Collagenase-3 (MMP-13) is expressed in periapical lesions: an immunohistochemical study.

AIM: To determine whether or not matrix metalloproteinase 13 (MMP-13) is present in periapical granulomas with and without epithelium. METHODOLOGY: Seventeen open periapical granulomas of pulpal origin (seven lesions without epithelium and 10 with proliferating epithelium) were fixed in formalin and then embedded in paraffin prior to being processed for immunohistochemical analysis. A monoclonal antibody against human MMP-13 was used to evaluate MMP-13 expression. Immunocomplexes were subsequently treated with the secondary antibody and then detected by means of streptavidin peroxidase. Immun

[Application of tricalcium phosphate in one visit root canal treatment].

This paper reports the successful use of tricalcium phosphate (TCP) as periapical barrier material in animal experiments and clinics. The results of animal experiments showed that TCP had good biocompatibility with periapical tissues. At 6 weeks there were osteoids deposited on the surface of TCP. At 12 weeks osteogenesis occurred around the TCP particles and combined each other closely. At 24 weeks around the periapex there were bone deposits and a tendency of apical closure. On the basis of experiments TCP was used to treat 17 chronic periapicitis in 15 patients. All cases were young permane…

Detection of interleukin-1 beta in human periapical lesions.

Interleukins are involved in modulating bone cell activity. Interleukin-1 (IL-1) has been shown to be potent stimuli of bone resorption in organ culture. This hormone-peptide is produced primarily by monocytes and macrophages. Diverse inflammatory cell types are clearly present in periapical lesions. The purpose of this study was to detect IL-1 beta activity in human periapical lesions. Eight human periapical lesions were examined for the presence of IL-1 beta. Pulp tissue of clinically impacted teeth were used as controls. Each specimen was quick-frozen and stored in liquid nitrogen. IL-1 bet

Human Umbilical Cord Mesenchymal Stem Cells-Derived Extracellular Vesicles for Rat Jawbone Regeneration in Periapical Periodontitis.

Extracellular vesicles derived from mesenchymal stem cells (MSCs-EVs) have great potential for bone remodeling and anti-inflammatory therapy. For the repair and reconstruction of inflammatory jawbone defects caused by periapical periodontitis, bone meal filling after debridement is commonly used in the clinic. However, this treatment has disadvantages such as large individual differences and the need for surgical operation. Therefore, it is of great significance to search for other bioactive substances that can promote jawbone regeneration in periapical periodontitis. Herein, it is found that

Magnetic resonance imaging in the diagnosis of periodontal and periapical disease.

OBJECTIVES: Early pre-clinical inflammatory changes in periodontal and/or periapical lesions, which typically precede bone loss, are challenging to diagnose using ionizing-radiation-based imaging modalities. MRI provides relevant additional diagnostic information of inflammatory processes in soft and hard tissues. The aim of the present study is to undertake a systematic review of the literature on MRI in the diagnosis of periodontal and/or periapical disease. METHODS AND MATERIALS: The PubMed/MEDLINE and Scopus bibliographic databases were searched (2000-2021) using the search string: ("MRI"

The Expression of Semaphorin 7A in Human Periapical Lesions.

INTRODUCTION: Semaphorin 7A (SEMA7A) is a membrane-bound or secretory protein exerting multiple functions in the regulation of inflammation, neural degradation, and cancer progression. Human periapical lesions are chronic and infectious diseases mainly caused by bacteria. However, the involvement of SEMA7A in human periapical lesions is still unclear. This study aimed to explore the expression of SEMA7A in human periapical lesions accompanied by the potential association of SEMA7A with matrix metalloproteinase (MMP)-1 and MMP-3 during the progression of apical periodontitis. METHODS: Samples

Local Immunomodulatory Effects of Intracanal Medications in Apical Periodontitis.

The immune system is an extremely complex biological network that plays a crucial role in the hemostasis of periapical tissue, pathogenesis of apical periodontitis (AP), and periapical tissue healing. The successful elimination of microbial infections remains a significant challenge, mostly because of the ever-growing development of antimicrobial-resistant pathogens. The bacterial endurance in the root canal system contributes to features ranging from altered posttreatment healing to exacerbation of chronic periradicular immune response, which compromise the outcome of endodontic treatment. A …

Nlrp3 inflammasome drives regulatory T cell depletion to accelerate periapical bone erosion.

AIM: Apical periodontitis is an inflammatory disorder triggered by an immune response to bacterial infection, leading to the periapical tissue damage and alveolar resorption. However, the underlying mechanisms driving this process remain elusive, due to the complex and interconnected immune microenvironment within the local lesion site. In this study, the influence of Nlrp3 inflammasome-mediated immune response on the apical periodontitis was investigated. METHODOLOGY: RNA sequencing, immunohistochemistry and ELISA assay were performed to investigate the activation of Nlrp3 inflammasome signa…

Extracellular matrix degradation by host matrix metalloproteinases in restorative dentistry and endodontics: An overview.

Matrix metalloproteinases (MMPs) are a group of over 25 secreted and membrane-bound enzymes responsible for pericellular substrate degeneration. In response to injury, they play key roles in morphogenesis, wound healing, tissue repair and remodeling. They have been isolated from dentin, odontoblasts, pulp and periapical tissue. They play a major role in the formation of dentin matrix and secondary and tertiary dentin. These are also responsible for releasing dentinal growth factors. MMP family proteins elicit a dual role in the pathogenesis of inflammation, stimulating protective innate and/or…
