DNA replication dynamics during erythrocytic schizogony in the malaria parasites Plasmodium falciparum and Plasmodium knowlesi.

Malaria parasites are unusual, early-diverging protozoans with non-canonical cell cycles. They do not undergo binary fission, but divide pri … st time the dynamics of schizogony in two human malaria parasite species, Plasmodium falciparum and Plasmodium knowlesi. These differ in their cell-cycle length, the number of progeny produced and the genome composition, among other factors. Comparing them could therefore yield new information about the parameters and limitations of schizogony. We report that the dyn …

Plasmodium knowlesi and global changes: socio-ecological drivers and challenges.

Plasmodium knowlesi, a malaria species predominantly found in Southeast Asia, is influenced by climate change in its transmission dynamics. Variations in temperature and precipitation impact both mosquito vectors and the parasites they carry. Additionally, deforestation and alterations in land use, often driven by climate change, facilitate the zoonotic transmission of the disease from simian hosts to humans. Climatic fluctuations also influence human and community behaviours, potentially altering exposure risks. Although increased awareness of climate risks may prompt behavioural changes, the…

Recombinase-Aided Loop-Mediated Isothermal Amplification on Human Plasmodium knowlesi.

Loop-mediated isothermal amplification (LAMP) is a nucleic acid amplification technique that can amplify specific nucleic acids at a constant temperature (63-65°C) within a short period (<1 hour). In this study, we report the utilization of recombinase-aided LAMP to specifically amplify the 18S sRNA of Plasmodium knowlesi. The method was built on a conventional LAMP assay by inclusion of an extra enzyme, namely recombinase, into the master mixture. With the addition of recombinase into the LAMP assay, the assay speed was executed within a time frame of less than 28 minutes at 65°C. We screened…

Anti-inflammatory cytokines predominate in acute human Plasmodium knowlesi infections.

Plasmodium knowlesi has entered the human population of Southeast Asia. Naturally acquired knowlesi malaria is newly described with relatively little available data, including data on the host response to infection. Therefore pre-treatment cytokine and chemokine profiles were determined for 94 P. knowlesi, and for comparison, 20, P. vivax and 22 P. falciparum, patients recruited in Malaysian Borneo. Nine, five and one patient with P. knowlesi, P. falciparum and P. vivax respectively had complicated malaria as defined by World Health Organisation. Patients with uncomplicated P. knowlesi had low…

Swedish traveller with Plasmodium knowlesi malaria after visiting Malaysian Borneo.

Plasmodium knowlesi is typically found in nature in macaques and has recently been recognized as the fifth species of Plasmodium causing malaria in human populations in south-east Asia. A case of knowlesi malaria is described in a Swedish man, who became ill after returning from a short visit to Malaysian Borneo in October 2006. His P. knowlesi infection was not detected using a rapid diagnostic test for malaria, but was confirmed by PCR and molecular characterization. He responded rapidly to treatment with mefloquine. Evaluation of rapid diagnostic kits with further samples from knowlesi mala…

Human Plasmodium knowlesi infections in young children in central Vietnam.

BACKGROUND: Considering increasing reports on human infections by Plasmodium knowlesi in Southeast Asian countries, blood samples collected during two large cross-sectional malariometric surveys carried out in a forested area of central Vietnam in 2004 and 2005 were screened for this parasite. METHODS: Blood samples collected at the 2004 survey and positive for Plasmodium malariae were randomly selected for PCR analysis detecting P. knowlesi. Blood samples collected in 2005 from the same individuals were screened again for P. knowlesi. Positive samples were confirmed by sequencing. Family mem…

Renal plasma flow in rhesus monkeys infected with Plasmodium knowlesi.

Renal plasma flow was studied in six rhesus monkeys before and during infection with Plasmodium knowlesi using 125I-sodium orthoiodohippurate (125I-OIH) as a tracer. The mean renal plasma flow and the rate constant from the intravascular compartment to the kidneys were significantly reduced in the infected monkeys. As both intravascular and extravascular compartments were slightly but not significantly elevated, which resulted in the prolonged mean transit time of 125I-OIH in monkeys infected with P. knowlesi. The parasitemia showed a reverse relationship with the renal plasma flow and a direc…

Plasmodium knowlesi sporozoite antigen: expression by infectious recombinant vaccinia virus.

The gene coding for the circumsporozoite antigen of the malaria parasite Plasmodium knowlesi was inserted into the vaccinia virus genome under the control of a defined vaccinia virus promoter. Cells infected with the recombinant virus synthesized polypeptides of 53,000 to 56,000 daltons that reacted with monoclonal antibody against the repeating epitope of the malaria protein. Furthermore, rabbits vaccinated with the recombinant virus produced antibodies that bound specifically to sporozoites. These data provide evidence for expression of a cloned malaria gene in mammalian cells and illustrate…

Erythrocyte receptors for (Plasmodium knowlesi) malaria: Duffy blood group determinants.

Duffy blood group negative human erythrocytes (FyFy) are resistant to infection by Plasmodium knowlesi, a simian malaria that infects Duffy positive human erythrocytes. The P. knowlesi resistance factor, Duffy negative erythrocytes, occurs in high frequency in West Africa, where the people are resistant to vivax malaria. This suggests that Duffy blood group determinants (Fya or Fyb) may be erythrocyte receptors for P. vivax.

Plasmodium knowlesi malaria: assessing the risk to the British Armed Forces.

Plasmodium knowlesi is a zoonosis and is now recognised as the fifth commonly occurring form of human malaria. It is endemic in South East Asia, including some areas previously declared malaria free or at low risk for malaria. The epidemiology of the disease is very different to other forms of malaria which are determined by transmission by anthrophilic mosquitoes from human reservoirs. In contrast Plasmodium knowlesi malaria has a monkey reservoir and disease is transmitted to humans by mosquitoes that normally feed on animals. People become accidentally infected when they enter the ranges of…

Plasmodium knowlesi: reservoir hosts and tracking the emergence in humans and macaques.

Plasmodium knowlesi, a malaria parasite originally thought to be restricted to macaques in Southeast Asia, has recently been recognized as a significant cause of human malaria. Unlike the benign and morphologically similar P. malariae, these parasites can lead to fatal infections. Malaria parasites, including P. knowlesi, have not yet been detected in macaques of the Kapit Division of Malaysian Borneo, where the majority of human knowlesi malaria cases have been reported. In order to extend our understanding of the epidemiology and evolutionary history of P. knowlesi, we examined 108 wild maca…

Plasmodium knowlesi in humans: a review on the role of its vectors in Malaysia.

Plasmodium knowlesi in humans is life threatening, is on the increase and has been reported from most states in Malaysia. Anopheles latens and Anopheles cracens have been incriminated as vectors. Malaria is now a zoonoses and is occurring in malaria free areas of Malaysia. It is also a threat to eco-tourism. The importance of the vectors and possible control measures is reviewed here.

Molecular epidemiological investigation of Plasmodium knowlesi in humans and macaques in Singapore.

Singapore reported its first locally acquired human Plasmodium knowlesi infection in 2007, involving a soldier who had undergone training in a forested area where long-tailed macaques are frequently seen. Comprehensive disease surveillance and monitoring system that was set up after the initial case detected four additional human P. knowlesi cases in 2007 and one in 2008. All involved military personnel who had undergone training in the forested area, and none had traveled out of Singapore 1 month before the onset of symptoms. Screening for malaria parasites on blood obtained from long-tailed …

Nuclear and mitochondrial DNA of the primate malarial parasite Plasmodium knowlesi.

Restriction analyses and DNA/DNA hybridisation of parasite DNA isolated from monkeys infected with the malarial parasite Plasmodium knowlesi has permitted unambiguous identification of the nuclear DNA of this species. Its (G+C) content, as determined by estimations of buoyant density as well as by direct analysis, is about 38%, essentially indistinguishable from that of its primate laboratory host, and grossly different from that of the major human malaria parasite, P. falciparum, which has a (G+C) content of approx. 19%. In addition, gradient fractionation of total P. knowlesi DNA revealed a …

Neutralization of the infectivity of sporozoites of Plasmodium knowlesi by antibodies to a synthetic peptide.

Antibodies against a synthetic peptide representing the repetitive epitope of the circumsporozoite protein (CS) of Plasmodium knowlesi have properties similar to those of antibodies against the native protein. Either antibody reacts with the synthetic peptide, cross-links the CS protein on the membrane of the parasite giving the CSP reaction, and neutralizes the infectivity of sporozoites. The synthetic peptide and sporozoite extracts were equally effective when used in an immunoradiometric assay as antigens to detect antibodies to CS proteins. It is likely that the corresponding synthetic rep…
