Roles of Extracellular Polysaccharides and Biofilm Formation in Heavy Metal Resistance of Rhizobia.

Bacterial surface components and extracellular compounds, particularly flagella, lipopolysaccharides (LPSs), and exopolysaccharides (EPSs), in combination with environmental signals and quorum-sensing signals, play crucial roles in bacterial autoaggregation, biofilm development, survival, and host colonization. The nitrogen-fixing species Sinorhizobium meliloti (S. meliloti) produces two symbiosis-promoting EPSs: succinoglycan (or EPS I) and galactoglucan (or EPS II). Studies of the S.meliloti/alfalfa symbiosis model system have revealed numerous biological functions of EPSs, including host sp

Which steps are essential for the formation of functional legume nodules?

Nodulation is reviewed in terms of the phenotypes proposed by Vincent (1980). Individual legumes may be infectible by one or more of the three bacterial genera (collectively known as rhizobia) Rhizobium, Bradyrhizobium, or Azorhizobium. The type of infection process by which rhizobia gain entry is largely governed by the host genotype. In addition to the widely studied root-hair pathway, infections may be associated with lateral root emergence or occur between root epidermal cells. The exact chemical and physical nature of the root hair/epidermal cell wall is likely to be a critical factor in

Functional Nitrogenase Cofactor Maturase NifB in Mitochondria and Chloroplasts of Nicotiana benthamiana.

Engineering plants to synthesize nitrogenase and assimilate atmospheric N2 will reduce crop dependency on industrial N fertilizers. This technology can be achieved by expressing prokaryotic nitrogen fixation gene products for the assembly of a functional nitrogenase in plants. NifB is a critical nitrogenase component since it catalyzes the first committed step in the biosynthesis of all types of nitrogenase active-site cofactors. Here, we used a library of 30 distinct nifB sequences originating from different phyla and ecological niches to restore diazotrophic growth of an Azotobacter vineland

Control of SBR switching by fuzzy pattern recognition.

The sequencing batch reactor (SBR) is a widely used process for biological removal of nutrients (nitrogen and phosphorus) from wastewater. It is based on the metabolism of specialised bacteria, which under alternate anaerobic/aerobic conditions uptake phosphorus and perform denitrification. Intermittent operation is normally operated on a fixed switching schedule with ample margin for possible inaccuracies, with the result that the process operation is highly inefficient. This paper proposes a switching strategy based on the indirect observation of process state through simple physico-chemical

Recent advances and fundamentals of microalgae cultivation technology.

Microalgae are considered to be a promising group of organisms for fuel production, waste processing, pharmaceutical applications, and as a source of food components. Unicellular algae are worth being considered because of their capacity to produce comparatively large amounts of lipids, proteins, and vitamins while requiring little room for growth. They can also grow on waste and fix CO2 and nitrogen compounds. However, production costs limit the industrial use of microalgae to the most profitable applications including micronutrient production and fish farming. Therefore, novel microalgae bas

Comparing methanol and glycerol as carbon sources for mainstream partial denitrification/anammox in an IFAS process.

This study explored the implementation of mainstream partial denitrification with anammox (PdNA) in the second anoxic zone of a wastewater treatment process in an integrated fixed film activated sludge (IFAS) configuration. A pilot study was conducted to compare the use of methanol and glycerol as external carbon sources for an IFAS PdNA startup, with a goal to optimize nitrogen removal while minimizing carbon usage. The study also investigated the establishment of anammox bacteria on virgin carriers in IFAS reactors without the use of seeding, and it is the first IFAS PdNA startup to use meth

Electron transfer to nitrogenase in Paenibacillus polymyxa.

UNLABELLED: Nitrogenase requires low-potential electrons to reduce N2 to produce ammonia. The direct electron donors to nitrogenase are reduced ferredoxin (Fd) and flavodoxin (Fld). The enzymes that reduce Fd or Fld for N2 fixation mainly include pyruvate-flavodoxin (ferredoxin) oxidoreductase (PFOR), ferredoxin-NADP+ reductase (FNR), Rhodobacter nitrogen fixation protein complex (Rnf), and bifurcating electron transfer flavoprotein complex (Fix). Paenibacillus polymyxa is a model for studies of nitrogen fixation, but the pathways of electron transport to nitrogenase are unknown. In this study

Plant-Compatible Xenium In Situ Sequencing: Protocol for Spatial Transcriptomics in Medicago truncatula Roots and Nodules.

Elucidating the spatial and temporal regulation of gene expression during plant organogenesis is crucial for enabling precise crop improvement strategies that incorporate beneficial traits into crops while avoiding adverse effects. Root nodules, specialized organs formed in symbiosis with nitrogen-fixing bacteria, provide a valuable system to study cell-type-specific gene networks in a symbiosis-induced developmental context. However, capturing these dynamics at cellular resolution in intact plant tissues remains technically challenging. Spatial transcriptomics technologies developed for anima…

Genomic and Experimental Analysis of the Biostimulant and Antagonistic Properties of Phytopathogens of Bacillus safensis and Bacillus siamensis.

The B. safensis RGM 2450 and B. siamensis RGM 2529 strains were isolated from the rhizosphere of plants presenting resilience to abiotic and biotic stress conditions. To understand the implications of bacteria in resilience, a genomic and experimental analysis was carried out on their biostimulant and phytopathogenic antagonist properties. Genome analyses of both strains indicated that they have the potential to synthesize bioactive compounds such as the battery of non-ribosomal peptides, polyketides, extracellular enzymes and phytohormones. These results were consistent with the antagonistic

Plant Growth-Promoting Traits of a Thermophilic Strain of the Klebsiella Group with its Effect on Rice Plant Growth.

In agriculture, instead of synthetic fertilizers, natural bio-inoculants can be used to increase growth and yield of crops. For this purpose, we report a thermophilic bacteria Klebsiella sp. strain PMnew, isolated from Paniphala hot spring. The strain was characterized and assessed for plant growth-promoting traits. Oryza sativa L. var Swarna (rice) seeds were inoculated with the strain to study the bacterization effect on vegetative and reproductive growth of rice plants. The results indicate that PMnew produces organic acids to solubilize phosphate (550.16 ± 0.04 µg/ml), fixes nitrogen, prod

Ensifer, Phyllobacterium and Rhizobium species occupy nodules of Medicago sativa (alfalfa) and Melilotus alba (sweet clover) grown at a Canadian site without a history of cultivation.

Phage-resistant and -susceptible bacteria from nodules of alfalfa and sweet clover, grown at a site without a known history of cultivation, were identified as diverse genotypes of Ensifer, Rhizobium and Phyllobacterium species based on sequence analysis of ribosomal (16S and 23S rRNA) and protein-encoding (atpD and recA) genes, Southern hybridization/RFLP and a range of phenotypic characteristics. Among phage-resistant bacteria, one genotype of Rhizobium sp. predominated on alfalfa (frequency approximately 68 %) but was recovered infrequently ( approximately 1 %) from sweet clover. A second ge

Characterization and description of plant-growth-promoting methanotrophic bacteria belonging to the genus of Methylocaldum.

Arid soil microbiomes present untapped resources of microbial diversity. Here, we describe twelve isolates, all belonging to the Methylocaldum genus. Based on metagenomic studies, the isolates represent the major clades of methanotrophic bacteria inhabiting the arid biomes of Southern California, comprising up to 0.03 % of the total soil microbiota. Phenotyping of isolates indicates that they are obligate methanotrophic bacteria, some capable of methanol utilization. All strains can fix nitrogen, use nitrate and ammonia as a N-source, and have key genetic signatures of autotrophy, methylotroph

Bio-capture of Cr(VI) in a denitrification system: Electron competition, long-term performance, and microbial community evolution.

Chromium is widely applied in industries as an important metal resource, but the discharge of Cr(VI) containing wastewater leads to the loss of chromium resources. This study proposed a bio-capture process of chromium in a denitrification system. The bio-capture potentiality was explored by investigating the electron competition between Cr(VI) and nitrogen compounds reduction, the long-term bio-capture performance, and the microbial community evolution. In the competition utilization of electron donors, both NO3--N and NO2--N took precedence over Cr(VI), and NO2--N reduction was proved to be t

In vitro antagonistic activity, plant growth promoting traits and phylogenetic affiliation of rhizobacteria associated with wild plants grown in arid soil.

The role of plant growth-promoting rhizobacteria (PGPR) in adaptation of plants in extreme environments is not yet completely understood. For this study native bacteria were isolated from rhizospeheric arid soils and evaluated for both growth-promoting abilities and antagonistic potential against phytopathogenic fungi and nematodes. The phylogentic affiliation of these representative isolates was also characterized. Rhizobacteria associated with 11 wild plant species from the arid soil of Almadinah Almunawarah, Kingdom of Saudi Arabia (KSA) were investigated. From a total of 531 isolates, only

Impact of Plant Growth Promoting Rhizobacteria Exiguobacterium Acetylicum RGK and Enterobacter Mori RGK1 on Growth Parameters and Phytochemicals of Asparagus Racemosus.

Rhizobacteria that promote plant growth (PGPR) are a wide range of soil bacteria that inhabit the rhizosphere of plants. In the present study, two plant growth-promoting bacterial strains were isolated from the rhizospheric soil of the Asparagus plant, Namely Enterobacter mori RGK1 and Exiguobacterium acetylicum RGK, which can enhance plant growth. It was shown that both of these bacterial strains were able to synthesize exopolysaccharide, hydrogen cyanide (HCN), indole acetic acid and siderophores. These strains fix atmospheric nitrogen, solubilize phosphate, zinc and potassium. Additionally,

Heterotrophy in tropical scleractinian corals.

The dual character of corals, that they are both auto- and heterotrophs, was recognized early in the twentieth Century. It is generally accepted that the symbiotic association between corals and their endosymbiotic algae (called zooxanthellae) is fundamental to the development of coral reefs in oligotrophic tropical oceans because zooxanthellae transfer the major part of their photosynthates to the coral host (autotrophic nutrition). However, numerous studies have confirmed that many species of corals are also active heterotrophs, ingesting organisms ranging from bacteria to mesozooplankton. H

Soil Microbial Resources for Improving Fertilizers Efficiency in an Integrated Plant Nutrient Management System.

Tomorrow's agriculture, challenged by increasing global demand for food, scarcity of arable lands, and resources alongside multiple environment pressures, needs to be managed smartly through sustainable and eco-efficient approaches. Modern agriculture has to be more productive, sustainable, and environmentally friendly. While macronutrients such as nitrogen (N), phosphorus (P), potassium (K), and sulfur (S) supplied by mineral fertilizers are vital to crop production, agriculturally beneficial microorganisms may also contribute directly (i.e., biological N2 fixation, P solubilization, and phyt

Cellular Stoichiometry of Methyl-Accepting Chemotaxis Proteins in Sinorhizobium meliloti.

The chemosensory system in Sinorhizobium meliloti has several important deviations from the widely studied enterobacterial paradigm. To better understand the differences between the two systems and how they are optimally tuned, we determined the cellular stoichiometry of the methyl-accepting chemotaxis proteins (MCPs) and the histidine kinase CheA in S. meliloti Quantitative immunoblotting was used to determine the total amount of MCPs and CheA per cell in S. meliloti The MCPs are present in the cell in high abundance (McpV), low abundance (IcpA, McpU, McpX, and McpW), and very low abundance (
