Venom alkaline phosphatase was detected using a blotting method following electrophoresis. The enzyme gave strong reactions in some venoms, but was absent in other venoms, some within the same species. The mol. wt of the enzyme is close to 100,000 and its pI is between 3.6 and 4.8. A method is described of the isolation of exonuclease from cultural medium of Actinomyces sp. of coelicolor type, strain 5. The enzyme preparation is practically free from phosphatase activity (the ratio between exonuclease and phosphatase activities is about 10000). Chromatography on DEAE-Sephadex after the heating of the enzyme preparation at 37 degrees C and pH 5.5 in the presence of 2-mercaptoethanol was used to elimate the phosphatase contamination. The usefulness and sensitivity of a direct agglutination test (DAT) in the diagnosis of cutaneous leishmaniasis infection has been investigated. Trypsin treated, formalin fixed and coomassie blue stained Leishmania promastigotes were used as antigens: L. infantum, L. donovani, L. aethiopica. HCG of chorionic tissues of hydatidiform mole was purified using ammonium sulfate fractionation, ion exchange chromatography and gel filtration. The purified hCG has a molecular weight of 1.5x10(5). BACKGROUND: Polymerized immunoglobulins can be difficult to interpret when observed on serum immunofixation electrophoresis (IFE). The use of reducing agents, such as beta-mercaptoethanol (BME), has been instrumental in resolving cases in which the monoclonal protein is unidentifiable due to polymerization. However, this procedure can be inconvenient to laboratories. We therefore investigated the use of immunotyping (IT) to resolve immunoglobulin aggregates as a potential replacement for BME pretreatment.

Optimal conditions for the use of 2-mercaptoethanol (2-ME) and dithiothreitol (DTT) in the inactivation of IgM red cell antibodies were investigated. 0.2 M 2-ME was better than 0.01 M DTT; higher concentrations of these 2 substances could not conveniently be used because of gel formation. Monoclonal antibodies can be labelled with technetium-99m by prereduction of the antibody with 2-mercaptoethanol, then reduction of pertechnetate with an aliquot of a stannous kit, resulting in greater than 97% labelling without the need for further purification. Silver stainability of nucleolus organizer regions (NORs) appears to be correlated with the presence of grouped sulfhydryl (SH) side chains of proteins. In fact, heavy metals with high affinity for SH groups, such as Hg and Cu, do prevent the silver staining reaction. In this study, the direct agglutination test (DAT), using 2-mercaptoethanol (2-ME), kaolin or N-acetylcysteine (NAC) as sample diluents, was used to assay 89 samples from visceral leishmaniasis (VL) patients and 130 samples from patients with other diseases and healthy individuals. Specific IgM antibodies were detected indirectly in sera of patients of typhoid by observing fourfold or more fall in the agglutinin titres after treatment of the serum with 2-mercaptoethanol (2-ME) for removal of IgM antibodies.
