GENETIC REGULATORY MECHANISMS AT THE POPULATION LEVEL IN MAN.

The distribution of the Rhesus and ABO blood groups, thalassemia, and glucose-6-phosphate dehydrogenase deficiency in Europe and in Jewish populations argues for a regulatory mechanism at the population level which adjusts gene frequencies so that the overall genetic load is not excessive, since gene frequencies giving a high genetic load at one locus are often associated with those giving low genetic loads at other loci.

Genetic Determinants of Infectious Diseases in the Qatari Population.

BACKGROUND AND OBJECTIVES: Human populations show striking phenotypic disparities in traits and diseases, including responses to infectious diseases (IDs). The impact of host genetic diversity on infection susceptibility and outcomes is increasingly recognized, yet remains largely unknown in Qatar. Here, we explored the distribution of infection-related host genetic variants in Qatar. MATERIALS AND METHODS: Infection-related genetic variants from the GWAS Catalog were analyzed in 6047 Qatari whole genomes from the Qatar Genome Program (QGP) and compared with populations from the 1000 Genomes

Genetic Causes of Oculocutaneous Albinism in Pakistani Population.

Melanin pigment helps protect our body from broad wavelength solar radiation and skin cancer. Among other pigmentation disorders in humans, albinism is reported to manifest in both syndromic and nonsyndromic forms as well as with varying inheritance patterns. Oculocutaneous albinism (OCA), an autosomal recessive nonsyndromic form of albinism, presents as partial to complete loss of melanin in the skin, hair, and iris. OCA has been known to be caused by pathogenic variants in seven different genes, so far, according to all the currently published population studies. However, the detection rate

Population-based Genetic Testing for Precision Prevention.

Global interest in genetic testing for cancer susceptibility genes (CSG) has surged with falling costs, increasing awareness, and celebrity endorsement. Current access to genetic testing is based on clinical criteria/risk model assessment which uses family history as a surrogate. However, this approach is fraught with inequality, massive underutilization, and misses 50% CSG carriers. This reflects huge missed opportunities for precision prevention. Early CSG identification enables uptake of risk-reducing strategies in unaffected individuals to reduce cancer risk. Population-based genetic testi

Compliance for genetic screening in the Arab population in Israel.

BACKGROUND: Genetic screening tests for cystic fibrosis (CF), fragile X (FRAX) and spinal muscular atrophy (SMA) have been offered to the entire Arab population of Israel in the last few years. Since 2008, screening for CF is provided free of charge, but for FRAX and SMA the screening is privately funded with partial coverage by complementary health insurance programs. OBJECTIVES: To assess the compliance of Arab couples with regard to genetic screening tests, and the factors that affect their decisions. METHODS: We analyzed compliance for genetic screening tests at the Emek Medical Center G

Population heterogeneity in the genetic control of serum urate.

Scanning of the genome in Caucasian cohorts for genes that control serum urate levels have revealed eight confirmed associations. Knowledge of genetic control of serum urate in other populations can be extrapolated from the study of gout, a condition of extreme hyperuricemia; three urate transport genes (SLC2A9, ABCG2, and SLC17A1/NPT1) have been studied in diverse populations (Caucasian, Asian, and Polynesian). Between-population heterogeneity is evident in allele frequency and association with gout, notably within Polynesian populations, which are defined by a geographic region and shared an

Susceptibility to lung tuberculosis--a population-genetic study.

The aim of this study was to compare the genetic structure of lung tuberculosis (LTB) patients (Pts) to healthy controls i.e. to evaluate dependency of LTB occurrence and course of the disease in regard to subject's genetic constitution. The study group consisted of 100 LTB Pts, while the same number of healthy non-sibling individuals served as controls. The groups were analyzed with respect to homozygous recessive characteristics (HRC) that served as genetic markers, which were studied in each subject. Twenty-three HRCs were chosen according to HRC-test rules. The results show significant dif

Antenatal genetic counselling: implications for population screening.

Only 1-2% of newborns have a congenital abnormality, yet it is responsible for the much greater proportion of mortality and morbidity in infancy and in childhood as well as during pregnancy. Because of this prevalence, there have been many developments in screening and diagnosis of congenital abnormality. Yet there are a number of limitations about how the screening and diagnostic service is currently operated. These limitations centre on the information that is provided, who undertakes the counselling, and their training and support provided to parents in making important decisions.

Genetic polymorphism of HLA-DR in the Japanese population.

As a result of carrying out sequence analyses on the HLA-DR genes of several Japanese donors, we found three new DRB1 alleles, DRB1-12b, DRB1-14c, and DRB1-JX6, that had not been identified using immunological procedures. Sequence-specific oligonucleotide (SSO) probes directed against various DRB1 alleles, including the above three, enabled DNA typing of all the DRB types in the Japanese population and calculation of their gene frequencies based on this typing to be carried out for the first time. The SSO-DNA typing yielded higher DR13 and DR14 gene frequencies than those reported by serologic

Population genetic data of 30 insertion-deletion markers in Punjabi population of Pakistan.

Insertion-deletion polymorphism (Indels) is valuable diallelic markers for forensic as well as parentage analysis. The Investigator DIPplex Kit (Qiagen) contains thirty autosomal Indels markers along with amelogenin. These thirty markers were tested in the Pakistani Punjabi Population but no significant deviations were observed from Hardy-Weinberg equilibrium rule expectations (Bonferroni corrected) except HLD58, HLD56, HLD99, and HLD40. The mean expected and observed heterozygosity was found 0.4701 and 0.4667 respectively; combined matching probability was computed as 7.31867 × 10-13. However

Genomic landscape of the human vaginal microbiome is linked to host genetics and population of origin.

The vaginal microbiome is essential for women's health, yet its genomic diversity and interaction with the host remain incompletely characterized. Here we present the Global Vaginal Metagenome-assembled Genomes catalog, an extensive repository of vaginal microbial genomes generated by integrating 10,665 in-house Chinese metagenomes, with 2,967 publicly available metagenomes and 1,433 bacterial isolates. The catalog comprises 65,055 genomes from 890 prokaryotes, 11 eukaryotes and 6,590 viral taxonomic units, many not represented in public reference databases. We investigate virus-bacteria inter

Shotgun sequencing of airborne eDNA achieves rapid assessment of whole biomes, population genetics and genomic variation.

Biodiversity and its associated genetic diversity are being lost at an unprecedented rate. Simultaneously, the distributions of flora, fauna, fungi, microbes and pathogens are rapidly changing. Novel technology can help to capture and record genetic diversity before it is lost and to measure population shifts and pathogen distributions. Here we report the rapid application of shotgun long-read environmental DNA (eDNA) analysis for non-invasive biodiversity, genetic diversity and pathogen assessments from air. We also compared air eDNA with water and soil eDNA. Coupling long-read sequencing wit

Comparative analysis of population genetics between Schistosoma japonicum-infected and uninfected Oncomelania hupensis snails using microsatellites.

Источник пометил публикацию как отозванную

Schistosomiasis is now at a low prevalence in China, but schistosome infections in intermediate host Oncomelania hupensis have been constantly reported. Previous studies have revealed a behavioral modification of snails associated with infection. However, there is a lack of comparative population genetics between schistosome-infected and uninfected snails. This study aimed to investigate any differences in genetic diversity and/or structure in O. hupensis associated with Schistosoma japonicum infection, with the purpose of providing information on snail control. Snail survey was performed in D

Comparative population genetics of the Antarctic grenadier fish (Macrourus whitsoni) and its parasitic copepod (Lophoura szidati).

Large ectoparasitic copepods, nearly as long as the length of their host, are often found on Antarctic fish, yet little is known about their biology. In this study, we investigated the genetic structure and host-parasite relationships in Macrourus whitsoni, a deep-sea fish, and its copepod ectoparasite, Lophoura szidati, using 10,569 biallelic SNPs collected from 38 copepods and 5,009 biallelic SNPs from 35 fish individuals across three populations in the Ross Sea, Antarctica. The Fst, DAPC, and admixture analyses revealed distinct genetic patterns between the two species. For M. whitsoni, our

Population genetics of Aedes albopictus in the port cities of Hainan Island and Leizhou Peninsula, China.

BACKGROUND: Aedes albopictus is an important vector of arboviral diseases, transmitting yellow fever, dengue fever, chikungunya and Zika. Monitoring its population genetic diversity and genetic differentiation has become essential for the control of infectious disease epidemics, especially in the functional areas of ports of entry. Population genetic monitoring of Ae. albopictus in the port area can help in the monitoring of port mosquito invasions and establishing port sanitary and quarantine measures to prevent the introduction and transmission of vector-borne diseases. METHODS: Seventeen p

Population genomics of Group B Streptococcus reveals the genetics of neonatal disease onset and meningeal invasion.

Group B Streptococcus (GBS), or Streptococcus agalactiae, is a pathogen that causes preterm births, stillbirths, and acute invasive neonatal disease burden and mortality. Here, we investigate bacterial genetic signatures associated with disease onset time and meningeal tissue infection in acute invasive neonatal GBS disease. We carry out a genome-wide association study (GWAS) of 1,338 GBS isolates from newborns with acute invasive disease; the isolates had been collected annually, for 30 years, through a national bacterial surveillance program in the Netherlands. After controlling for the popu

Heritability of non-HLA genetics in coeliac disease: a population-based study in 107 000 twins.

BACKGROUND AND OBJECTIVE: Almost 100% individuals with coeliac disease (CD) are carriers of the human leucocyte antigen (HLA) DQ2/DQ8 alleles. Earlier studies have, however, failed to consider the HLA system when estimating heritability in CD, thus violating an underlying assumption of heritability analysis. We examined the heritability of CD in a large population-based sample of twins, considering HLA. DESIGN: In a population-representative sample of 107 912 twins, we identified individuals with CD (equal to villous atrophy) through biopsy reports from all Swedish pathology departments. We c
