Histamine H3 receptor agonists decrease hypothalamic histamine levels and increase stereotypical biting in mice challenged with methamphetamine.

The effects of the histamine H(3) receptor agonists (R)-α-methylhistamine, imetit and immepip on methamphetamine (METH)-induced stereotypical behavior were examined in mice. The administration of METH (10 mg/kg, i.p.) to male ddY mice induced behaviors including persistent locomotion and stereotypical behaviors, which were classified into four categories: stereotypical head-bobbing (1.9%), circling (1.7%), sniffing (14.3%), and biting (82.1%). Pretreatment with (R)-α-methylhistamine (3 and 10 mg/kg, i.p.) significantly decreased stereotypical sniffing, but increased stereotypical biting induce…

Histamine increases anti-CD3 induced IL-5 production of TH2-type T cells via histamine H2-receptors.

Besides its proinflammatory functions histamine released from basophils and mast cells during immediate-type hypersensitivity reactions is known to inhibit several lymphocyte functions like IL-2 and gamma-IFN production. Recently, it has been shown that T helper cells of type 2 phenotype (TH2) represent the T cell fraction which may play a pivotal role in the promotion of the allergic inflammatory eosinophilic late-phase reaction by secretion of cytokines, especially IL-4 and IL-5. We have investigated the effect of histamine on anti-CD3 induced IL-4 and IL-5 production by TH2 cells. Histamine…

Histamine levels and activities of histamine metabolizing enzymes in mammalian ocular tissues: comparison between day and night.

Iris, ciliary body, choroid and retina of the cow, rabbit and man contain histamine; the retinas are less rich in histamine (55-135 ng/g tissue) than the uvea (209-3073 ng/g tissue). The tissues show no diamine oxidase activity, no or marginal histidine decarboxylase activity, and well expressed histamine-N-methyltransferase activity. The killing of the rabbits at noon, light-midnight and dark-midnight revealed some day-night and light-dark dependent variations in both histamine content and histamine-N-methyltransferase activity in the iris-ciliary body, choroid and retina. It is suggested tha…

Histamine Neuroimaging in Stress-Related Disorders.

Although histamine plays a major role in animal models of stress-related disorders, human neuroimaging data are sparse. Histamine H1 receptors in the human brain were first imaged by Professor Kazuhiko Yanai in 1992 by using 11C-doxepin, a potent ligand of H1 receptors, and positron emission tomography (PET). Subsequent work revealed that H1 receptors are reduced in the prefrontal and anterior cingulate cortices in patients with major depressive disorders. A sex difference in H1 receptor binding in the brain has also been found, with women exhibiting more abundant H1 receptor binding than men.…

Opioid toxicity: histamine, hypersensitivity, and MRGPRX2.

Insights into the pathophysiology of many non-immune-mediated drug reactions referred to as toxicities, sensitivities, intolerances, or pseudoallergies have resulted from research identifying the mastocyte-related G-protein-coupled receptor (GPCR) member X2 (MRGPRX2), a human mast cell receptor mediating adverse reactions without the involvement of antibody priming. Opioid-induced degranulation of mast cells, particularly morphine, provoking release of histamine and other preformed mediators and causing hemodynamic and cutaneous changes seen as flushing, headache and wheal and flare reactions …

Considering histamine in functional gastrointestinal disorders.

In westernized countries, adverse reactions to ingested foods are reported to affect up to 20% of the population. Functional, nonspecific, non-allergic gastrointestinal complaints are mainly due to the intolerance/malabsorption of carbohydrates (lactose and fructose), proteins (gluten), and biogenic amines (histamine). Food intolerance/malabsorption is defined by one or several of the above mentioned food components not being degraded and/or absorbed properly within the gastrointestinal tract. Food intolerance/malabsorption causes variable, functional, nonspecific, non-allergic gastrointestina…

Histamine: neural circuits and new medications.

Histamine was first identified in the brain about 50 years ago, but only in the last few years have researchers gained an understanding of how it regulates sleep/wake behavior. We provide a translational overview of the histamine system, from basic research to new clinical trials demonstrating the usefulness of drugs that enhance histamine signaling. The tuberomammillary nucleus is the sole neuronal source of histamine in the brain, and like many of the arousal systems, histamine neurons diffusely innervate the cortex, thalamus, and other wake-promoting brain regions. Histamine has generally e…

Histamine H2-receptor antagonists for urticaria.

BACKGROUND: Urticaria is a common skin disease characterised by itching weals or hives, which can occur almost anywhere on the body. There a … with a clinical diagnosis of urticaria of any duration or of any subtype. Studies including H1-antihistamines for chronic urticaria are the topic of a separate Cochrane review; thus, they were not included in this review. DATA COLLECTION AND ANALYSIS: Two reviewers independently assessed trial quality and extracted and analysed data. MAIN RESULTS: Four s …

Physiological significance of ECL-cell histamine.

In the oxyntic mucosa of the mammalian stomach, histamine is stored in ECL cells and in mucosal mast cells. In the rat, at least 80 percent of oxyntic mucosal histamine resides in the ECL cells. Histamine is a key factor in the regulation of gastric acid secretion. Following depletion of ECL-cell histamine by treatment with alpha-fluoromethylhistidine (alpha-FMH), basal acid secretion was reduced, and gastrin-stimulated acid secretion was abolished. Vagally-induced acid secretion (by insulin injection or pylorus ligation) was unaffected by alpha-FMH treatment but inhibited by an H2 antagonist.…

Helicobacter pylori produces histamine and spermidine.

The mechanism by which Helicobacter pylori associates with the development of upper gastrointestinal diseases is still not well understood. Toxic metabolites of H. pylori are discussed as possible factors. We were interested to investigate, whether biogenic amines might be involved. Ten monocultures of H. pylori from the antrum of patients with H. pylori associated diseases were analyzed for the content of the biogenic amine histamine. The bacteria were isolated on Columbia blood agar with Skirrow's supplement in a microaerophilic environment. After three passages onto fresh agar plates the ba…

Cutaneous histamine metabolism in chronic urticaria.

Impaired metabolism of histamine in the skin of patients with chronic idiopathic urticaria (CIU) might explain the observed enhanced and prolonged skin responses to intradermal histamine. Histamine metabolism was measured in homogenates from unaffected forearm skin in nine patients with CIU and in skin of age- and sex-matched control subjects with a radiochromatographic assay, and the results are expressed as nanograms of histamine metabolized per milligram of protein per hour. Endogenous histamine content was determined by RIA. There was a highly significant increase in endogenous histamine c…

Histamine release from leucocytes in migraine.

The spontaneous histamine release (SHR) from leucocytes in migraine patients during symptom-free periods has been investigated, and also the influence of migraine sufferer serum on histamine release (HR) from blood donor leucocytes. The leucocytes were isolated according to the Day method and histamine assay was made by the Shore spectrofluorometric method modified by Bergendorff and Uväs (1). SHR in the 17 female migraine patients was significantly higher (39.7% +/- 13.1%) than in 8 controls (25.4% +/- 4.2%). Migraine serum increased HR from blood donor leucocytes by 14.6% +/- 20.2% compared …

Tricyclic antidepressants and histamine H1 receptors.

Tricyclic antidepressants and some structurally related compounds were tested for their ability to antagonize histamine H1 and muscarinic acetylcholine receptors of cultured mouse neuroblastoma cells. As a group, tertiary amine tricyclic antidepressants tended to be more potent than secondary amine drugs at both receptors. The most potent antihistamine, doxepin hydrocholoride, was about 4 times more potent than amitriptyline hydrochloride, about 800 times more potent than diphenhydramine hydrochloride, and about 8,000 times more potent than desipramine hydrochloride, the least potent tricyclic…
