Ketogenic Diet in Cancer Prevention and Therapy: Molecular Targets and Therapeutic Opportunities.

Although cancer is still one of the most significant global challenges facing public health, the world still lacks complementary approaches that would significantly enhance the efficacy of standard anticancer therapies. One of the essential strategies during cancer treatment is following a healthy diet program. The ketogenic diet (KD) has recently emerged as a metabolic therapy in cancer treatment, targeting cancer cell metabolism rather than a conventional dietary approach. The ketogenic diet (KD), a high-fat and very-low-carbohydrate with adequate amounts of protein, has shown antitumor effe

PPAR Gamma: From Definition to Molecular Targets and Therapy of Lung Diseases.

Peroxisome proliferator-activated receptors (PPARs) are members of the nuclear receptor superfamily that regulate the expression of genes related to lipid and glucose metabolism and inflammation. There are three members: PPARα, PPARβ or PPARγ. PPARγ have several ligands. The natural agonists are omega 9, curcumin, eicosanoids and others. Among the synthetic ligands, we highlight the thiazolidinediones, clinically used as an antidiabetic. Many of these studies involve natural or synthetic products in different pathologies. The mechanisms that regulate PPARγ involve post-translational modificati

Molecular targeted therapies: Ready for "prime time" in biliary tract cancer.

The prognosis for patients with biliary tract cancers (cholangiocarcinoma and gallbladder cancer) is poor, while the incidence of these cancers is increasing. Most patients are diagnosed with advanced disease when treatment options are limited to palliative approaches, mainly focused on chemotherapy. In recent years, novel treatment targets of relevance to biliary tract cancers, mainly present in patients with intrahepatic cholangiocarcinoma, have been identified and are rapidly changing the field. These include fibroblast growth factor receptor (FGFR) fusions and isocitrate dehydrogenase (IDH

[Epstein-Barr Virus Genome Replication as a Molecular Target for Cancer Therapy].

Epstein-Barr virus (EBV), a human oncogenic virus, is a B cell-tropic herpesvirus and has the ability to immortalize normal B cells during latent infection. The Epstein-Barr nuclear antigen 1 (EBNA1) protein of EBV is expressed in the most EBV latently infected cells and binds to a specific viral genome region termed "oriP" (origin of plasmid replication) to maintain the stability of the approximately 170 kb double-stranded circular virus genomic DNA (episome) in cells. EBV elimination is thought to inhibit progression of EBV-associated malignancies, and the EBNA1-dependent mechanisms for EBV

Molecular and cellular pathways as treatment targets for biologic therapies in systemic sclerosis.

Recent advances have shed light on the complex pathogenic processes that underlie the development and progression of Systemic Sclerosis (SSc) but management of the disease remains problematic and curative treatment is not available. Better understanding of the underlying pathology has enabled novel therapeutic approaches to be investigated, as therapies in rheumatology are becoming increasingly disease/ organ-specific, targeting unique biological networks and signalling pathways. The pathophysiologic and clinical pleiomorphism of SSc however, represents a major barrier to conducting large well

Molecular advancements in targeting fibroblast activation protein (FAP) in cancer therapies.

INTRODUCTION: Fibroblast activation protein (FAP) is a membrane-bound serine protease highly expressed by activated stromal cells in the majority of epithelial-derived malignancies, on the contrary FAP expression is limited in normal adult tissues. Beyond the recognizing role as a marker of cancer-associated fibroblasts (CAFs), accumulating evidence indicates that FAP actively contributes to extracellular matrix remodeling, immune suppression, tumor progression and therapeutic resistance, making it an attractive target for both cancer diagnosis and treatment. Emerging molecular targets in gynaecologic cancers: clinical implications for personalized therapy.

Gynaecologic malignancies, such as ovarian, cervical, endometrial, vulvar, and vaginal cancers, are a significant health burden in the world, characterised by significant heterogeneity in their mode of expression on the molecular scale, high rates of recurrence, and low rates of sustained response to standard treatment. Recent progress in cancer genomics has essentially redefined the nature of these diseases, discovering multifaceted genetic, epigenetic, and tumour microenvironment-based processes that regulate tumour progression, therapeutic response, and resistance. This review summarises th

Targeting rheumatoid arthritis: a molecular perspective on biologic therapies and clinical progress.

To surpass challenges with traditional approaches to treat Rheumatoid arthritis (RA), an improved understanding of molecular-level pathogenesis brought forth targeted therapy with biologics as a great promise in halting the progression of RA. Novel biologics are being designed with the help of synthetic fusion proteins, monoclonal antibodies, and protein fragments, with or without drugs, to target various signaling pathways, including TNF-α, IL-6, JAK, Th-17, IL-family, GM-CSF, B-cell, and T-cell signaling. The journey of biologics in RA management began in 1998 with etanercept (Enbrel®). Sinc

Potent molecular-targeted therapies for gastro-entero-pancreatic neuroendocrine carcinoma.

Neuroendocrine neoplasms (NENs), which are characterized by neuroendocrine differentiation, can arise in various organs. NENs have been divided into well-differentiated neuroendocrine tumors (NETs) and poorly differentiated neuroendocrine carcinomas (NECs) based on morphological differentiation, each of which has a distinct etiology, molecular profile, and clinicopathological features. While the majority of NECs originate in the pulmonary organs, extrapulmonary NECs occur most predominantly in the gastro-entero-pancreatic (GEP) system. Although platinum-based chemotherapy is the main therapeut

Three targets in one complex: A molecular perspective of TFIIH in cancer therapy.

The general transcription factor II H (TFIIH) plays an essential role in transcription and nucleotide excision DNA repair (NER). TFIIH is a complex 10 subunit containing molecular machine that harbors three enzymatic activities while the remaining subunits assume regulatory and/or structural functions. Intriguingly, the three enzymatic activities of the CDK7 kinase, the XPB translocase, and the XPD helicase exert different impacts on the overall activities of TFIIH. While the enzymatic function of the XPD helicase is exclusively required in NER, the CDK7 kinase is deeply involved in transcript

Targeting Molecular Inflammatory Pathways in Granuloma as Host-Directed Therapies for Tuberculosis.

Globally, more than 10 million people developed active tuberculosis (TB), with 1.4 million deaths in 2020. In addition, the emergence of drug-resistant strains in many regions of the world threatens national TB control programs. This requires an understanding of host-pathogen interactions and finding novel treatments including host-directed therapies (HDTs) is of utter importance to tackle the TB epidemic. Mycobacterium tuberculosis (Mtb), the causative agent for TB, mainly infects the lungs causing inflammatory processes leading to immune activation and the development and formation of granul

Chromosomal alterations, prognostic factors, and targeted molecular therapies for malignant meningiomas.

Meningiomas are the second most common intracranial neoplasm in adults and originate from arachnoidal cap cells. Malignant meningiomas are resistant to conventional treatments such as surgery, chemotherapy, and radiotherapy. Unlike benign meningiomas, atypical and anaplastic tumors generally display more complex karyotypes associated with aggressive behavior. While these chromosomal anomalies are associated with greater malignancy in meningiomas, the specific genes involved remain unknown. Malignant meningiomas are characterized by increased tumor aggressiveness, rapid recurrence, local invasi

mTOR in squamous cell carcinoma of the oesophagus: a potential target for molecular therapy?

AIMS: The mammalian target of rapamycin (mTOR), an important regulator of protein translation and cell proliferation, is activated in various malignancies. In a randomised controlled trial of advanced renal cell carcinoma patients, targeted therapy to mTOR by means of rapamycin analogues has been shown to significantly improve survival. An in vitro study has revealed that mTOR is activated in oesophageal squamous cell carcinoma (OSCC) cell lines and that mTOR expression is inhibited by rapamycin. The objectives of this histological study were to determine the proportion of OSCC tissues with ac

Molecular targets for emerging anti-tumor therapies for neurofibromatosis type 1.

Neurofibromatosis type 1 (NF1) is the most common cancer predisposition syndrome. NF1 patients present with a constellation of clinical manifestations and have an increased risk of developing certain benign and malignant tumors. This disease results from mutation within the gene encoding neurofibromin, a GTPase activating protein (GAP) for Ras. Functional loss of this protein compromises Ras inactivation, which leads to the aberrant growth and proliferation of neural crest-derived cells and, ultimately, tumor formation. Current management of NF1-associated malignancy involves radiation, surgic
