Piperonyl butoxide (PBO) is a synergist used in some pyrethroid and pyrethrin pesticide products and has been used in toxicity identification evaluations (TIEs) of water samples to indicate organophosphate or pyrethroid-related toxicity. Methods were developed and validated for use of PBO as a TIE tool in whole-sediment testing to help establish if pyrethroids are the cause of toxicity observed in field-collected sediments. Pyrethroid toxicity was increased slightly more than twofold in 10-d sediment toxicity tests with Hyalella azteca exposed to 25 microg/L of PBO in the overlying water. This

The pyrethroids are a class of natural and synthetic pesticides which were associated with an epidemic of gynecomastia in Haitian men in 1981. In the present study we tested several pyrethroids for their ability to interact with androgen binding sites in dispersed, intact human genital skin fibroblasts and in human plasma to sex hormone binding globulin (SHBG). All the pyrethroids tested inhibited fibroblast binding of [3H]methyltrienolone (R1881) at 22 degrees C with the following rank order of potency:pyrethrins greater than bioallethrin greater than fenvalerate greater than fenothrin greate…

Leptospermone, a natural β-triketone and major constituent of manuka oil (Leptospermum scoparium), is an established inhibitor of plant HPPD … osquitocide that induces rapid knockdown and toxicity to Ae. aegypti at doses that approach natural pyrethrins against pyrethroid-resistant mosquito strains. Further, the data indicate leptospermone mode of action is CA inhibition, which is a novel mosquitocide target and is different when compared to the mode of action in plants.

BACKGROUND: The Anopheles gambiae species complex comprises about nine sibling species, including An. gambiae, An. coluzzii, and An. arabiensis, three of the four major malaria vector species across Africa. Insecticide-based vector control remains the most critical tool in the fight against malaria, with a heavy reliance on pyrethroid insecticides. However, widespread pyrethroid resistance jeopardises the effectiveness of these methods. Currently, the number of diagnostic markers available to track pyrethroid resistance in endemic settings is limited, focusing mainly on changes at the insectic

Pesticides with vastly different modes of action are needed to combat pests that have become multipesticide resistant. One proposed alternative is the pyrrole insecticide chlorfenapyr. This propesticide needs bioactivation by CYP450 enzymes to form the more potent metabolite tralopyril. However, its effects on nontarget soil organisms are largely unknown. This study aimed to assess the toxicity of chlorfenapyr to Folsomia candida and Enchytraeus crypticus, two model species for soil ecotoxicology, following Organisation for Economic Co-operation and Development (OECD) guidelines. Tests were pe

Since the 1980s, monarch butterfly (Danaus plexippus plexippus) populations across North America have declined by 80%-95%. Although several studies have implicated pesticides as a contributing factor to their population declines, our understanding of monarch exposure levels in nature remains limited. In January 2024, a mass mortality event near an overwintering site in Pacific Grove, California, USA, provided an opportunity to analyze dead overwintering monarch butterflies for pesticide residues. Ten recently deceased butterflies were collected and analyzed using liquid and gas chromatography

Aedes albopictus (Ae. albopictus) is widely distributed and can transmit many infectious diseases, and insecticide-based interventions play an important role in vector control. However, increased insecticide resistance has become a severe public health problem, and the clarification of its detailed mechanism is a matter of urgence. This study found that target-site resistance and metabolic resistance could not fully explain insecticide resistance in field Ae. albopictus, and there were likely other resistance mechanisms involved. The 16S and internal transcribed spacer sequencing revealed sign

The extensive use of chemical insecticides for public health and agricultural purposes has increased the occurrence and development of insecticide resistance. This study used transcriptome sequencing to screen 10 upregulated metabolic detoxification enzyme genes from Aedes albopictus resistant strains. Of these, CYP6A14 and CYP6N6 were found to be substantially overexpressed in the deltamethrin-induced expression test, indicating their role in deltamethrin resistance in Ae. albopictus. Furthermore, the corresponding 60-kDa recombinant proteins, CYP6A14 and CYP6N6, were successfully expressed u

Although reptiles are non-target organisms of pesticide applications, their ecological niche and trophic role suggest that the use of these compounds in agriculture can have toxicological effects on them. Our recent field study on Italian wall lizard Podarcis siculus in hazelnut orchards evidenced that the use of pesticides-mixtures, consisting of thiophanate-methyl (TM), tebuconazole (TEB), deltamethrin (DM), lambda-cyhalothrin (LCT), besides copper sulphate, induced an increase of the total antioxidant capacity toward hydroxyl radicals and caused DNA damage; however, it did not cause neuroto

In China, the extensive use of the pesticide chlorfenapyr has led to an increase in chlorfenapyr poisoning. However, there are limited reports on chlorfenapyr poisoning, and most of them are fatal cases. This study retrospectively analyzed four patients admitted to the emergency room after chlorfenapyr intake and detected different concentrations of chlorfenapyr in their plasma. Among them, one patient died and three patients survived. Case 1 suffered respiratory and circulatory failure with a deep coma shortly after oral administration of 100 mL of a the chlorfenapyr-containing mixture and di

BACKGROUND: The increasing reports of resistance to pyrethroid insecticides associated with reduced efficacy of pyrethroid-only interventions highlight the urgency of introducing new non-pyrethroid-only control tools. Here, we investigated the performance of piperonyl-butoxide (PBO)-pyrethroid [Permanet 3.0 (P3.0)] and dual active ingredients (AI) nets [Interceptor G2 (IG2): containing pyrethroids and chlorfenapyr and Royal Guard (RG): containing pyrethroids and pyriproxyfen] compared to pyrethroid-only net Royal Sentry (RS) against pyrethroid-resistant malaria vectors in Cameroon.

Use of agrochemicals, including insecticides, is vital to food production and predicted to increase 2-5 fold by 2050. Previous studies have shown a positive association between agriculture and the human infectious disease schistosomiasis, which is problematic as this parasitic disease infects approximately 250 million people worldwide. Certain insecticides might runoff fields and be highly toxic to invertebrates, such as prawns in the genus Macrobrachium, that are biocontrol agents for snails that transmit the parasites causing schistosomiasis. We used a laboratory dose-response experiment and

Pyrethroid insecticides have been broadly used as pest control in agriculture and residential spaces, exerting high effectiveness of insecticidal property and relatively low toxicity to humans. Several animal studies suggested that exposure to pyrethroids may induce hematological abnormalities, thereby altering the number of blood cells and resulting in blood disorders. However, no epidemiologic study has reported on the effect of pyrethroid insecticide exposure on hematological changes, except for occupational exposure. This study aimed to investigate the effect of urinary 3-phenoxybenzoic ac

The increased use of pyrethroid insecticides raises concern for exposure to non-target aquatic species, such as Chinook salmon (Oncorhynchus tshawytscha). Cypermethrin, a type II pyrethroid, is frequently detected in surface waters and sediments at concentrations that exceed levels that induce toxicity to several invertebrate and salmonid species. To better understand the effects of cypermethrin to salmonids following dietary exposure, juvenile Chinook salmon were dietarily exposed to a 0, 200, or 2000 ng/g cypermethrin diet for a duration of 7, 14, or 21 days and assessed for body burden resi

Anopheles epiroticus is a malaria vector in Thailand found primarily along coastal areas with brackish water habitats. Insecticides, particularly pyrethroid class compounds, are commonly used to control malaria vectors in Thailand. The lack of specific discriminating lethal concentrations for An. epiroticus has possibly compromised a more accurate assessment of physiological susceptibility to various chemicals. The routine assessment of vector response to insecticides is a key program management component to prevent or mitigate the development of resistance. The purpose of this study was to de

Prothrin, a synthetic pyrethroid insecticide, was synthesized from the biomass-derived platform chemical 5-(chloromethyl)furfural in six steps and overall 65% yield. Two structural analogues of prothrin were also prepared following the same synthetic approach. Preliminary testing of these furan-based pyrethroids against the yellow fever mosquito Aedes aegypti indicates promising insecticidal activities.
