Six patients with acute trichinellosis were studied. Three patients were treated with mebendazole and the other three with thiabendazole. Both drugs were given at a daily dose of 50 mg per kg body weight for seven consecutive days. No substantial differences were evident in the efficacy of the two drugs when symptoms, clinical findings, and laboratory data were followed for three months. However, mebendazole was better tolerated than thiabendazole. The diagnosis was based on epidemiological data and positive antibody tests. In two cases, diagnosis was confirmed by isolation of trichinella larv…

Four drug regimens for treating onchocerciasis were compared in a double-blind study in persons from an endemic area in southern Mexico. 40 men were randomly assigned to receive either diethylcarbamazine (DEC) (200 mg/day), mebendazole (2 g/day), levamisole (150 mg per week), or mebendazole plus levamisole. DEC produced the most rapid fall in skin microfilaria counts, but by 6 months the two groups receiving mebendazole showed similar or slightly greater reductions. Despite the administration of corticosteroids to persons receiving DEC, more systemic side-effects were seen in this group. Ocula…

The effect of high dosage therapy with Mebendazole was studied in 5 patients with cystic echinococcosis and 2 patients with alveolar echinococcosis involving several organs. Follow-up studies at regular intervals from 7 to 20 months indicated an improvement of the pathological findings initially observed. This could clearly be demonstrated by the radiographic findings in cases with pulmonary involvement. The desintegration or dissolution of pulmonary cysts was observed within a few months after treatment with Mebendazole had begun. However, consolidation of previously emptied cysts occurred du…

In July 1974, mebendazole was reported to be effective against the larval stage of Echinococcus granulosus in experimentally infected mice. A clinical trial of mebendazole was initiated in December 1974 in 4 patients with far advanced, nonresectable lesions caused by Echinococcus multilocularis. A daily dose of 40 mg of the drug per kg of body weight was administered to the 4 patients for more than 3 years. No evidence of toxicity or adverse reactions has been observed. Detectable plasma mebendazole concentrations were achieved with high-dose mebendazole therapy. Serum concentrations of IgE in…

The anthelmintic drugs mebendazole (Vermox) and levamisole were tested in patients for their possible ovicidal effects on the eggs of Trichuris trichiura and Ascaris lumbricoides. The patients studied were 20 Africans of Sierra Leone, and 11 children of Nassau, Bahamas. Egg counts were made before treatment, and for 5 consecutive days following treatment. Eggs were incubated in 2.5% aqueous potassium dichromate and percentages of larval development were determined. Mebendazole completely inhibited the development of the eggs of T. trichiura by day 3 of the follow-up period. The ovicidal action…

Acute myeloid leukemia (AML) is one of the most common types of acute leukemia in adults with the lowest survival rate of all leukemia. Resistance to cytarabine and anthracycline-based chemotherapy is a major cause of treatment failure. Thus, finding new drugs with anti-leukemia activities and minimal side effect is urgently needed. Here through screening more than 1000 drugs approved by the Food and Drug Administration (FDA) of United States, the anthelmintic drug mebendazole (MBZ) was found to inhibit the growth of AML cell lines (THP-1, U937, NB4 and K562) and bone marrow mononuclear cells …

Long-term chemotherapy of human alveolar echinococcosis with benzimidazole compounds (mebendazole, albendazole) has been shown to be primarily parasitostatic, but its curative (parasitocidal) efficacy is debated. This article reports on a 67-year-old male patient with non-resectable alveolar echinococcosis of the liver who had been continuously treated for 13 years with mebendazole (approximately 45-48 mg/kg body weight per day) and who was closely monitored according to a specific protocol. At the age of 80 years the patient died of oesophageal variceal bleeding. During treatment the hepatic …

Two patients receiving oral high-dose mebendazole therapy for echinococcosis were found to have severe, reversible neutropenia, apparently due to marrow suppression; platelets and RBCs were also reversibly suppressed in one. High blood levels of mebendazole (239 ng/mL) in one patient may have resulted in the neutropenia and several toxic side effects, as well as a striking shrinkage of the patient's pulmonary and liver cysts. Neutropenia with high-dose mebendazole therapy may occur in up to 5% of patients and may be much more common than previously recognized. The WBC count should be monitored…

Authors present three cases of peritoneal hydatidosis, two of which are due to spontaneous rupture of hepatic hydatid cysts and the other due to operative rupture of ovarian hydatid cyst. A dose of 1 g./day of mebendazole was administered for 11 months with 30 day break periods every three months. Mebendazole has proved effective in the treatment of secondary echinococcosis. In authors judgement this may be an effective treatment for peritoneal spreading if mebendazole is used at an early stage since they have verified the absence of dissemination in one of the cases and an inactive disseminat…

Two patients with inoperable multiple Echinococcus granulosus infections involving the lungs and the liver (one patient) were treated with the following daily doses of mebendazole (Vermox): 3 days 1000 mg, 3 days 1500 mg, the following 3 months 30-40 mg/kg body weight. One of the patients had been treated before with mebendazole in similar doses for 3 periods of 5-6 weeks each. The patients received the drug as 500 mg tablets divided into 3 daily doses after meals. After termination of treatment a distinct regression of lung cysts was demonstrable in both patients, with the exception of one cy…

The progressive micromorphological changes in Taenia taeniaeformis cysticerci, induced by a single parenteral treatment of the infected mice with mebendazole, are described. The time-related alterations concerned the tegument and tegumental cells and were successively: disappearance of microtubules, accumulation of secretory substances in the Golgi areas, decrease in number to complete loss of microtriches, "ballooning" of all tegumental cells with subsequent burst, vacuolization and degeneration of the tegument, and finally necrosis of the pseudoproglottids. Similar but less pronounced injuri…

Single 500 mg doses of mebendazole (Vermox; Janssen) were administered to 211 children aged 5 - 16 years in a rural area for treatment of single and mixed infestations with Ascaris lumbricoides, hookworm and Trichuris trichiura; the majority of subjects had low egg counts. Treatment was particularly effective against Ascaris, less effective against hookworm and promising against Trichuris. In most refractory cases egg output was considerably reduced. Treatment resulted in fewer Ascaris (95%) and hookworm (84%) ova reaching the environment. The results compare well with those obtained elsewhere…
