[Study of interaction of polystirolsulphonate with polymerization degree of 8 and polyallylamin with bilayer lipids membranes].

Interaction of polystirolsulphonate with polymerization degree of 8 (PSS-8) and polyallylamin PAA (molecular mass 60 kilodaltons) with viruses from bloodline of paramixo- and orthomixoviruses by the example of measles virus, parotitis and flu leads to the decreasing of infective activity. The possible mechanism of viral inhibitive action of these chemical compounds is damaging of interfacial antigenic proteins of paramixo- and orthomixoviruses. In this study it was detected the change of surface tension of bilayer lipid membrane in the presence of PSS-8 and PAA. The change of surface tension l

Enhanced Brownian ratchet molecular separation using a self-spreading lipid bilayer.

A new approach is proposed for two-dimensional molecular separation based on the Brownian ratchet mechanism by use of a self-spreading lipid bilayer as both a molecular transport and separation medium. In addition to conventional diffusivity-dependence on the ratchet separation efficiency, the difference in the intermolecular interactions between the target molecules and the lipid bilayer is also incorporated as a new separation factor in the present self-spreading ratchet system. Spreading at the gap between two ratchet obstacles causes a local change in the lipid density at the gap. This eff

Adhesion and merging of lipid bilayers: a method for measuring the free energy of adhesion and hemifusion.

Lipid bilayers can be induced to adhere to each other by molecular mediators, and, depending on the lipid composition, such adhesion can lead to merging of the contacting monolayers in a process known as hemifusion. Such bilayer-bilayer reactions have never been systematically studied. In the course of our studies of membrane-active molecules, we encountered such reactions. We believe that they need to be understood whenever bilayer-bilayer interactions take place, such as during membrane fusion. For illustration, we discuss three examples: spontaneous adhesion between phospholipid bilayers in…

Properties and behaviour of tetracyclic allopsoralen derivatives inside a DPPC lipid bilayer model.

Allopsoralens are angular psoralen derivatives presenting advantages over the parent compound because of monofunctional DNA-photobinding and consequent lower toxicity. Allopsoralen molecules with three different substituents and different protonation states were studied using the molecular dynamics technique. The location of these molecules when inside the lipid bilayer is of major importance because their photochemical properties can change with the environment. Also, the ability of psoralens to form photoadducts with unsaturated phospholipids depends on the preference of the molecules to loc

In excess proIAPP, increased aggregation levels are finally observed, however, which could be attributed to seed-induced cofibrillation of proIAPP.

The effect of microwave radiation on the stability and formation of gramicidin-A channels in lipid bilayer membranes.

The effects of microwaves on the single-channel kinetics of gramicidin-A channels in lipid bilayer membranes were examined. Attempts were made to separate thermal and athermal effects by accurate measurements of temperature at the site of the membrane and by relating the measured parameters to their previously characterized temperature dependence. It was found that microwave radiation does not affect single-channel conductance or channel life time to a degree that is significantly different from that expected of a purely thermal effect. On the other hand, the rate of channel formation is decre

Black widow spider venom: effect of purified toxin on lipid bilayer membranes.

A purified toxin (the B5 fraction) from black widow spider venom added to the solution on one side of a lipid bilayer membrane interacts irreversibly with the membrane to produce a continuous, linear rise of membrane conductance with time. Conductances greater than 10(-4) reciprocal ohm per square centimeter can eventually be attained without any loss of membrane stability. Membranes treated with toxin are ideally selective for alkali cations over anions and are substantially permeable to calcium ion. These effects of the toxin result from the formation of permanent channels in the membrane of

Visible-Light Sensitized Isomerization in the Lipid Bilayer Enables Activation of a Transmembrane Transporter.

Currently, there is major interest in embedding light-responsive (supra)molecular systems in phospholipid bilayer membranes, for example, to control transmembrane transport processes. Most of these systems are operated by harmful UV-light. Although in solution phase the isomerization of photoswitchable systems has been achieved using visible-light-absorbing photosensitizers, no cases are known yet of sensitized isomerization in a lipid bilayer environment. Herein, we show that the active isomer of a stiff-stilbene based chloride transporter can be generated in situ by irradiation and energy tr

Regeneration of tethered bilayer lipid membrane biosensors for repetitive use in toxin detection.

The development of reusable biosensors based on tethered bilayer lipid membranes (tBLMs) is crucial to achieve cost-effective and sustainable pathogen detection. In this study, we investigated the regeneration of tBLMs assembled on fluorine-doped tin oxide (FTO) substrates using organic silane-based molecular anchors and a lipid mixture of dioleoylphosphatidylcholine and cholesterol. The sensors were exposed to α-hemolysin (αHL), a pore-forming toxin from Staphylococcus aureus, and regenerated by a two-step bilayer removal protocol. Electrochemical impedance spectroscopy (EIS) was used to asse

Pegylated gold nanoparticles interact with lipid bilayer and human serum albumin and transferrin.

Gold nanoparticles (AuNPs) are potentially applicable in drug/nucleic acid delivery systems. Low toxicity, high stability, and bioavailability are crucial for the therapeutic use of AuNPs and they are mainly determined by their interactions with proteins and lipids on their route to the target cells. In this work, we investigated the interaction of two pegylated gold nanoparticles, AuNP14a and AuNP14b, with human serum proteins albumin (HSA) and transferrin (Tf) as well as dimyristoyl-phosphatidylcholine (DMPC) liposomes, which can be a representative of biomembranes. We showed that AuNP14a/b

Effect of C60 on the phase transition behavior of a lipid bilayer under high pressure.

Interactions between fullerenes and cells and effects on the main transition of lipid bilayers have attracted much attention in biophysics in recent years. By employing coarse-grained molecular dynamics simulations, we obtained the temperature-pressure phase diagrams of a dipalmitoylphosphatidylcholine bilayer, which exhibits a gel phase and a fluid phase, with variation of the C60 versus lipid ratios. The simulation results show that the critical area per lipid at the fluid-gel main phase transition boundary increases with the increasing ratios of C60. A critical area per lipid of 0.594 ± 0.0…

Continuity of Monolayer-Bilayer Junctions for Localization of Lipid Raft Microdomains in Model Membranes.

We show that the selective localization of cholesterol-rich domains and associated ganglioside receptors prefer to occur in the monolayer across continuous monolayer-bilayer junctions (MBJs) in supported lipid membranes. For the MBJs, glass substrates were patterned with poly(dimethylsiloxane) (PDMS) oligomers by thermally-assisted contact printing, leaving behind 3 nm-thick PDMS patterns. The hydrophobicity of the transferred PDMS patterns was precisely tuned by the stamping temperature. Lipid monolayers were formed on the PDMS patterned surface while lipid bilayers were on the bare glass sur…

Following Natures Lead: On the Construction of Membrane-Inserted Toxins in Lipid Bilayer Nanodiscs.

Bacterial toxin or viral entry into the cell often requires cell surface binding and endocytosis. The endosomal acidification induces a limited unfolding/refolding and membrane insertion reaction of the soluble toxins or viral proteins into their translocation competent or membrane inserted states. At the molecular level, the specific orientation and immobilization of the pre-transitioned toxin on the cell surface is often an important prerequisite prior to cell entry. We propose that structures of some toxin membrane insertion complexes may be observed through procedures where one rationally

Influence of inhalation anesthetics on ion transport across a planar bilayer lipid membrane.

Ion transport from one aqueous phase (W1) to another (W2) across a planar bilayer lipid membrane (BLM) in the presence of inhalation anesthetics was electrochemically investigated. In the absence of inhalation anesthetics in the BLM system, no ion transport current flowed between W1 and W2 across the BLM. When inhalation anesthetics such as halothane, chloroform, diethyl ether and trichloroethylene were added to the two aqueous phases or the BLM, the ion transport current quite clearly appeared. When the ratio of the concentration of KCl or NaCl in W1 to that in W2 was varied, the zero current

Fabrication and photoelectric properties of self-assembled bilayer lipid membranes on conducting glass.

Supported bilayer lipid membranes (s-BLMs with and without the doping of fullerene C60) self-assembled on indium-tin oxide (ITO) glass were fabricated and characterized by cyclic voltammetry and electrochemical impedance spectroscopy using a three-electrode system. The photoelectric properties of the ITO supported planar lipid bilayers were studied. Light intensity of irradiation, bias voltage, and concentration of donors have been found to be limiting factors of the transmembrane photocurrent. The facilitation effect of C60 doping in s-BLMs on the photoinduced electron transfer across s-BLM i…

Orientation and pore-forming mechanism of a scorpion pore-forming peptide bound to magnetically oriented lipid bilayers.

The orientation and pore-forming mechanisms of pandinin 2 (pin2), an antimicrobial peptide isolated from venom of the African scorpion Pandinus imperator, bound to magnetically oriented lipid bilayers were examined by 31P and 13C solid-state, and 15N liquid-state NMR spectroscopy. 31P NMR measurements at various temperatures, under neutral and acidic conditions, showed that membrane lysis occurred only under acidic conditions, and at temperatures below the liquid crystal-gel phase transition of the lipid bilayers, after incubation for two days in the magnet. Reconstitution of a calcium-activated potassium channel in basolateral membranes of rabbit colonocytes into planar lipid bilayers.

A highly enriched preparation of basolateral membrane vesicles was isolated from rabbit distal colon surface epithelial cells employing the method described by Wiener, Turnheim and van Os (Weiner, H., Turnheim, K., van Os, C.H. (1989) J. Membrane Biol. 110:147-162) and incorporated into planar lipid bilayers. With very few exceptions, the channel activity observed was that of a high conductance. Ca2+-activated K+ channel. This channel is highly selective for K+ over Na+ and Cl-, displays voltage-gating similar to "maxi" K(Ca) channels found in other cell membranes, and kinetic analyses are con…

Effects of gabergic phenols on the dynamic and structure of lipid bilayers: A molecular dynamic simulation approach.

γ-Aminobutyric acid (GABA) is the major inhibitory neurotransmitter in the vertebrate and invertebrate nervous system. GABAA receptors are activated by GABA and their agonists, and modulated by a wide variety of recognized drugs, including barbiturates, anesthetics, and benzodiazepines. The phenols propofol, thymol, chlorothymol, carvacrol and eugenol act as positive allosteric modulators on GABAA-R receptor. These GABAergic phenols interact with the lipid membrane, therefore, their anesthetic activity could be the combined result of their specific activity (with receptor proteins) as well as
