Transcriptome Analysis of Tryptophan-Induced Resistance against Potato Common Scab.

Potato common scab (CS) is a worldwide soil-borne disease that severely reduces tuber quality and market value. We observed that foliar application of tryptophan (Trp) could induce resistance against CS. However, the mechanism of Trp as an inducer to trigger host immune responses is still unclear. To facilitate dissecting the molecular mechanisms, the transcriptome of foliar application of Trp and water (control, C) was compared under Streptomyces scabies (S) inoculation and uninoculation. Results showed that 4867 differentially expressed genes (DEGs) were identified under S. scabies uninocula

Dwarf and Increased Branching 1 controls plant height and axillary bud outgrowth in Medicago truncatula.

Optimizing plant architecture is an efficient approach for breeders to increase crop yields, and phytohormones such as gibberellins (GAs) play an important role in controlling growth. Medicago truncatula is a model legume species, but the molecular mechanisms underlying its architecture are largely unknown. In this study, we examined a tobacco retrotransposon Tnt1-tagged mutant collection of M. truncatula and identified dwarf and increased branching 1 (dib1), which exhibited extreme dwarfism and increased numbers of lateral branches. By analysis of the flanking sequences of Tnt1 insertions in

Dissecting the Regulatory Network of Leaf Premature Senescence in Maize (Zea mays L.) Using Transcriptome Analysis of ZmELS5 Mutant.

Leaf premature senescence largely determines maize (Zea mays L.) grain yield and quality. A natural recessive premature-senescence mutant was selected from the breeding population, and near-isogenic lines were constructed using Jing24 as the recurrent parent. In the near-isogenic lines, the dominant homozygous material was wild-type (WT), and the recessive material of early leaf senescence was the premature-senescence-type ZmELS5. To identify major genes and regulatory mechanisms involved in leaf senescence, a transcriptome analysis of the ZmELS5 and WT near-isogenic lines (NILs) was performed

The dynamic regulatory network of stamens and pistils in papaya.

BACKGROUND: Papaya exhibits three sex types: female (XX), male (XY), and hermaphrodite (XYh), making it an unusual trioecious model for studying sex determination. A critical aspect of papaya sex determination is the pistil abortion in male flowers. However, the regulatory networks that control the development of pistils and stamens in papaya remain incompletely understood. RESULTS: In this study, we identified three organ-specific clusters involved in papaya pistils and stamens development. We found that pistil development is primarily characterized by the significant expression of auxin-rel

Secondary product glucosyltransferase and putative glucosyltransferase expression during Citrus paradisi (c.v. Duncan) growth and development.

Flavonoids are secondary metabolites that have significant roles in plant defense and human nutrition. Glucosyltransferases (GTs) catalyze the transfer of sugars from high energy sugar donors to other substrates. Several different secondary product GTs exist in the tissues of grapefruit making it a model plant for studying their structure and function. The goal of this investigation was to determine the expression patterns of seven putative secondary product GTs during grapefruit growth and development by quantifying mRNA expression levels in the roots, stems, leaves, flowers, and mature fruit

A Xanthomonas effector increases virulence quantitatively by inducing at least 3 minor susceptibility genes.

The transcription activator-like effector Tal12a is widely conserved among Xanthomonas campestris pv. campestris strains that cause black rot in Brassica crops. However, its role in disease remains unclear. To investigate how Tal12a contributes to pathogenesis, we combined transcriptomic profiling of cauliflower leaves infected with Tal12a-expressing strains, prediction of TALE-binding elements, and heterologous expression assays in Nicotiana benthamiana. The aim of this approach was to identify candidate susceptibility genes. Artificial TALEs were further employed to validate the contribution

Genome-wide identification of genes associated with enhanced carbon secretion in cluster roots of Lupinus albus L.

Several plant species adapted to low-phosphorus (P) conditions develop cluster roots, specialized structures that release organic acids and acid phosphatases (APases) to increase inorganic phosphate (Pi) availability. White lupin (Lupinus albus L.) is used as a model for studying cluster root function. Using a positron-emitting tracer imaging system (PETIS), we previously observed spot-like carbon (C) secretion patterns in the cluster roots of white lupin, the amounts of which differed widely among spots, suggesting variation in secretion activity among cluster roots. Here, we combined PETIS w

Identification of genes controlling grain size in barley through RNA-seq and weighted gene co-expression network analysis.

To elucidate the molecular mechanisms underlying grain size formation in barley, this study selected two distinct barley cultivars with significant differences in grain dimensions: the large-grained cultivar 'jianglinhuang' and the small-grained cultivar 'heiseyeqingke'. We conducted a comprehensive comparative analysis by integrating phenotypic measurements with RNA-seq data to elucidate molecular-level differences between barley varieties with distinct grain sizes. The study focused on three critical grain developmental stages. Dynamic phenotypic monitoring revealed that grain length and thi

Downregulation of HbFPS1 affects rubber biosynthesis of Hevea brasiliensis suffering from tapping panel dryness.

Tapping panel dryness (TPD) is a century-old problem that has plagued the natural rubber production of Hevea brasiliensis. TPD may result from self-protective mechanisms of H. brasiliensis in response to stresses such as excessive hormone stimulation and mechanical wounding (bark tapping). It has been hypothesized that TPD impairs rubber biosynthesis; however, the underlying mechanisms remain poorly understood. In the present study, we firstly verified that TPD-affected rubber trees exhibited lower rubber biosynthesis activity and greater rubber molecular weight compared to healthy rubber tree

Overexpression of the Liriodendron tulipifera BOP2 Gene (LtuBOP2) Affects Leaf Margin Development in Transgenic Arabidopsis thaliana.

BLADE-ON-PETIOLE 2 (BOP2) plays a pivotal role in leaf morphogenesis. Liriodendron tulipifera is a suitable model for exploring the molecular mechanisms underlying leaf serration formation, which are largely unknown. Here, we isolated the full-length LtuBOP2 gene and its promoter from L. tulipifera and characterized its function in leaf morphogenesis through multidimensional approaches. The spatiotemporal expression pattern of LtuBOP2 indicated the high expression of LtuBOP2 in stems and leaf buds. We constructed LtuBOP2 promoter, fused the promoter sequences to the β-glucuronidase (GUS) gene,

Functional Genomics and Genetic Control of Compound Leaf Development in Medicago truncatula: An Overview.

Diverse forms of leaves are present in nature. However, the regulatory mechanisms that underpin the development of diverse leaf forms remain enigmatic. The initiation of leaf primordia from the periphery of shoot apical meristem (SAM) requires downregulation of the class 1 knotted-like homeobox KNOXI proteins. In plants with simple leaves, this downregulation is permanent, consistent with leaves being determinant organs. In most of plants with compound leaves, the KNOXI proteins are reactivated in developing leaf primordia, and this reactivation is required for the development of compound leav

TsCYP94B1 regulates male fertility in androdioecious Tapiscia sinensis via jasmonic acid metabolism.

Tapiscia sinensis Oliv. (Tapisciaceae), a functionally androdioecious species, displays male individuals with higher pollen viability compared to hermaphrodites. This study elucidates the molecular basis underlying reduced pollen viability and abnormal tapetum development in hermaphroditic flowers. Integrated transcriptomic and functional analyses identified TsCYP94B1, a jasmonate-responsive gene overexpressed in hermaphroditic flowers, as the homolog of AtCYP94B1 from Arabidopsis thaliana that catabolizes bioactive JA-Ile into 12-OH-JA-Ile and 12-COOH-JA-Ile. Elevated TsCYP94B1 expression in

Genome-wide identification and functional analysis of the BES1-like (VfBES1) gene family in Vernicia fordii reveals its role in floral development.

BACKGROUND: Vernicia fordii Hemsl (also known as Tung tree), an significant commercial oil-producing tree species, is a monoecious and diclinous species with male and female flowers on the same inflorescence; however, the molecular mechanisms governing its floral sex determination remain elusive, particularly the genetic basis underlying the skewed female-to-male flower ratio and the evolutionary dynamics of sex-related gene families, which severely restrict targeted breeding for yield enhancement. In the model plant Arabidopsis, the BRI1 EMS SUPPRESSOR 1 (BES1) transcription factor family pla

Differential expression of genes encoding the light-dependent and light-independent enzymes for protochlorophyllide reduction during development in loblolly pine.

The expression patterns of the two distinct subfamilies of genes (designated porA and porB) encoding the light-dependent NADPH:protochlorophyllide oxidoreductases (PORs) in loblolly pine (Pinus taeda L.) were examined. Transcripts arising from both gene subfamilies were shown to be present at high levels in the cotyledons of dark-grown pine seedlings and to a lesser extent in their stems. Exposure of dark-grown seedlings to light resulted in increased levels of both porA and porB transcripts, as well as increased levels of mRNAs encoding other photosynthesis-related gene products, suggesting t

Coordinated homeostasis of essential mineral nutrients: a focus on iron.

In plants, iron (Fe) transport and homeostasis are highly regulated processes. Fe deficiency or excess dramatically limits plant and algal productivity. Interestingly, complex and unexpected interconnections between Fe and various macro- and micronutrient homeostatic networks, supposedly maintaining general ionic equilibrium and balanced nutrition, are currently being uncovered. Although these interactions have profound consequences for our understanding of Fe homeostasis and its regulation, their molecular bases and biological significance remain poorly understood. Here, we review recent know

Root secreted metabolites and proteins are involved in the early events of plant-plant recognition prior to competition.

The mechanism whereby organisms interact and differentiate between others has been at the forefront of scientific inquiry, particularly in humans and certain animals. It is widely accepted that plants also interact, but the degree of this interaction has been constricted to competition for space, nutrients, water and light. Here, we analyzed the root secreted metabolites and proteins involved in early plant neighbor recognition by using Arabidopsis thaliana Col-0 ecotype (Col) as our focal plant co-cultured in vitro with different neighbors [A. thaliana Ler ecotype (Ler) or Capsella rubella (C

Transcriptional insights into molecular events occurring during tuber maturation of tigernut (Cyperus esculentus L.).

Tigernut (Cyperus esculentus L. var. sativus Boeck., Cyperaceae), an emerging oilcrop, is unique for producing desiccation-tolerant tubers. However, thus far, the underlying mechanisms are largely unknown. To gain insights into molecular events occurring during tuber maturation, deep sequencing was used to investigate gene expression profiles of two representative tuber stages of Reyan3, i.e., immature and mature, whose tubers were characterized by light and dark brown in color and with the moisture content of 74.80% and 48.30%, respectively. A total of 16,499 genes were shown to be expressed,

Comparative transcriptomics reveals hormone signaling and MADS-box genes in divergent development of inflorescences and tendrils in grapevine lateral shoots.

Hormone signaling and MADS-box genes regulate grapevine tendril and inflorescence growth divergence, offering molecular insights for managing tendril growth. Grapevine (Vitis vinifera L.) tendrils and inflorescences are homologous organs; however, their divergent development has important agronomic consequences because excessive tendril growth increases vineyard management costs. To explore the regulatory mechanisms, we compared the inflorescence-prone cultivar 'Einset Seedless' (ENT) with the tendril-prone cultivar 'Pinot Noir' (PN) using anatomical observation, transcriptome analysis of spec

From dormancy to germination: Transcriptome analysis reveals the potential involvement of heat shock proteins in gibberellin-induced seed dormancy release in Solanum torvum.

Solanum torvum, a superior vegetable grafting rootstock and medicinal Solanaceae plant, exhibits strong seed dormancy, which limits its commercial cultivation. Among various strategies explored to improve the germination rate of S. torvum, exogenous application of gibberellin (GA) has been shown to be effective. In this study, a GA concentration of 2.5 mM was established as the optimal for breaking dormancy in S. torvum seeds. Transcriptome analysis of dry, water-soaked, GA-soaked, and GA-induced germinated seeds was conducted to investigate the molecular mechanism of GA-mediated dormancy rele
