Eosinophil and foam cell accumulation in lungs of Sprague-Dawley rats fed purified, biotin-deficient diets.

One hundred twenty male Sprague-Dawley rats (3 weeks old) were given biotin-deficient diets containing ovalbumin as the protein source. Ten control rats of the same origin were fed a commercially available purified diet that used casein as a protein source. Eosinophils and histiocytes were observed at a higher frequency in lungs of rats fed the purified diets containing ovalbumin than in the controls. Foam cells were confined to subpleural and peribronchial regions, reacting positively to anti-lysozyme antibody. The incidence of pulmonary histiocytosis was 76/120 rats (63.3%) in the groups fed…

Evidence of foam cell and cholesterol crystal formation in macrophages incubated with oxidized LDL by fluorescence and electron microscopy.

Macrophage-derived foam cells are a prominent component of developing atherosclerotic lesions. We describe an in vitro model of foam cell formation which mimics some aspects of the evolution of foam cells in mature atherosclerotic lesions. Thioglycollate-elicited mouse peritoneal macrophages were incubated with copper-oxidized LDL (ox-LDL) for periods up to 168 hr. Identifiable foam cells were present after incubation with ox-LDL at 24, 72, and 168 hr. Control cells incubated without ox-LDL did not form foam cells. Fluorescence microscopy after staining with Nile red exhibited progressive accu…

Diffuse panbronchiolitis: diagnosis and distinction from various pulmonary diseases with centrilobular interstitial foam cell accumulations.

Diffuse panbronchiolitis (DPB) is a clinicopathologic entity characterized histologically by chronic inflammation principally affecting the respiratory bronchioles. Few pathologists outside Japan are familiar with this entity. The most distinctive pathologic feature of DPB is chronic inflammation and an accumulation of foam cells in the walls of the respiratory bronchioles, adjacent alveolar ducts, and alveoli (PB unit lesion). The differential diagnosis is important both clinically and histologically because of the similarity of DPB to other chronic airway diseases. We report seven cases of D…

Phenotype and Response to PAMPs of Human Monocyte-Derived Foam Cells Obtained by Long-Term Culture in the Presence of oxLDLs.

Cholesterol-laden, foam macrophages constitute the most characteristic component of human atherosclerotic plaques. Persistent uptake of oxLDLs results in accumulation of lipid bodies inside the cells and determines their phenotype and subsequent functions. In this work, we describe the phenotype of human monocyte-derived foam cells obtained by differentiation in the constant presence of oxLDLs for 30 days (prolonged-hMDFCs). Although neither the total cellular nor the cell surface expression of Toll-like receptors (TLR) was regulated by oxLDLs, the prolonged-hMDFCs changed dramatically their r…

Krüppel-like Factor 2 (KLF2) in the Regulation of Lipid Accumulation, ROS, and Mitochondrial Functions During Foam Cell Formation in RAW264.7 Cells.

Foam cell formation, a hallmark of early atherosclerotic lesion development, is closely associated with mitochondrial dysfunction and excessive reactive oxygen species (ROS) production. Disruption in mitochondrial activity leads to electron leakage, elevated ROS generation, and collapse of mitochondrial membrane potential, contributing to vascular pathogenesis. In this study, we investigated the role of Krüppel-like factor 2 (KLF2), a transcription factor known for its vasculoprotective effects, in regulating mitochondrial function during foam cell (FC) formation in RAW264.7 cells. This study

Immunocytochemical characteristics of perivascular and intratumoral foam cells in neoplasms of neuroectodermal origin with lysozyme, alfa1-antitrypsin, protein S-100 and GFAP.

Investigations were performed to become acquainted with the immunohistochemical features of foam cells localized perivascular and intratumoral in neoplasms of neuroectodermal origin. Antibodies against lysozyme (muramidase) (LO), alfa 1-antitrypsin (AT), protein S-100 and glial fibrillary acid protein (GFAP) were used. A weak or medium intense reaction result has been obtained in the cytoplasm of the foam cells if antibodies against LO, and alfa 1-antitrypsin and almost negative result if antibodies against protein S-100 and GFAP were used. Only very few cells which differ from the foam cells …

Epigallocatechin-3-gallate prevents TNF-α-induced NF-κB activation thereby upregulating ABCA1 via the Nrf2/Keap1 pathway in macrophage foam cells.

The ATP-binding membrane cassette transporter A1 (ABCA1) plays a protective role in the development of atherosclerosis for the reverse cholesterol transport process. Epigallocatechin-3-gallate (EGCG), which exists abundantly in green tea, exerts an anti-atherosclerotic effect via anti-inflammatory and metabolic regulation activities. Many genes and proteins related to lipid metabolism are involved in the lowering cholesterol effects of EGCG. However, effects of EGCG on ABCA1 have rarely been described. In the study presented here, we found that exposure of macrophage foam cells to TNF-α result…

PCSK9-Mediated Inflammation in Foam Cell Formation and Exploring the Biologically Active Compounds Derived From Natural Resources.

Atherosclerosis develops as a persistent inflammatory condition that gradually alters the vascular structure and condition, in which the formation of macrophage-derived foam cells plays a central role in disease initiation and progression. Although proprotein convertase subtilisin/kexin type 9 (PCSK9) is widely recognised for its involvement in low-density lipoprotein receptor regulation and systemic lipid homeostasis, growing evidence indicates that PCSK9 also exerts a direct influence on macrophage inflammatory responses and intracellular lipid metabolism. Within atherosclerotic lesions, PCS…

Decoding dexmedetomidine's multiple mechanisms in atherosclerosis: effects on lipid metabolism, inflammation, cholesterol efflux, and foam cell formation.

Atherosclerosis is a leading cause of cardiovascular disease and is characterized by dysregulated lipid metabolism and chronic vascular inflammation. Dexmedetomidine (DEX) has been reported to exert anti-inflammatory, antioxidant and lipid-lowering effects across several disease settings, yet its potential therapeutic impact on atherosclerosis remains unclear. In this study, we investigated the anti-atherosclerotic efficacy of DEX and explored the underlying mechanisms in vivo and in vitro. An atherosclerosis model was established by feeding ApoE−/− mice a Western diet for 13 weeks, followed b

Biodegradable cell-laden starch foams for the rapid fabrication of 3D tissue constructs and the application in neural tissue engineering.

Cells encapsulation by biomaterials has been widely studied as a strategy of building tissue construct in tissue engineering. Conventional encapsulation of cells using hydrogels often needs the polymerization process or relatively complex molding process. In this study, we developed a facile strategy for the in situ fabrication of biodegradable cell-laden starch foams. By utilizing the unique gelatinization property of starch, cell-laden starch foams with tunable architecture were rapidly prepared in a green and biological-friendly process. The bubble size and stiffness of starch foams could b

Mitochondrial Redox Failure Links Senescence-like Foam Cell Stress to Ferroptosis and Plaque Non-Resolution in Atherosclerosis.

Foam cells are central to atherosclerotic plaque development, but their pathological significance in advanced lesions extends beyond lipid accumulation. Under chronic exposure to oxidized lipoproteins, cholesterol crystals, inflammatory cytokines, hypoxia, lysosomal stress, and mitochondrial injury, lipid-loaded foam cells of macrophage and vascular smooth muscle cell (VSMC) origin may acquire maladaptive stress phenotypes. In this focused review, we propose a redox-threshold model, defined as the transition point at which mitochondrial antioxidant and metabolic buffering capacity is exceeded,…

The foam cell-derived exosomes exacerbate ischemic white matter injury via transmitting metabolic defects to microglia.

Atherosclerosis (AS) has been shown to be an independent risk factor for vascular cognitive impairment (VCI), but the mechanisms remain unclear. Here, we found that AS circulating exosomes exacerbated ischemic white matter injury and VCI. Exosomes originating from macrophage-derived foam cells targeted microglia. Mechanistically, foam cell-derived exosomes transmitted redox imbalance, mitochondrial dysfunction, and metabolic defects to microglia via the miR-101-3p-Nrf2-Slc2a1 axis. Anti-miR-101-3p or activation of Nrf2, both genetically and pharmacologically, could antagonize AS exosomes and a…

Electrochemical oxidation of gaseous benzene on a Sb-SnO2/foam Ti nano-coating electrode in all-solid cell.

An all-solid cell with a solid polymer electrolyte was applied to electrochemical oxidation of low-concentration indoor gaseous aromatic pollution. Antimony-doped tin dioxide nanocoatings deposited on a titanium foam substrate (Ti/Sb-SnO2) with different Sb/Sn ratios (4.8-14.0 mol%) and loading weight of Sb-SnO2 (4.4-7.7 mg cm-2) were used as dimensionally stable anodes. Sn and Sb were homogeneously dispersed on the substrate, and a crack-free nanocoating was built when the loading of nanocoating was increased to 6.3 mg cm-2. The activity tests for oxidation of benzene showed that 40 ppm gaseo
