In eukaryotes, the nucleocytoplasmic export of bulk poly(A)+-mRNAs through the nuclear pore complex is mediated by the ubiquitously expressed NXT1-NXF1 heterodimer. In humans, NXT1 has an X-chromosomal paralog, NXT2, which exhibits testis-enriched expression, suggesting a role in spermatogenesis. Here, we report the in vivo interaction of NXT2 with crucial components of the nuclear export machinery, including NXF1, the testis-specific NXF1 paralogs NXF2 and NXF3, and nuclear pore complex proteins. Binding to NXF2 and NXF3 is mediated by the NTF2-like domain of NXT2. By identifying infertile me…

Reduction in POM121 appears to initiate a decrease in the expression of seven additional nucleoporins, ultimately affecting the localization of Ran GTPase and subsequent cellular toxicity in …

The nuclear pore complex (NPC) is a multi-protein complex that regulates the trafficking of macromolecules between the nucleus and cytoplasm. Genetic variants in components of the NPC have been shown to cause a range of neurological disorders, including intellectual disability and microcephaly. Translocated promoter region, nuclear basket protein (TPR) is a critical scaffolding element of the nuclear facing interior of the NPC. Here, we present two siblings with biallelic variants in TPR who present with a phenotype of microcephaly, ataxia and severe intellectual disability. The variants resul…

Long-standing evidence indicates that human immunodeficiency virus type 1 (HIV-1) preferentially integrates into a subset of transcriptionally active genes of the host cell genome. However, the reason why the virus selects only certain genes among all transcriptionally active regions in a target cell remains largely unknown. Here we show that HIV-1 integration occurs in the outer shell of the nucleus in close correspondence with the nuclear pore. This region contains a series of cellular genes, which are preferentially targeted by the virus, and characterized by the presence of active transcri…

Nanoporous materials synthesized in a wet-chemical process often show different pore sizes in their as-prepared state in the reaction solution and their dried state. Depending on the materials, these changes can be reversible or irreversible. Determination of the exact pore sizes of such materials in their as-prepared state is challenging as conventional characterization methods for pore size determination only work on dry materials. Cryoporometry is applicable directly to wet materials though. Thus, it offers a unique opportunity for analyzing unaffected pores of materials prior to drying. Wi

Nucleoporin 98 (NUP98) fusion oncoproteins are observed in a spectrum of hematologic malignancies, particularly pediatric leukemias with poor patient outcomes. Although wild-type full-length NUP98 is a member of the nuclear pore complex, the chromosomal translocations leading to NUP98 gene fusions involve the intrinsically disordered and N-terminal region of NUP98 with over 30 partner genes. Fusion partners include several genes bearing homeodomains or having known roles in transcriptional or epigenetic regulation. Based on data in both experimental models and patient samples, NUP98 fusion onc…

In response to the challenges of complex pore structures and low recovery rates in shale oil reservoirs of an offshore oilfield, this study investigates the evaluation technology for the lower limit of pore mobilization based on multiscale information fusion. Dynamic imbibition physical simulation experiments were conducted on core samples, and nuclear magnetic resonance (NMR) technology was employed to obtain T2 spectra at various imbibition stages, enabling the analysis of fluid distribution within the pores. Pore throat structures were characterized using high-pressure mercury intrusion (HP

Components of the nuclear periphery coordinate a multitude of activities, including macromolecular transport, cell-cycle progression, and chromatin organization. Nuclear pore complexes (NPCs) mediate nucleocytoplasmic transport, mRNA processing, and transcriptional regulation, and NPC components can define regions of high transcriptional activity in some organisms at the nuclear periphery and nucleoplasm. Lineage-specific features underpin several core nuclear functions and in trypanosomatids, which branched very early from other eukaryotes, unique protein components constitute the lamina, kin…

Infantile striatonigral degeneration is caused by a homozygous variant of the nuclear-pore complex (NPC) gene NUP62, involved in nucleo-cytoplasmic trafficking. By querying sequencing-datasets of patients with dystonia and/or Leigh(-like) syndromes, we identified 3 unrelated individuals with biallelic variants in NUP54. All variants clustered in the C-terminal protein region that interacts with NUP62. Associated phenotypes were similar to those of NUP62-related disease, including early-onset dystonia with dysphagia, choreoathetosis, and T2-hyperintense lesions in striatum. In silico and protei

Human genomics is identifying candidate genes for congenital heart disease (CHD), but discovering the underlying mechanisms remains challenging. In a patient with CHD and heterotaxy (Htx), a disorder of left-right patterning, we previously identified a duplication in Nup188. However, a mechanism to explain how a component of the nuclear pore complex (NPC) could cause Htx/CHD was undefined. Here, we show that knockdown of Nup188 or its binding partner Nup93 leads to a loss of cilia during embryonic development while leaving NPC function largely intact. Many data, including the localization of e…

In vitro airway epithelial models, such as air-liquid interface (ALI) cultures, are widely used for disease modeling, therapeutic and toxicology testing, and studying epithelial-environmental interactions. However, it remains unknown whether the pore density of the permeable membrane inserts, on which primary human bronchial epithelial cells (HBEC) are grown, influences their morphological and differentiation characteristics. To investigate this, HBEC from seven donors were cultured on inserts with low (4 × 106 pores/cm2) and high (1 × 108 pores/cm2) pore densities and differentiated for 28 da

An electron microscope study of the nuclear membrane of Amoeba proteus by thin sectioning techniques has revealed an ultrastructure in the outer layer of the membrane that is homologous to the pores and annuli observed in the nuclear membranes of many other cell types studied by these techniques. An inner honeycombed layer apparently unique to Amoeba proteus is also described.

The technology for geological storage of carbon dioxide (CO2) involves a complex gas-water two-phase seepage process. It is essential to clarify the two-phase seepage law in porous media to increase the efficiency of the CO2 storage. In this paper, based on the classical capillary bundle seepage model, parameters such as roughness, water film thickness, and pore throat ratio are introduced to modify the model, and the physical model of a rough curved capillary bundle considering pore throat structure is constructed. According to the concept of critical displacement aperture, the distribution s

In this issue of Neuron, Gasset-Rosa et al. (2017) and Grima et al. (2017) describe defects in the nuclear pore complex and impaired nucleocytoplasmic transport in Huntington's disease (HD). The findings suggest that erosion of nuclear gatekeeping function, which is found in normal brain aging, may play an important role in the pathogenesis of multiple neurodegenerative disorders, including HD.
