The encoding of olfactory information in the central nervous system (CNS) depends on spatially distributed patterns of activity generated simultaneously in many neuronal circuits. Optical neurophysiological recording permits analysis of neural activity non-invasively and with high spatial and temporal resolution. Here, a video method for imaging voltage-sensitive dye fluorescence in vivo is used to map neuronal activity in local circuits of the salamander olfactory bulb. The method permits the imaging of simultaneous ensemble transmembrane activity in real time. After electrical stimulation of

Nicotinamide adenine dinucleotide (NAD+) plays a central role in energy metabolism, and its decline is linked to various degenerative diseases. While NAD+ restoration holds therapeutic promise, its long term, tissue-specific consequences remain poorly understood. We investigated effects of nicotinamide riboside (NR) supplementation for "mutator" mice manifesting mitochondrial progeria. Our results reveal strikingly divergent outcomes: in proliferative bone marrow, NR-treated mutators show reductive stress with accumulation of NADH/NADPH, altered amino acid, nucleotide, folate levels, and impai

Organophosphorus agents, also known as nerve agents, are very dangerous chemicals that were used as chemical warfare agents. HI-6 is one of the most promising reactivators which is effective in reactivating AChE inhibited by many nerve agents. However, the fast in-vivo clearance of HI-6 became a large barrier for first aid use under some sophisticated circumstances. In this study, PEGylated liposomes loading HI-6 were prepared and evaluated in vitro and in vivo. For PEG-LP-HI-6, the optimal formulation's loading efficiency and encapsulation efficiency were 6.47 ± 0.10% and 71.2 ± 1.15%, respec

The ongoing activity of the ganglionic cells innervating the normal and partial denervated submandibular gland of rat was investigated with intracellular recording. With the aim to be identified these cells were preliminary labeled by the fluorescent dye Di-1. The most of neurons innervating the normal gland were entirely silent. Ongoing activity was found only in 11% of these neurons and the average frequency of discharge was 0.1 +/- 0.01/s. After partial denervation of the gland, the number of active ganglion cells among the neurons innervating the gland increased from 11 to 42%. The average

Rapid, unidirectional pollen tube tip growth is a widely employed model for polar cell expansion, which is essential for plant morphogenesis. At the pollen tube tip, various proteins and lipids playing pivotal roles in regulating polar cell expansion are associated with distinct domains of the plasma membrane (PM) or accumulate within different cytoplasmic regions. These PM domains and cytoplasmic regions must be dynamically maintained during tip growth, which is contingent on massive secretory and endocytic membrane trafficking. The molecular and cellular mechanisms responsible for PM and cyt

C2 toxin from Clostridium botulinum represents the prototype of clostridial binary actin ADP-ribosylating toxins which destroy the actin-cytoskeleton of mammalian cells and cause severe enteric diseases in humans and animals. After receptor-mediated endocytosis of the C2 toxin complex, the binding/translocation component C2IIa forms a heptameric transmembrane pore in membranes of acidified endosomal vesicles. The separate ADP-ribosyltransferase component C2I translocates through this C2IIa-pore from the endosomal lumen into the cytosol. Here we demonstrate that positively charged heterocyclic

The potency of currently used oximes to reactivate sarin-inhibited acetylcholinesterase (AChE) in various parts of pig brain and whole pig brain was evaluated using in vitro methods. Significant differences in reactivation potency among all tested oximes were observed. At concentrations (10(-4) M) corresponding to recommended doses in vivo, the oxime HI-6 seems to be a more efficacious reactivator of sarin-inhibited AChE in whole pig brain as well as in cerebral hemispheres and cerebellum compared with the other oximes studied. Nevertheless, there are not any differences in the potency of oxim

Nucleophilic copper-mediated radioiodination (CMRI) of organoboronic precursors with radioiodides is a promising method of radioiodination. The previously reported CMRI has demonstrated its great potential and scope of labeling for the radiosynthesis of radioiodine-labeled compounds. However, the reported protocols (using a small amount/volume of radioactivity) are practically not reproducible in large-scale CMRI, in which the radioactivity was usually provided in a bulk alkaline solution. A large amount of water and a strong base are incompatible with CMRI. To overcome these issues in large-s

Perovskite-based solar cells have recently been catapulted to the cutting edge of thin-film photovoltaic research and development because of their promise for high-power conversion efficiencies and ease of fabrication. Two types of generic perovskites compounds have been used in cell fabrication: either Pb- or Sn-based. Here, we describe the performance of perovskite solar cells based on alloyed perovskite solid solutions of methylammonium tin iodide and its lead analogue (CH3NH3Sn(1-x)Pb(x)I3). We exploit the fact that, the energy band gaps of the mixed Pb/Sn compounds do not follow a linear

Using supramolecular chiral cages to create a favorable chiral environment can effectively address the limitations of traditional metal asymmetric catalysis in controlling chiral catalytically active centers. However, achieving harmonious interactions among the molecular cage, the metal, and the substrate within the cavity remains a significant challenge. To overcome this, we have designed a pyridinium-modified, chiral-diamine-functionalized cage with a distinct bowl-shaped geometry. This structure features three quaternary ammonium linkers at the base and three chiral cyclohexanediamine units

With the widespread use of antibiotics, bacterial resistance has developed rapidly. To make matters worse, infections caused by persistent bacteria and biofilms often cannot be completely eliminated, which brings great difficulties to clinical medication. In this work, three series of quinolone pyridinium quaternary ammonium small molecules were designed and synthesized. Most of the compounds showed good antibacterial activity against Gram-positive bacteria (S. aureus and E. faecalis) and Gram-negative bacteria (E. coli and S. maltophilia). The activity of the para-pyridine quaternary ammonium

Benzolamide, an orphan drug belonging to the pharmacological class of sulfonamide carbonic anhydrase (CA, EC 4.2.1.1) inhibitors (CAIs) is widely used in many physiological and pharmacological studies, together with the clinically employed classical drugs, acetazolamide, methazolamide, ethoxzolamide or dichlorophenamide, it being frequently stated that benzolamide is a membrane-impermeant inhibitor. We prove here that this is false: in fact benzolamide is rather similar to acetazolamide from the point of view of penetrability through blood red cell membranes. Unlike these neutral drugs, the ca

Spatially confining the membrane-fusion effects of fusogenic delivery systems in tumors can enhance therapeutic efficacy and reduce systemic toxicity. However, current existing fusion-regulating strategies often suffer from insensitivity and unidirectional control, leading to suboptimal antitumor effects and potential safety risks. Here, we designed a pyridinium betaine-capped lipid, 2-(4-((2,3-di(stearoyloxy)propyl)carbamoyl)pyridin-1-ium-1-yl)acetate (DSPCPA)), as a pH-responsive regulator to precisely manipulate the membrane-fusion behavior of fusogenic liposomes. Incorporating DSPCPA, we o

Mitochondria are known as the powerhouse of eukaryotic cells. They play a crucial role in several biological processes and maintain cellular health. The ideal condition of mitochondria depends not only on their morphology but also on various micro-environmental factors, including pH, polarity, and temperature. Changes in these factors or malfunctions of mitochondrial species, such as Reactive Oxygen Species (ROS), active nitrogen species, metal cations, anions, and protons, can lead to several diseases in humans, including heart failure, kidney disorders, diabetes, Alzheimer's disease, and Par

BACKGROUND: Aging exacerbates post-stroke inflammation, contributing to worse neurological outcomes. However, the mechanisms underlying this age-dependent immune dysregulation remain unclear. Because immune cell metabolism critically shapes inflammatory responses, we investigated whether the metabolic state of circulating monocytes, key immune cells that traffic to the ischemic brain, is altered by age after stroke. We further examined whether enhancing cellular NAD+ availability with nicotinamide riboside (NR) could mitigate age-associated neuroinflammatory responses and improve stroke outcom

The present report is Werner's-syndrome associated with a neurogenic bladder. The patient visited our clinic with the chief complaint of retention of urine and urinary tract infections. The diagnosis of Werner's syndrome was made because the patient had such clinical features as bird-like or "masked" face, short stature, graying of the hair in youth, cataracts in youth, skin changes like scleroderma principally on limbs, early menopause. After trans-urethral-resection of bladder neck and administration of distigmine bromide, her bladder function was improved, she could urinate by herself and s
