The design of spin probes for electron magnetic resonance spectroscopy and imaging.

The problems that have to be addressed in designing suitable spin probes are manifold. In this article it will be shown that some the properties the probes need to possess are (i) a simple EMR spectrum (in some types of imaging or oximetry a single line is desirable), (ii) sharp lines with no unresolved splittings, to provide maximum sensitivity to detection, (iii) chemical, metabolic and thermal stability, (iv) easy possible synthesis to permit substitution with isotopes, prosthetic groups or groups to control the partition coefficients in lipid/water systems, and (v) electronic anisotropy, i

[1H and 31P-magnetic resonance spectroscopy of cerebral infarction in rats].

Magnetic resonance spectroscopy (MRS) allows the noninvasive study of metabolism in vivo. In order to further understand the time course of biochemical changes during cerebral infarction, we performed the MRS study with pathological analysis. The left middle cerebral artery (MCA) was occluded in spontaneously hypertensive male rats (SHR) by the method of Tamura et al. The spectra were obtained from the infarcted hemisphere by placing the surface coils over the left side of the calvarium. 31P and 1H-MRS were performed at 3 hours, 24 hours and 7 days after MCA occlusion. Ischemic lesions caused …

In vivo proton magnetic resonance spectroscopy in a case of Reye's syndrome.

A case of a 14-year-old boy with Reye's syndrome (RS) and complete neurologic recovery is presented. 1H magnetic resonance spectroscopy was performed on days 1 (admission to ICU), 8 and 62: During the acute phase of RS substantial cerebral metabolic imbalances were observed and their normalization monitored. The spectra from day 1 featured extremely high glutamine content (approximately 18 mmol/kg excess) and low concentrations of choline compounds pounds (approximately 1 mmol/kg deficit). Also some excess lactate was present. The subsequent spectra demonstrated the return to an almost normal …

Diagnosis of meconium in amniotic fluids by nuclear magnetic resonance spectroscopy.

Some 40 samples of amniotic fluids were studied by nuclear magnetic resonance (NMR) spectroscopy as the first step in developing techniques for analysis of amniotic fluids in vivo without amniocentesis. It was found possible to distinguish amniotic fluids containing meconium (fetal feces) from all other kinds of amniotic fluids (hydramnios or normal fluids) by proton NMR, but precise chemical or cytological analysis seemed unavailable. Also reported are results of in vitro analysis of amniotic fluids, and of measurements performed in vivo on other biologic fluids to investigate possible in sit

Bone marrow fat content in 70 adolescent girls with anorexia nervosa: Magnetic resonance imaging and magnetic resonance spectroscopy assessment.

BACKGROUND: Adolescents and women with anorexia nervosa have increased bone marrow fat and decreased bone formation, at least in part due to hormonal changes leading to preferential stem cell differentiation to adipocytes over osteoblasts. OBJECTIVE: The purpose of this study was to evaluate marrow fat content and correlate with age and disease severity using knee MRI with T1 relaxometry (T1-R) and MR spectroscopy (MRS) in 70 adolescents with anorexia nervosa. MATERIALS AND METHODS: We enrolled 70 girls with anorexia nervosa who underwent 3-T knee MRI with coronal T1-W images, T1-R and singl

Nuclear magnetic resonance spectroscopy as a tool to probe molecular condensates.

Biomolecular condensates formed via liquid-liquid phase separation regulate essential cellular processes, with dysregulation implicated in neurodegeneration and cancer. Nuclear magnetic resonance (NMR) spectroscopy provides label-free, atomic-resolution insights into condensate composition, stoichiometry, physicochemical properties (e.g., viscosity and water content), and molecular interactions, overcoming the limitations of imaging and bulk techniques. This review highlights NMR's capabilities in quantifying selective partitioning of proteins, RNAs, ions, and small molecules, as well as mappi

Identifying the translational complexity of magnetic resonance spectroscopy in neonates and infants.

Little attention has been paid to relating MRS outputs of vendor-supplied platforms to those from research software. This comparison is crucial to advance MRS as a clinical prognostic tool for disease or injury, recovery, and outcome. The work presented here investigates the agreement between metabolic ratios reported from vendor-provided and LCModel fitting algorithms using MRS data obtained on Siemens 3 T TIM Trio and 3 T Skyra MRI scanners in a total of 55 premature infants and term neonates with hypoxic ischemic encephalopathy (HIE). We compared peak area ratios in single voxels placed in

Effect of gold marker seeds on magnetic resonance spectroscopy of the prostate.

PURPOSE: Magnetic resonance stereoscopic imaging (MRSI) of the prostate is an emerging technique that may enhance targeting and assessment in radiotherapy. Current practices in radiotherapy invariably involve image guidance. Gold seed fiducial markers are often used to perform daily prostate localization. If MRSI is to be used in targeting prostate cancer and therapy assessment, the impact of gold seeds on MRSI must be investigated. The purpose of this study was to quantify the effects of gold seeds on the quality of MRSI data acquired in phantom experiments. METHODS AND MATERIALS: A cylindri

Quantitative Assessment of Liver Fat with Magnetic Resonance Imaging and Spectroscopy.

Hepatic steatosis is characterized by abnormal and excessive accumulation of lipids within hepatocytes. It is an important feature of diffuse liver disease, and the histological hallmark of non-alcoholic fatty liver disease (NAFLD). Other conditions associated with steatosis include alcoholic liver disease, viral hepatitis, HIV and genetic lipodystrophies, cystic fibrosis liver disease, and hepatotoxicity from various therapeutic agents. Liver biopsy, the current clinical gold standard for assessment of liver fat, is invasive and has sampling errors, and is not optimal for screening, monitorin

In vivo proton magnetic resonance spectroscopy of intraventricular tumours of the brain.

The aim of this study was to assess the usefulness of proton MR spectroscopy in the diagnosis of intraventricular tumours. Fifty-two intraventricular tumours pertaining to 16 different tumour types were derived from our database. All cases had single-voxel proton MR spectroscopy performed at TE at both 30 and 136 ms at 1.5 T. The Mann-Whitney U test was used to search for the most discriminative datapoints each tumour type. Characteristic trends were found for some groups: high Glx and Ala in meningiomas (p < 0.001 and p < 0.01, respectively), high mobile lipids in metastasis (p < 0.001), high

In vivo proton magnetic resonance spectroscopy of breast lesions: an update.

In vivo proton magnetic resonance spectroscopy ((1)H-MRS) has been demonstrated to be successful in the differentiation of benign and malignant breast lesions in a non-invasive manner by detecting increased levels of composite choline (Cho) compounds. Currently there is molecular evidence of increased Cho metabolism in breast cancer cells. In breast malignancies, (1)H-MRS achieved a high-overall sensitivity (82%). Most test cases were infiltrating duct carcinoma, but infiltrating lobular, medullary, mucinous and adenoid cystic carcinomas were also positive by (1)H-MRS. Large lesional size is a…

Magnetic resonance spectroscopy and imaging of the thalamus in idiopathic generalized epilepsy.

Experimental work in animal models of generalized epilepsy and clinical data in humans with idiopathic generalized epilepsy (IGE) indicate that the thalamo-cortical circuitry is involved in the generation of epileptic activity. The purpose of this study was to evaluate in vivo the chemical and structural integrity of the thalamus in patients with IGE. Thalamic proton magnetic resonance spectroscopic imaging (1H-MRSI), measuring N-acetylaspartate (NAA), choline-containing compounds and creatine (Cr) was performed in 20 IGE patients and in a group of age-matched healthy subjects. Additionally, 1

Proton magnetic resonance spectroscopy in a case of subacute sclerosing panencephalitis.

Subacute sclerosing panencephalitis is an encephalopathy caused by a persistent measles virus infection. We examined a 13-year-old girl with subacute sclerosing panencephalitis and performed a magnetic resonance spectroscopic study to evaluate the in vivo pathophysiologic abnormality. The results suggested the occurrence of inflammatory processes and glial proliferation prior to neuronal loss even in magnetic resonance imaging (MRI)-negative regions as well as in MRI-positive regions. The additional resonance peaks were also detected, but further studies should be performed to determine the pr

Aspects of human bioenergetics as studied in vivo by magnetic resonance spectroscopy.

We outline the relevant capabilities of in vivo phosphorus MR spectroscopy by discussing some aspects of normal human biochemistry as studied by this technique. The transport of inorganic phosphate from cytosol into mitochondria in the human skeletal muscle was studied by exploiting a new experimental protocol. We found that Pi was transported into mitochondria in the absence of ATP biosynthesis and in the presence of a pH gradient. The control of CoQ on the efficiency of oxidative phosphorylation in the skeletal muscle and brain was studied by administering CoQ to patients with mitochondrial

Applications of magnetic resonance spectroscopy to diagnosis and monitoring of mitochondrial disease.

Magnetic resonance spectroscopy (MRS) can now be performed on routine high-field clinical magnetic resonance imaging systems. Over the last decade it has provided several useful insights into the pathophysiology of mitochondrial disorders. More recently, the feasibility of applications to clinical diagnosis and monitoring have been demonstrated. Exciting new work suggests that carefully supervised physical conditioning in conjunction with sodium dichloroacetate administration can markedly enhance both biochemical measures of aerobic metabolism and functional performance of patients with mitoch…

Proton magnetic resonance spectroscopy of the anterior cingulate region in schizophrenia.

The authors measured N-acetylaspartate (NAA, a putative neuronal marker), choline and creatine in the anterior cingulate region of 26 schizophrenic patients and 16 control subjects using in vivo proton magnetic resonance spectroscopic imaging (1H MRSI). Relative to the control group, the patients with schizophrenia demonstrated significantly lower NAA in both the right and left anterior cingulate regions. There was no association between NAA and duration of illness or medication dosage. No group differences or lateralized asymmetries in choline or creatine were noted. The NAA findings provide
