Divergent Chiral Self-Sorting of Covalent Organic Pillars Switched by Subtle Conformational Preferences.

Chiral self-sorting enables molecular systems to translate stereochemical information from small building blocks into highly organized architectures across length scales, echoing the hierarchical expression of chirality in biological systems. However, predicting whether a system will favor homochiral or heterochiral organization, let alone rationally controlling the sorting outcome, remains challenging. Here we show that the chiral self-sorting of covalent organic pillars (COPs) can be selectively switched through minimal structural variation in the linker units. Two diamine linkers, 2,6-bis(a

An optimized base editor with efficient C-to-T base editing in zebrafish.

BACKGROUND: Zebrafish is a model organism widely used for the understanding of gene function, including the fundamental basis of human disease, enabled by the presence in its genome of a high number of orthologs to human genes. CRISPR/Cas9 and next-generation gene-editing techniques using cytidine deaminase fused with Cas9 nickase provide fast and efficient tools able to induce sequence-specific single base mutations in various organisms and have also been used to generate genetically modified zebrafish for modeling pathogenic mutations. However, the editing efficiency in zebrafish of currentl

Use of drugs, perceived drug efficacy and preferred providers for febrile children: implications for home management of fever.

BACKGROUND: Community distribution of anti-malarials and antibiotics has been recommended as a strategy to reduce the under-five mortality due to febrile illnesses in sub-Saharan Africa. However, drugs distributed in these interventions have been considered weak by some caretakers and utilization of community medicine distributors has been low. The aim of the study was to explore caretakers' use of drugs, perceptions of drug efficacy and preferred providers for febrile children in order to make suggestions for community management of pneumonia and malaria. METHODS: The study was conducted in

Septic arthritis in a previously healthy man with pan-negative infectious and rheumatologic work-up.

Neisseria gonorrhoeae is the causative organism in 0.6%-1.2% of septic arthritis cases in North America and Europe, and classically presents as migratory polyarthralgias and tenosynovitis, with later development of septic oligoarthritis. In men, urine gonorrhoea nucleic amplification testing (NAAT) is the preferred diagnostic test, as its sensitivity surpasses that of joint and blood culture in disseminated infections. We present a case of a previously healthy man who presented with septic arthritis of the wrist. He denied any sexual activity in the previous year. Urine gonorrhoea NAAT and cul

Personal Health Libraries for People Returning From Incarceration: Protocol for a Qualitative Study.

BACKGROUND: Individuals released from carceral facilities have high rates of hospitalization and death, especially in the weeks immediately after their return to community settings. During this transitional process, individuals leaving incarceration are expected to engage with multiple providers working in separate, complex systems, including health care clinics, social service agencies, community-based organizations, and probation and parole services. This navigation is often complicated by individuals' physical and mental health, literacy and fluency, and socioeconomic status. Personal healt

Real-Time Viability Assessment of Ex Vivo Mouse Kidneys for Transplant Applications Using Dynamic Optical Coherence Tomography.

Kidney transplantation remains the preferred treatment for patients with end-stage kidney disease. However, the ongoing shortage of donor organs continues to limit the availability of transplant treatments. Existing evaluation methods, such as the kidney donor profile index (KDPI) and pre-transplant donor biopsy (PTDB), have various limitations, including low discriminative power, invasiveness, and sampling errors, which reduce their effectiveness in organ quality assessment and contribute to the risk of unnecessary organ discard. In this study, we explored the dynamic optical coherence tomogr

The 'gut bucket': a novel training tool for standardised patients.

BACKGROUND: Standardised patients (SPs) are often asked to portray complex physical roles in which authenticity is paramount; however, SPs come from a variety of backgrounds, and may lack knowledge of basic anatomy and physiology that can hamper their portrayal. This lack of knowledge can lead to gaps in accuracy and credibility. CONTEXT: In our efforts to bridge the gap and create training that would lead to authentic and confident portrayals, we developed the 'gut bucket'. This three-dimensional learning tool is tactile, portable, simple and affordable. INNOVATION: The 'gut bucket' has lif

Construction and Evaluation of Novel Dual-function Antifungal Inhibitors and Covalent Organic Framework Carriers Based on the Infection Microenvironment.

In this study, PD-L1 and CYP51 were selected as key dual-target enzymes, which play an important role in the process of fungal proliferation and immune suppression. A series of novel bifonazole dual-target compounds were designed through the method of fragment combination. Their chemical structure was synthesized, characterized, and evaluated. Among them, the compounds (10c-1, 14a-2, 17c-2) exhibited excellent antifungal and antidrug-resistant fungal activity in vitro. In particular, the preferred compound 14a-2 with high-efficiency dual-target inhibitor ability could block the fungal prolifer

Obesity in Cardiac Rehabilitation: Considerations in Offering Weight Management As Part of Cardiac Rehabilitation Programs.

Despite its relevance to cardiovascular health, obesity is rarely targeted during cardiac rehabilitation (CR). The objective of this paper was to review evidence regarding whether measures to address excess body fat should be offered as a standard component of CR for patients with obesity. We organize the paper around three themes: 1) outcomes of obesity management, 2) the complexity of obesity management, and 3) patient attitudes, experiences, and preferences. Our discussion of each theme was informed by a narrative literature review and a survey of Canadian CR healthcare providers (n=80). We

Control of Mycoplasma hyopneumoniae infections in pigs.

Mycoplasma hyopneumoniae, the primary pathogen of enzootic pneumonia, occurs worldwide and causes major economic losses to the pig industry. The organism adheres to and damages the ciliated epithelium of the respiratory tract. Affected pigs show chronic coughing, are more susceptible to other respiratory infections and have a reduced performance. Control of the disease can be accomplished in a number of ways. First, management practices and housing conditions in the herd should be optimized. These include all-in/all-out production, limiting factors that may destabilize herd immunity, maintaini

Retrospective and prospective perspectives on zoonotic brucellosis.

Members of the genus Brucella are pathogenic bacteria exceedingly well adapted to their hosts. The bacterium is transmitted by direct contact within the same host species or accidentally to secondary hosts, such as humans. Human brucellosis is strongly linked to the management of domesticated animals and ingestion of their products. Since the domestication of ungulates and dogs in the Fertile Crescent and Asia in 12000 and 33000 ya, respectively, a steady supply of well adapted emergent Brucella pathogens causing zoonotic disease has been provided. Likewise, anthropogenic modification of wild

Predictors of public attitude toward living organ donation in Kano, northern Nigeria.

Organ shortage is a major public health challenge for transplant programs globally. The sustenance of such programs as an effective therapy for end-stage organ failure (ESOF) requires an exploration of public awareness and willingness to donate organs. This is imperative, especially in developing countries where ESOF is highly prevalent. We studied the awareness and predictors of public attitude toward organ donation in Kano city in northern Nigeria. Using interviewer-administered questionnaires, we assessed the awareness and willingness to donate solid organs among 400 adults in the Kano metr

Cryo-EM structure of Escherichia coli HPII catalase serendipitously co-purified with a recombinant adhesin construct.

Catalases are key antioxidant enzymes that protect aerobic organisms from reactive oxygen species generated through metabolic activities and host immune responses. In Escherichia coli, the monofunctional HPII catalase (encoded by katE) forms a stable tetrameric enzyme that contributes to oxidative stress resistance. During the purification of a recombinantly produced construct of the Vibrio cholerae FrhA adhesin, we unexpectedly observed a co-purifying protein species which peptide mass spectrometry identified as E. coli HPII catalase. Initial reconstruction was limited by severe preferred ori
