivary F.

Benzene-1,3-disulfonyl Fluoride Mediated Synthesis of Glycosyl Fluorides from Glycosyl Hemiacetals.

We developed an efficient method for synthesizing glycosyl fluorides from glycosyl hemiacetals using benzene-1,3-disulfonyl fluoride (S2) as the fluorinating agent in combination with the base DBU (1,8-diazabicyclo[5.4.0]undec-7-ene) under mild conditions. This approach enabled the successful synthesis of 17 glycosyl fluorides with various functional groups, including disaccharides and deoxysulfates, with yields of up to 99%. The utility of these synthesized glycosyl fluorides was further demonstrated in the synthesis of important O- and C-glycosides.

Arginine Accelerates Sulfur Fluoride Exchange and Phosphorus Fluoride Exchange Reactions between Proteins.

Sulfur fluoride exchange (SuFEx) and phosphorus fluoride exchange (PFEx) click chemistries are advancing research across multiple disciplines. By genetically incorporating latent bioreactive unnatural amino acids (Uaas), these chemistries have been integrated into proteins, enabling precise covalent linkages with biological macromolecules and paving the way for new applications. However, their suboptimal reaction rates in proteins limit effectiveness, and traditional catalytic methods for small molecules are often incompatible with biological systems or in vivo applications. We demonstrated th

Fluoride exposure and indicators of thyroid functioning in the Canadian population: implications for community water fluoridation.

BACKGROUND: There are concerns that altered thyroid functioning could be the result of ingesting too much fluoride. Community water fluoridation (CWF) is an important source of fluoride exposure. Our objectives were to examine the association between fluoride exposure and (1) diagnosis of a thyroid condition and (2) indicators of thyroid functioning among a national population-based sample of Canadians. METHODS: We analysed data from Cycles 2 and 3 of the Canadian Health Measures Survey (CHMS). Logistic regression was used to assess associations between fluoride from urine and tap water sampl…

Comparative antimicrobial activity of an essential oil and an amine fluoride/stannous fluoride mouthrinse in vitro.

Although laboratory studies are not necessarily predictive of clinical activity; they can help to elucidate mechanisms underlying clinical activity when the latter has been established. In a recent clinical study, an essential oil mouthrinse (Listerine Antiseptic) was shown to be significantly more effective than an amine fluoride/stannous fluoride mouthrinse (Meridol) in inhibiting supragingival plaque formation. This paper reports the results of laboratory studies comparing the antimicrobial effectiveness of these 2 mouthrinses using a kill kinetics assay and a plaque biofilm kill assay. In
