Unique ability of the Proteus mirabilis capsule to enhance mineral growth in infectious urinary calculi.

Struvite (MgNH4PO4.6H2O) calculi are a common complication of Proteus mirabilis urinary tract infections. Although urease is a major virulence factor in calculus formation, the polysaccharide capsule (CPS) of this organism also enhances struvite crystallization and growth in vitro (L. Clapham, R. J. C. McLean, J. C. Nickel, J. Downey, and J. W. Costerton, J. Crystal Growth 104:475-484, 1990). We obtained purified CPS, of known structure and varying anionic character, from P. mirabilis ATCC 49565 and several other organisms. Artificial urine was added to CPS, and the pH was elevated from 5.8 to…

Investigation of the effects of ampicillin and ceftazidime on Proteus mirabilis using metabolomic approaches and bioinformatics analyses.

Antibiotic resistance is the ability of microorganisms to survive and proliferate despite exposure to antibiotics that would normally inhibit or kill susceptible strains. This resistance can make antibiotics ineffective or diminish their ability to combat microorganisms, complicating the treatment of infections and potentially leading to serious complications. To combat antibiotic resistance, a comprehensive analysis of changes in the metabolic activities of microorganisms is crucial for understanding the underlying mechanisms. Proteus mirabilis is a critical pathogen, particularly as a common…

The effectiveness and risk comparison of EDTA with EGTA in enhancing Cd phytoextraction by Mirabilis jalapa L.

In the previous study, Mirabilis jalapa L. had revealed the basic Cd hyperaccumulator characteristics, but the accumulation ability was not as strong as that of other known Cd hyperaccumulators. In order to improve the accumulation ability of this ornamental plant, the chelants were used to activate the Cd in soil. As a substitute, ethylene glycol bis(2-aminoethyl) tetraacetic acid (EGTA) was selected to testify whether it has better effectiveness and can bring lesser metal leaching risk than EDTA. The data showed that the growth of M. jalapa was inhibited, while the Cd concentration of the pl…

ZapA, a virulence factor in a rat model of Proteus mirabilis-induced acute and chronic prostatitis.

Our knowledge of pathogenesis has benefited from a better understanding of the roles of specific virulence factors in disease. To determine the role of the virulence factor ZapA, a 54-kDa metalloproteinase of Proteus mirabilis, in prostatitis, rats were infected with either wild-type (WT) P. mirabilis or its isogenic ZapA(-) mutant KW360. The WT produced both acute and chronic prostatitis showing the typical histological progressions that are the hallmarks of these diseases. Infection with the ZapA(-) mutant, however, resulted in reduced levels of acute prostatitis, as determined from lower le…

The detection of raised levels of IgM to Proteus mirabilis in sera from patients with rheumatoid arthritis.

An analysis by ELISA of 100 rheumatoid factor (RF)-positive sera selected at random from a collection of sera from patients with various auto-immune diseases and joint pains, and 100 RF-negative sera from the same collection matched by patient age and gender, showed that the RF-positive sera had highly significantly (p < 0.0001) raised levels of IgM antibody, but not IgG antibody, to Proteus mirabilis over those of the RF-negative sera. This response was subsequently found to be associated with sera from patients who clinically had rheumatoid arthritis (RA). Sera from the RA patients had signi…

Evaluation of biofilm activity, virulence genes, and antibiotic susceptibility profiles in clinical Proteus mirabilis isolates.

INTRODUCTION: Proteus mirabilis is a common opportunistic pathogen in wound and urinary tract infections (UTIs), whose clinical significance is driven by biofilm formation. This virulence-mediated process provides a shield against antimicrobials and host immunity, promoting difficult-to-treat infections. This study aimed to elucidate the relationship between biofilm-forming capacity, presence of virulence genes, and antibiotic resistance profiles in P. mirabilis isolates. METHODOLOGY: A total of 76 P. mirabilis isolates from urine and wound samples were analyzed. Isolates were identified usin…

Draft genome sequencing of Proteus mirabilis strain Indica (ST286) reveals antimicrobial resistance and virulence determinants.

BACKGROUND: Proteus mirabilis is a Gram-negative, motile bacterium that typically colonizes the human gastrointestinal tract as a commensal organism. Under favorable conditions, particularly in hospitalized or immunocompromised individuals, as an opportunistic pathogen, causing a range of infections including catheter-associated urinary tract infections, wound infections and bacteremia. Its pathogenic potential is linked to a combination of virulence traits such as urease production, swarming motility, biofilm formation and the ability to acquire antimicrobial resistance determinants, which to…

Biosynthesized gold nanoparticles from Proteus mirabilis: A synergistic strategy against multidrug-resistant Gram-negative bacteria.

Gold nanoparticles (AuNPs) are increasingly explored as antimicrobial agents for the detection and treatment of multidrug-resistant (MDR) bacterial infections. This research tries to establish an eco-friendly, biological process for synthesizing AuNPs using Proteus mirabilis and studies their antibacterial efficacy against MDR Gram-negative pathogens and their potential to enhance standard antibiotic efficacy. Using the Vitek-2 system, 51 P.mirabilis isolates were analyzed, of which 74.5 % were MDR. Netilmicin was effective against 75 % of these isolates. P.mirabilis extracellularly biosynthes…

Metabolic interplay between Proteus mirabilis and Enterococcus faecalis facilitates polymicrobial biofilm formation and invasive disease.

Biofilms play an important role in the development and pathogenesis of catheter-associated urinary tract infection (CAUTI). Proteus mirabilis and Enterococcus faecalis are common CAUTI pathogens that persistently co-colonize the catheterized urinary tract and form biofilms with increased biomass and antibiotic resistance. In this study, we uncover the metabolic interplay that drives biofilm enhancement and examine the contribution to CAUTI severity. Through compositional and proteomic biofilm analyses, we determined that the increase in biofilm biomass stems from an increase in the protein fra…

A small-molecular inhibitor against Proteus mirabilis urease to treat catheter-associated urinary tract infections.

Infection and blockage of indwelling urinary catheters is significant owing to its high incidence rate and severe medical consequences. Bacterial enzymes are employed as targets for small molecular intervention in human bacterial infections. Urease is a metalloenzyme known to play a crucial role in the pathogenesis and virulence of catheter-associated Proteus mirabilis infection. Targeting urease as a therapeutic candidate facilitates the disarming of bacterial virulence without affecting bacterial fitness, thereby limiting the selective pressure placed on the invading population and lowering …

Potentially Probiotic Lactobacillus Strains Derived from Food Intensify Crystallization Caused by Proteus mirabilis in Urine.

Proteus mirabilis is a common cause of infectious urolithiasis. The first stage in the formation of urinary stones is the crystallization of mineral salts in the urine induced by urease activity of this microorganism. Lactobacillus spp. are an important component of the human microbiota and in large quantities occur in foods. Regardless of their origin, those with probiotic properties are proposed as an alternative to antibiotic therapy in the treatment of urinary tract infections. The aim of the study was to check the effect of selected Lactobacillus plantarum and Lactobacillus brevis strains…

Production Optimization, Partial Purification, and Biochemical Characterization of an Extracellular Lipase from Proteus mirabilis SAB2.

Lipases are industrially important biocatalysts with broad applications in biotechnology and environmental processing. In this study, an extracellular lipase-producing bacterial isolate, SAB2, recovered from soil collected in Rijal Almaa, Saudi Arabia, was identified as Proteus mirabilis using phenotypic, biochemical, and 16S rRNA gene sequence evidence. Lipase production was evaluated by a one-factor-at-a-time (OFAT) approach examining temperature, initial pH, carbon and nitrogen sources, inoculum size, and incubation period. The highest temperature-series production response was 2.660 U/mL a…

Genomic divergence and sex-linked region of Welwitschia mirabilis revealed by whole-genome re-sequencing.

A central goal of population genomics is to unveil the evolutionary forces driving genomic divergence. Although genomic islands have been extensively studied in angiosperms, the patterns of genomic divergence remain poorly understood in gymnosperms. Here, by re-sequencing the whole genome of 130 individuals of Welwitschia mirabilis, we reveal significant genomic differentiation among its geographically close populations. Population genomic analyses suggest that geographical isolation and species characteristics play important roles in triggering and maintaining population divergence. We also i…
