The rising cost of orthodox medication has endeared so many to the use of herbs for the management of neurological conditions. Rauwolfia vomitoria (RV) one of such herbs is a rainforest shrub whose parts are used locally in the management of psychiatry and other medical issues. Its usefulness though not in doubt is wrapped with adverse reports as its active constituents depletes brain monoamine and dopamine stores. This motivated this research on the effects of the root bark extract on olfaction and the olfactory bulb of adult Wistar rats. Eighteen adult Wistar rats (220g average) were divided…

Secretagogin (SCGN) is a recently discovered calcium binding protein of the EF hand family. We studied the structural features of SCGN-positive neurons in the mouse main olfactory bulb (MOB). SCGN-positive neurons were localized throughout layers but clustered in the glomerular layer (GL), mitral cell layer (MCL) and granule cell layer (GCL). They were heterogeneous, including numerous juxtaglomerular neurons, granule cells, small to medium-sized neurons in the external plexiform layer (EPL), and a few small cells in the ependymal/subependymal layer. Calretinin and/or tyrosine hydroxylase occa…

Synaptogenesis is essential for the development of neuronal networks in the brain. In the olfactory bulb (OB) glomeruli, numerous synapses must form between sensory olfactory neurons and the dendrites of mitral/tufted and periglomerular cells. Glomeruli develop from E13 to E16 in the mouse, coincident with an increment of the neuropil in the border between the external plexiform (EPL) and olfactory nerve layers (ONL), coupled to an extensive labelling of phalloidin and GAP-43 from the ONL to EPL. We have tracked synaptogenesis in the OB during this period by electron microscopy (EM) and immuno…

We evaluated the distribution of apolipoprotein E (apoE) immunoreactivity in mouse and human olfactory bulbs. ApoE immunoreactivity was present in the olfactory nerve and around the glomeruli. Immunoreactivity was seen in somata that appeared to be glial. No neuronal staining was seen. The apoE immunoreactivity was also present in the mouse olfactory bulb subependymal layer. The specificity of immunoreactivity was confirmed with apoE-deficient mice (apoE gene knock-out mice, apoE KO) which did not display any immunoreactivity. The presence of apoE around glomeruli and nerve suggest that apoE i…

Men typically display larger olfactory bulb (OB) volumes, while women demonstrate superior olfactory performance. Olfactory training (OT) has been shown to induce structural changes in OB, but whether sex influences OT-related plasticity remains unclear. This study aimed to examine how sex differences modulate OB volume-function relationships and OT-related OB neuroplasticity. Participants included 27 women and 11 men with acquired olfactory dysfunction who underwent Sniffin' Sticks Test and T2-weighted MRI before and after a 12-week OT program of twice-daily exposure to four odorants. OB volu…

One of the most common reasons for partial nasal obstruction is nasal septal deviation (NSD). The effect of a partial lateralized nasal obstruction on olfactory bulb (OB) volume remains unclear. Thus, the aim of this study was to investigate the side differences in olfactory function and OB in patients with serious NSD. Sixty-five volunteers were included: 22 patients with serious right NSD and 43 patients with left NSD. The patients' mean age was 22 years. All participants received volumetric magnetic resonance imaging scans of the entire brain and detailed lateralized olfactory tests. The ma…

Sall1 is a zinc finger containing transcription factor that is highly expressed during mammalian embryogenesis. In humans, the developmental disorder Townes Brocks Syndrome is associated with mutations in the SALL1 gene. Sall1-deficient animals die at birth due to kidney deficits; however, its function in the nervous system has not been characterized. We examined the role of Sall1 in the developing olfactory system. We demonstrate that Sall1 is expressed by cells in the olfactory epithelium and olfactory bulb (OB). Sall1-deficient OBs are reduced in size and exhibit alterations in neurogenesis…

We analyzed the cellular composition of the juxtaglomerular region in the main olfactory bulb of C57B/6J strain mice, focusing on 1) the compartmental organization of the glomerulus and the presence of type 1 and 2 periglomerular cells, 2) the colocalization relationships among the 4 major chemically identified groups of periglomerular cells, glutamic acid decarboxylase (GAD)/gamma-aminobutyric acid (GABA), tyrosine hydroxylase, calretinin and calbindin D28k positive periglomerular cells, and 3) the chemical properties of the nitric oxide synthase (NOS)-positive juxtaglomerular cells. We confi…

The olfactory bulb (OB) serves as the first relay station that processes odor information. Within the OB, odor representations are dynamically modulated by reciprocal synapses between excitatory mitral and tufted (M/T) cells and local inhibitory interneurons. Among these interneurons, periglomerular cells (PGCs) are GABAergic neurons located in the glomerular layer that provide inhibitory inputs to M/T cells. Although their anatomical and molecular properties have been extensively characterized, the in vivo dynamics of PGCs during odor processing and learning remain poorly understood. To addre…

The olfactory bulb (OB), the first central relay of the olfactory pathway, plays a critical role in odor perception and exhibits remarkable structural plasticity shaped by environmental influences. This raises a fundamental question about the extent to which OB volume and its integration within the wider olfactory network is genetically determined. While both OB volume and function have been implicated in a range of neurodegenerative and neuropsychiatric disorders, such as Parkinson's disease, schizophrenia, and depression, all of which involve known genetic risk factors, the heritability of t…

Hyposmia is one of the most prevalent non-motor symptoms of Parkinson's disease and antecedes motor dysfunction by up to a decade. However, the underlying pathophysiology remains poorly understood. In this study, we investigated the mechanisms of dopamine metabolism in post-mortem olfactory bulbs from ten Parkinson's disease and ten neurologic control subjects. In contrast to the loss of dopaminergic neurons in the midbrain, we observed an increase in tyrosine hydroxylase-positive neurons in the Parkinson's disease olfactory bulb, suggesting a potential role for dopamine in the hyposmia associ…

Dopaminergic neurons are located in several brain areas including the olfactory bulb (OB) and involved in many physiological and pathophysiological processes. In the OB, dopamine (DA) is released exclusively by a population of interneurons termed short axon cells (SACs) in the glomerular layer, the initial synaptic integration site of the whole olfactory system. SACs corelease GABA and extend their processes to many glomeruli forming the interglomerular circuit. Two major groups of DA receptors D1-like (D1LRs) and D2-like (D2LRs) types are differentially distributed in the OB, i.e., D1LRs are …

Olfaction is a highly sophisticated sensory modality able to detect and discriminate thousands of different odours, even at very low concentration. How such a challenging task is achieved remains to be fully understood. A unique feature of the olfactory system is the dual role of the odorant receptor: it does detect odours in the olfactory epithelium but it also contributes to neuronal circuit formation in the olfactory bulb. The odorant receptors are indeed expressed on the cilia that protrude in the nasal cavity, where they bind odorants, and at the axon termini, where they could act as axon…

The raphe nuclei provide serotonergic innervation widely in the brain, thought to mediate a variety of neuromodulatory effects. The mammalian olfactory bulb (OB) is a prominent recipient of serotonergic fibers, particularly in the glomerular layer (GL), where they are thought to gate incoming signals from the olfactory nerve. The dorsal raphe nucleus (DRN) and the median raphe nucleus (MRN) are known to densely innervate the OB. The majority of such projections are thought to terminate in the GL, but this has not been explicitly tested. We sought to investigate this using recombinant adeno-ass…

Different types of oscillations in the olfactory bulb (OB), including θ (1 to 4 and 5 to 12 Hz), β (13 to 30 Hz), and γ oscillations (31 to 64 and 65 to 90 Hz), are important in olfactory information processing and olfactory-related functions and have been investigated extensively in recent decades. The awake and anesthetized states, 2 different brain conditions, are used widely in electrophysiologic studies of OB. Chloral hydrate, pentobarbital, and urethane are commonly used anesthetics in these studies. However, the influence of these anesthetics on the oscillations has not been reported. I…

Sall3 is a member of a gene family with homology to the spalt gene of Drosophila melanogaster, encoding transcription factors, and acts as downstream target of hedgehog. Vertebrate homologues of spalt have been shown to be involved in development of the limbs and nervous system and several organs including the kidney and heart; mutations in the genes are implicated in several human genetic disorders. Recent studies have shown a total loss of olfactory bulb (OB) dopaminergic (DA) neurons in Sall3-null mice. We assume that tyrosine hydroxylase (TH) may be regulated by Sall3 in OB. In this study,…

Damage to the olfactory bulb and tract is a frequently described complication of brain surgery in the frontal region, and it seems to be influenced by the surgical approaches. Eighty cerebral hemispheres and 5 formalin-fixed cadavers filled with colored latex were used. Parameters were directly measured, and after olfactory bulb and tract were mobilized with careful dissections, retraction of the frontal lobe was noted. The anterior border of the olfactory bulb is 22.21 (SD, 5.45) mm posterior to the frontomarginal sulcus, and arachnoidal dissection should be performed parallel to olfactory st…

INTRODUCTION: In vivo, most neurons in the main olfactory bulb exhibit robust spontaneous activity. This paper tests the hypothesis that spontaneous activity in olfactory receptor neurons drives much of the spontaneous activity in mitral and tufted cells via excitatory synapses.
METHODS: Single units were recorded in vivo from the main olfactory bulb of a rat before, during, and after application of lidocaine to the olfactory nerve. The effect of lidocaine on the conduction of action potentials from the olfactory epithelium to the olfactory bulb was assessed by electrically stimulating the ol…
