Parkinson's disease (PD) is characterized by dopaminergic neuron loss and α-synuclein aggregation in the substantia nigra pars compacta (SNpc). It is a multifactorial disorder with motor and non-motor manifestations and growing evidence suggests that gastrointestinal dysfunction may precede motor onset. Sex differences influence PD risk, onset and clinical features, with men exhibiting higher prevalence and earlier onset, driven by hormonal, genetic, and metabolic factors. The gut microbiota communicates bidirectionally with the central nervous system (CNS) via the gut-brain axis, modulating n

The human microbiota comprises a vast and diverse array of microorganisms that play critical roles in maintaining health and modulating diseases. Engineered live biotherapeutic products (eLBPs) harness genetically modified microbes to perform defined therapeutic functions within the host. A central challenge in developing effective eLBPs is the rational selection of an appropriate microbial chassis, which requires consideration of safety, genetic tractability, functional performance and the target host niche. As different body sites present distinct physiological and biochemical conditions tha

Hidradenitis suppurativa (HS) is an inflammatory disease of the skin with a chronic, relapsing-remitting course. The pathogenesis of the disease is poorly understood and involves multiple factors, including genetics, environment, host-microbe interactions, and immune dysregulation. In particular, the composition of the cutaneous microbiome shifts as the disease progresses, although it is unclear whether this is a primary or secondary process. Trials with immunomodulatory therapy elucidate the role of specific immune pathways and cytokine signaling in disease mechanism, such as TNF-α, IL-1β, IL

Cryptococcus neoformans is a major cause of fungal meningoencephalitis in immunocompromised patients. Despite recent advances in the genetics and molecular biology of C. neoformans, and improved techniques for molecular epidemiology, aspects of the ecology, population structure, and mode of reproduction of this environmental pathogen remain to be established. Application of recent insights into the life cycle of C. neoformans and its different ways of engaging in sexual reproduction under laboratory conditions has just begun to affect research on the ecology and epidemiology of this human path

Reproductive microbiota is known to modulate host immunity, reproductive physiology, and longevity, yet their interactions with host genetics, sex, and mating status remain relatively underexplored in most insects. Some studies suggest that species (and even populations) can vary in the microbial communities associated with their reproductive tissues. Sexually transmitted microbes can also directly influence population growth and reproductive fitness, making them relevant even for applied insect management strategies. The economically relevant, Black Soldier Fly (BSF; Hermetia illucens), is kn

The immune-oncology-microbiome (IOM) trio highlights the significant role of microbiomes in cancer progression by modulating immune evasion, genomic instability, and inflammation-key hallmarks of cancer. While microbiomes can exert both protective and detrimental effects on tumor development and treatment response, the mechanistic underpinnings-particularly those involving intratumoral microbiomes-remain poorly understood. To elucidate these dynamics, we frame the interplay between cancer, immune cells, and microbiomes through the lens of the Anna Karenina Principle (AKP)-using Leo Tolstoy's a

Microbial populations exhibit both genetic and non-genetic heterogeneity. In the clinical context, this heterogeneity is of concern as it provides subsets of microbial populations with enhanced immune evasion abilities and antimicrobial resistance. Fungal pathogens are of special concern as fungal diseases and antifungal resistance are increasing worldwide and similarities between eukaryotic cells make it challenging to identify targets that are toxic to fungi without also harming the human host. Engineered live biotherapeutic products (eLBPs) could provide an alternative and complementary app

Terpenoids represent the largest and most structurally diverse class of secondary metabolites, with extensive applications in the pharmaceutical, nutraceutical, cosmetic, agricultural, fragrance, and biofuel industries. The growing demand for these compounds has resulted in extensive exploitation of plant-derived terpenoids, raising concerns regarding resource availability and sustainability. Consequently, microbial production has emerged as a promising alternative because of the high genetic tractability, rapid growth, and ease of metabolic engineering offered by microbial hosts. Various meta

Celiac disease (CD) is a chronic immune-mediated disorder triggered by dietary gluten in genetically predisposed individuals and characterized by intestinal inflammation, epithelial damage, and loss of immune tolerance. While a strict lifelong gluten-free diet (GFD) remains the cornerstone of treatment, accumulating evidence indicates that it does not consistently restore gut microbiome composition or function, and many patients experience persistent symptoms despite good dietary adherence. The gut microbiome has emerged as a key modulator of immune homeostasis, intestinal barrier integrity, a

Increasing evidence indicates that alterations in the gastrointestinal microbiota are associated with the initiation and progression of gastrointestinal cancers. Abundant evidence suggests that gastrointestinal microbiota dysbiosis plays an important role in the development of gastrointestinal cancers by producing proinflammatory and immunosuppressive signals. This review examines the role of microbiome dysbiosis in the pathogenesis of gastrointestinal malignancies. The composition of the microbiota varies greatly among individuals and depends on various factors such as age, diet, lifestyle, g

Mycobacterium tuberculosis is one of most successful pathogens of mankind, infecting one-third of the global population and claiming two million lives every year. The ability of the bacteria to persist in the form of a long-term asymptomatic infection, referred to as latent tuberculosis, is central to the biology of the disease. The persistence of bacteria in superficially normal tissue was recognized soon after the discovery of the tubercle bacillus, and much of our knowledge about persistent populations of M. tuberculosis dates back to the first half of the last century. Recent advances in m

BACKGROUND: Atopic dermatitis (AD), food sensitization (FS), and food allergy (FA) frequently co-occur in infancy, but the factors underlying distinct atopic phenotypes remain unclear. Although FLG null mutations are major genetic risk factors for AD, they explain only part of disease heritability, suggesting a potential role for the skin microbiome. This study examined how early-life skin microbiome composition and its interaction with host genetics contribute to distinct atopic phenotypes in infancy.
METHODS: We analyzed > 1000 skin swabs from 429 infants in the VITALITY cohort using deep s

BACKGROUND: Rheumatoid arthritis (RA) is a systemic chronic inflammatory disease that leads to joint destruction and functional disability due to the targeting of self-antigens present in the synovium, cartilage, and bone. RA is caused by a number of complex factors, including genetics, environment, dietary habits, and altered intestinal microbial flora. Microorganisms in the gut bind to nod-like receptors and Toll-like receptors to regulate the immune system and produce various metabolites, such as short-chain fatty acids (SCFAs) that interact directly with the host. We developed an innovative approach to estimate the occurrence and extent of fecal pollution sources for urban river catchments. The methodology consists of 1) catchment surveys complemented by literature data where needed for probabilistic estimates of daily produced fecal indicator (FIBs, E. coli, enterococci) and zoonotic reference pathogen numbers (Campylobacter, Cryptosporidium and Giardia) excreted by human and animal sources in a river catchment, 2) generating a hypothesis about the dominant sources of fecal pollution and selecting a source targeted monitoring design, and 3) verifying t

Rumen, one of the most productive diverse microbial habitats plays a vital role in the breakdown of feed to produce energy for maintenance and milk production in cattle. Culture-based procedures could identify only 10% of microbial species present in the rumen. Kerala, one of the southern states of India, owns only one native cattle breed, the Vechur cattle, which is noted for its short stature, disease resistance and adaptability to hot humid climate. Lower population density and decreased milk production potential of Vechur cattle led to the development of crossbred cattle of Kerala, with hi

The host-microbiota relationship has evolved to shape mammalian physiology, including immunity, metabolism, and development. Germ-free models are widely used to study microbial effects on host processes such as immunity. Here, we find that both germ-free and T cell-deficient mice exhibit a robust sebum secretion defect persisting across multiple generations despite microbial colonization and T cell repletion. These phenotypes are inherited by progeny conceived during in vitro fertilization using germ-free sperm and eggs, demonstrating that non-genetic information in the gametes is required for

The golden jackal (Canis aureus) is a medium-sized canid widely distributed across Europe and the Mediterranean region. As a synanthropic species, it often inhabits areas near human settlements and is associated with zoonotic diseases such as rabies. This study characterizes the golden jackal’s fur microbiota and examines its interactions with host traits, fecal microbiota, host genetics and environmental factors. Analysis of the 16S rRNA amplicons indicated that the golden jackal fur microbiota is dominated by four bacteria phyla (Proteobacteria, Firmicutes, Actinobacteriota and Bacteroidota)

Among all non-communicable diseases, Cardiovascular Diseases (CVDs) stand as the leading global cause of mortality. Within this spectrum, Myocardial Infarction (MI) strikingly accounts for over 15 % of all deaths. The intricate web of risk factors for MI, comprising family history, tobacco use, oral health, hypertension, nutritional pattern, and microbial infections, is firmly influenced by the human gut and oral microbiota, their diversity, richness, and dysbiosis, along with their respective metabolites. Host genetic factors, especially allelic variations in signaling and inflammatory marker
