Heterozygous mutation of Drosophila Opa1 causes the development of multiple organ abnormalities in an age-dependent and organ-specific manner.

Optic Atrophy 1 (OPA1) is a ubiquitously expressed dynamin-like GTPase in the inner mitochondrial membrane. It plays important roles in mitochondrial fusion, apoptosis, reactive oxygen species (ROS) and ATP production. Mutations of OPA1 result in autosomal dominant optic atrophy (DOA). The molecular mechanisms by which link OPA1 mutations and DOA are not fully understood. Recently, we created a Drosophila model to study the pathogenesis of optic atrophy. Heterozygous mutation of Drosophila OPA1 (dOpa1) by P-element insertion results in no obvious morphological abnormalities, whereas homozygous

Undesired meeting of lymphocytes: organ-specific infiltration and the organization of ectopic lymphoid tissue in a murine experimental autoimmune gastritis.

Autoimmune diseases are caused by the misdirected attack of the immune system against the body's own components. Delicate balance between the activities of several CD4+ T cell subsets is crucial for determining the initiation of autoimmunity and disease state. In addition, complicated interactions between immune effector cells and tissue resident cells take place in the target organ and construct the inflammatory microenvironment. During the acute/deterioration phase, autoreactive and cytotoxic effector cells leave blood vessels to infiltrate the target site through a multi-step mechanism invo

Paxlovid shows organ-specific and age-specific impacts on risk of developing post-acute sequelae of COVID-19.

BACKGROUND: The impact of antiviral therapies, including Paxlovid, on post-acute sequelae of COVID-19 (PASC) remains inconclusive. METHODS: We analyzed data from 19,413 patients (age > 18) from a validated PASC research cohort in New England who experienced at least one COVID-19 infection episode between January 1, 2022, and June 7, 2022, totaling 22,094 episodes. Multivariable logistic regression with inverse probability weights was used to infer the causal effects of Paxlovid treatment during acute infection and the risk of PASC overall (primary outcome), stratified by age group and organ s

Cryptococcus neoformans-Specific and Non-Cryptococcus neoformans-Specific Antibody Profiles in Organ Transplant Recipients With and Without Cryptococcosis.

Antibody immunity has not been studied in organ transplant recipients (OTRs) with cryptococcosis. We determined serum antibody levels in OTRs: 23 cryptococcosis cases and 21 controls. Glucuronoxylomannan immunoglobulin M (IgM) and laminarin IgM were lower in cases than controls, were inversely associated with cryptococcosis status, and may hold promise as markers of cryptococcosis.

Cell-specific organization and paralog divergence of erythropoietin signaling in Atlantic salmon.

Erythropoietin (EPO) is the main hormonal regulator of erythropoiesis in vertebrates and also mediates non-hematopoietic functions. Despite extensive characterization in mammals, the cellular sources and targets of EPO signaling remain poorly defined in non-mammalian vertebrates. The Atlantic salmon provides a valuable model to investigate paralog-specific organization of EPO signaling due to a recent whole genome duplication (WGD). Here, we characterize the tissue- and cell-specific expression of paralogs encoding EPO and the erythropoietin receptor (EPOR) in Atlantic salmon (Salmo salar). Ph

Learning Gains in Organic Chemistry from Full and Hybrid Specifications Grading.

In recent years, alternative grading strategies to traditional points-based systems have gained popularity, particularly in STEM. Specifications grading is one type of alternative grading that focuses students on meeting course learning objectives rather than accumulating points, sets a threshold for satisfactory work, and often includes frequent, low-stakes assessments, feedback, and opportunities for reattempts or revision. However, the contribution of these features to student learning in authentic courses remains difficult to isolate. This study provides course-scale evidence that frequent

Heterogeneous Spheroid Engineering: Spatial Organization, Fabrication, and Tissue-Specific Design.

Three-dimensional (3D) spheroid models have emerged as advanced in vitro systems that overcome the limitations of conventional two-dimensional (2D) culture by better recapitulating native tissue architecture, cell-cell and cell-extracellular matrix interactions, and physiologically relevant gradients of oxygen, nutrients, and signaling factors, making them valuable platforms for tissue engineering and regenerative medicine. Realizing their full potential, however, requires rational control over cellular composition and spatial organization. In this review, we classify spheroids by cellular com

Organ-Specific Responses to Plant Secondary Metabolites Reveal Mechanisms of Adaptation.

Many insects feed on plants that produce toxic secondary metabolites. The Rattlebox plant (Crotalaria) produces Pyrrolizidine Alkaloids (PAs) that protect it from most herbivores, but some insects, including the Bella moth (Utetheisa ornatrix), can metabolize, sequester, and repurpose its chemical defenses. However, organ-specific responses to Crotalaria secondary metabolites remain poorly understood, and current knowledge is largely derived from a few model species and isolated organ studies. This study bridges this gap by evaluating organ-specific responses of the Bella moth to the Showy Rat

Sex-specific differences in clinical characteristics and organ damage in Hypertensive Urgencies.

Acute blood pressure disorders are a common yet heterogeneous clinical challenge in the Emergency Department (ED), often requiring rapid assessment and risk stratification. Among these, Hypertensive Urgency (HU) is a frequent presentation, characterized by severely elevated blood pressure (BP) without acute hypertension-mediated organ damage (HMOD). Despite its clinical relevance, sex-related differences in HU remain poorly understood. . This study aimed to investigate sex differences in clinical presentation, BP control and HMOD prevalence in subjects presenting with HU. Methods. A multicentr

Organ-Specific Histopathological Effects of Prenatal Alcohol Exposure: A Narrative Review.

The role of alcohol-induced epigenetic modifications that predispose the fetus to metabolic dysregulation, increased susceptibility to future substance use, and long-term behavioral and cognitive impairments has received increasing attention. Fetal alcohol syndrome (FAS) and fetal alcohol spectrum disorders (FASDs) remain underdiagnosed worldwide. As evidence continues to demonstrate that no safe level of alcohol consumption exists during pregnancy, strengthening public awareness, maternal education, and prenatal care services has become increasingly imperative. Prenatal alcohol exposure (PAE)

Assessment of Organ-Specific Fibrotic Biomarkers in Patients With Oral Submucous Fibrosis.

BACKGROUND: Oral submucous fibrosis is a potentially malignant disorder with a high malignant transformation rate. Areca nut, being the chief etiologic factor, when chewed, is known to be swallowed and absorbed into circulation resulting in several systemic effects. This is the first kind of report presenting serum organ-specific fibrosis biomarkers suggestive of functional and fibrotic changes in the visceral organs. METHODS: Various fibrotic biomarkers such as kidney injury molecule-1 (KIM-1), alanine aminotransferace (ALT), aspartate aminotransferase (AST), and its ratio to platelet index

Organ-specific colonization and niche remodeling in a human tissue model of metastasis.

For metastatic colonization to occur, disseminated tumor cells must survive, adapt to, and remodel distant microenvironments in an organ-specific manner. We established a human multitissue model of cancer spread, with engineered bone and lung linked by vascular flow containing circulating cancer cells. Parental MDA-MB-231 cells extravasated toward both tissues, remodeled their niches, and acquired transcriptional programs reflecting adaptation to the local microenvironment, particularly upon homing to bone. Tissue-specific colonization by the bone- and lung-tropic MDA-MB-231 derivatives was qu

Organism-Specific Sequence Motifs Link Ribosomal RNAs to Brain Disorders.

We report that in humans, mice, fruit flies, and worms, the ribosomal RNAs and the transcribed spacers of 45S are densely packed with organism-specific sequence motifs that are primarily shared with nervous system genes. The human ribosomal RNAs and 45S spacers contain 1,723 such motifs. Specific combinations of these motifs are predominantly found in 3,430 human nervous system genes, of which 1,046 are genes associated with brain disorders, including autism spectrum disorder and schizophrenia. The sequences of the 1,723 motifs and their locations in the introns and exons of nervous system gen

Organ-specific lithium accumulation and its toxic effects on female reproduction.

Li exposure is becoming a new prominent major issue in energy-environment-health field. However, its reproductive health risk has been less explored. In this study, we find that exposure to 28-fold REED (really environmental exposure dose) of Li is enough to induce mouse miscarriage by altering the structures and functions of placenta. Exposure to ≥ 5.6-fold REED of Li is enough to alter the structure and functions of ovary and placenta, and exposure to ≥ 28-fold REED of Li alters fetus morphology and uterus functions, showing a susceptibility to Li exposure as the order ovary > placenta > fet

Analysis of Organ-Specific Outcomes in Transferred Emergency General Surgery Patients.

INTRODUCTION: Interhospital transfer for emergency general surgery (EGS) is often necessary to access a higher level of care. Compared to EGS patients admitted at the operating institution (nontransfer patients), EGS transfer patients represent a higher-risk population. This study sought to evaluate the association between transfer status and postoperative complications in EGS patients. METHODS: Using the 2010-2022 American College of Surgeons' National Surgical Quality Improvement Program database, we identified patients aged ≥18 years who underwent an EGS operation. Inflammation and Fibrosis in Progeria: Organ-Specific Responses in an HGPS Mouse Model.

Hutchinson-Gilford Progeria Syndrome (HGPS) is an extremely rare genetic disorder that causes accelerated aging, due to a pathogenic variant in the LMNA gene. This pathogenic results in the production of progerin, a defective protein that disrupts the nuclear lamina's structure. In our study, we conducted a histopathological analysis of various organs in the LmnaG609G/G609G mouse model, which is commonly used to study HGPS. The objective of this study was to show that progerin accumulation drives systemic but organ-specific tissue damage and accelerated aging phenotypes. Our findings show sign

Polymicrobial infections with specific Actinomyces and related organisms, using the current taxonomy.

Actinomyces organisms reside on mucosal surfaces of the oropharynx and the genitourinary tract. Polymicrobial infections with Actinomyces organisms are increasingly being reported in the literature. Since these infections differ from classical actinomycosis, lacking of specific clinical and imaging findings, slow-growing Actinomyces organisms can be regarded as contaminants or insignificant findings. In addition, only limited knowledge is available about novel Actinomyces species and their clinical relevance. The recent reclassifications have resulted in the transfer of several Actinomyces spe

Cell line-specific features of 3D chromatin organization in hepatocellular carcinoma.

Liver cancer, particularly hepatocellular carcinoma (HCC), poses a significant global threat to human lives. To advance the development of innovative diagnostic and treatment approaches, it is essential to examine the hidden features of HCC, particularly its 3D genome architecture, which is not well understood. In this study, we investigated the 3D genome organization of four HCC cell lines-Hep3B, Huh1, Huh7, and SNU449-using in situ Hi-C and assay for transposase-accessible chromatin sequencing. Our findings revealed that HCC cell lines had more long-range interactions, both intra-and interch
