Local chemokine paralysis, a novel pathogenic mechanism for Porphyromonas gingivalis.

Periodontitis, which is widespread in the adult population, is a persistent bacterial infection associated with Porphyromonas gingivalis. Gingival epithelial cells are among the first cells encountered by both P. gingivalis and commensal oral bacteria. The chemokine interleukin 8 (IL-8), a potent chemoattractant and activator of polymorphonuclear leukocytes, was secreted by gingival epithelial cells in response to components of the normal oral flora. In contrast, P. gingivalis was found to strongly inhibit IL-8 accumulation from gingival epithelial cells. Inhibition was associated with a decre…

Life below the gum line: pathogenic mechanisms of Porphyromonas gingivalis.

Porphyromonas gingivalis, a gram-negative anaerobe, is a major etiological agent in the initiation and progression of severe forms of periodontal disease. An opportunistic pathogen, P. gingivalis can also exist in commensal harmony with the host, with disease episodes ensuing from a shift in the ecological balance within the complex periodontal microenvironment. Colonization of the subgingival region is facilitated by the ability to adhere to available substrates such as adsorbed salivary molecules, matrix proteins, epithelial cells, and bacteria that are already established as a biofilm on to…

Secretion of Porphyromonas gingivalis fimbrillin polypeptides by recombinant Streptococcus gordonii.

The fimbriae of Porphyromonas gingivalis plays an important role in the pathogenesis of periodontal disease. A structural subunit of the P. gingivalis fimbriae, fimbrillin, has been shown to promote adherence of the bacteria to host surfaces and also induce an immune response. Biologically active domains of fimbrillin responsible for adherence or eliciting immune responses have been determined. In a previous study, we engineered the human oral commensal organism Streptococcus gordonii to express such biologically active domains on the surface of the bacteria as a vaccine delivery system. In th…

Porphyromonas gingivalis outer membrane vesicles promote bacterial resistance to chlorhexidine.

Porphyromonas gingivalis has been frequently associated with some types of periodontal diseases and possesses various mechanisms favoring the pathogenic process. It has been recently observed that vesicles elaborated by P. gingivalis are able to protect bacteria from the bactericidal activity of human serum. The aim of the present investigation was to evaluate the ability of vesicles from P. gingivalis in protecting oral bacteria against chlorhexidine. Data indicate that vesicles released by P. gingivalis may bind chlorhexidine, thus allowing protection for itself and for other oral bacterial …

Chemical structure of lipid A from Porphyromonas (Bacteroides) gingivalis lipopolysaccharide.

The novel chemical structure of Porphyromonas (Bacteroides) gingivalis strain 381 lipid A was determined to be a glucosamine beta-(1-6) disaccharide 1-monophosphate acylated by 3-hydroxy-15-methylhexadecanoic acid and 3-hexadecanoyloxy-15-methylhexadecanoic acid at the 2- and 2'-positions, respectively. The absence of ester-linked phosphate at the 4'-position and fatty acids at the 3- and 3'-positions, and the presence of fatty acids possessing 16-17 carbon atoms are unique features, differentiating the lipid A from enterobacterial and other lipid As. These structural features may be related t…

A CRISPR array orchestrates virulence and host response in Porphyromonas gingivalis.

CRISPR-Cas systems are primarily recognized for their defensive role against foreign DNA. Recent studies, however, indicate involvement in regulatory functions. The persistence of a large spacer array without apparent phage targets in Porphyromonas gingivalis ATCC 33277 remains unexplained. This study demonstrates that deletion of the non-coding CRISPR array 30.1 in P. gingivalis ATCC 33277 results in increased biofilm formation, elevated virulence in a Galleria mellonella infection model, and significant alterations in the macrophage transcriptomic response. The ΔCRISPR-30.1 mutant forms twic…

Association of Porphyromonas gingivalis with Acute Myocardial Infarction: A Systematic Review.

Background/Objectives: Periodontitis is a chronic oral infection in which Porphyromonas gingivalis (Pg) acts as a keystone pathogen capable of systemic dissemination and immune evasion. A possible association between Pg and acute myocardial infarction (AMI) has been proposed; this systematic review is, to our knowledge, among the first to evaluate this association through a pathogen-specific synthesis integrating both microbial and serological evidence. Methods: Five electronic databases (PubMed, Scopus, Embase, Web of Science, Cochrane Library) were searched from inception to February 2025, w…

Development of a Quantitative Detection System for Porphyromonas gingivalis Based on Immunochromatography.

Background: Porphyromonas gingivalis is the principal bacterial species implicated in the pathogenesis of periodontitis. Despite its central role, direct detection of periodontopathic bacteria is not routinely performed in dental clinics. Although test kits using monoclonal antibodies against P. gingivalis have been reported, no practical chairside method has yet been established. Objectives: In this study, we developed an immunochromatography system that enables rapid and quantitative detection of P. gingivalis within 10 min. Methods: We evaluated the performance of the developed immunochroma…

Structural and biochemical characterization of a zinc metallopeptidase from Porphyromonas gingivalis.

The pathogen Porphyromonas gingivalis contributes to the pathogenesis of periodontitis and other systemic diseases. The zinc-dependent metallopeptidase PepO is a virulence factor that plays a crucial role in the adhesion and invasion of Porphyromonas gingivalis to human cells. Here, we solved the 2.04 Å crystal structure of wild-type PepO in complex with the inhibitor phosphoramidon. The active-site pocket of PepO appears to exhibit an increased hydrophobicity and a more pronounced negative charge, highlighting distinct structural features compared to its homologs. In addition to phosphoramido…

CD47 and thrombospondin-1 contribute to immune evasion by Porphyromonas gingivalis.

Porphyromonas gingivalis is a gram-negative anaerobic bacterium linked to periodontal disease. Remarkably, P. gingivalis thrives in an inflamed environment rich in activated neutrophils. Toll-like receptor 2 (TLR2) recognition is required for P. gingivalis to evade innate immune killing; however, the mechanisms through which P. gingivalis uncouples host inflammation from bactericidal activity are only partially known. Since integrin activation and alternative signaling are implicated in P. gingivalis TLR2-mediated immune escape, we explored the role of CD47, a widely expressed integrin-associa…

Porphyromonas gulae and PPAD antibodies are not related to citrullination in rheumatoid arthritis.

INTRODUCTION: Porphyromonas gulae have the enzyme PPAD, as P. gingivalis, which is responsible for citrullination related to the pathophysiology of rheumatoid arthritis and periodontitis; this implies the presence of two species of PPAD-producing bacteria in the mouth as well as the presence of citrullinated proteins. There are no previous reports or studies investigating an association between P. gulae PPAD in rheumatoid arthritis (RA). OBJECTIVE: To assess the presence of P. gulae and anti-citrullinated peptide antibodies of P. gulae PAD in patients with RA and their possible relationship w…

[Porphyromonas gingivalis HmuY and Its Possible Effect on the Pathogenesis of Periodontitis].

Periodontitis, one of the most common inflammatory oral diseases in human beings, threatens the health of teeth and mouth and is closely associated with the development of many systemic diseases. Existing research about the pathogenesis of periodontitis mainly focuses on the oral microbial homeostasis and its complex interaction with the immune system. Among all the oral microorganisms, Porphyromonas gingivalis ( P. gingivalis) is considered to be the main pathogen causing chronic periodontitis. Recent studies have shown that P. gingivalis poesseses HmuY, a special heme binding protein, which …

Elimination of Porphyromonas gingivalis inhibits liver fibrosis and inflammation in NASH.

AIM: Non-alcoholic steatohepatitis (NASH) is a critical liver disease showing potential progression to liver cirrhosis/cancer. Previously, we had reported that odontogenic infection of Porphyromonas gingivalis (P. gingivalis), a major periodontal pathogen, exacerbates fibrosis in NASH through the production of fibrosis mediators such as transforming growth factor-β1 (TGF-β1) and galectin-3. In this study, we determined the effects of therapeutic interventions using antibiotics on NASH progression induced by P. gingivalis odontogenic infection. MATERIALS AND METHODS: To eliminate P. gingivalis…
