Basic to virtually all relevant biological effects of ionizing radiation is the underlying damage produced in DNA and the subsequent cellular processing of such damage. The damage can be qualitatively different for different kinds of radiations, and the genetics of the biological systems exposed can greatly affect damage processing and ultimate outcome--the biological effect of concern. The accurate repair of DNA double-strand breaks (DSBs) is critical for the maintenance of genomic integrity and function. Incorrect repair of such lesions results in chromosomal rearrangements and mutations tha

Energy depositions by sparsely and densely ionizing radiation in the cellular DNA are responsible for deleterious radiobiological effects like the induction of chromosome aberrations or gene mutations. However, due to differences in the spatial pattern of energy deposition the action of densely ionizing radiation is quantitatively and qualitatively different from that of sparsely ionizing radiation. By the use of new molecular-biological techniques a deeper insight into the molecular events underlaying particle-induced genetic changes has been obtained. The present paper will attempt to give a

The clinical benefits associated with the use of ionizing radiation for diagnostic and therapeutic purposes are well established, particularly in cancer medicine. Unfortunately, it is now clear that prior exposure to radiation is associated with an excess risk of developing malignancy in the exposure field. Indeed, the development of a second primary malignancy is a devastating side effect that can often be attributed to radiotherapy for a first cancer. Research has focused on elucidating the relationship between therapeutic radiation dose and site-specific cancer risk, and how this relationsh

Ionizing radiation exposure can induce profound changes in intracellular components, potentially leading to diverse health effects in exposed individuals. Any cellular component can be damaged by radiation, but some components affect cellular viability more profoundly than others. The ionization caused by radiation lasts longer than the initial inciting incident, continuing as 1 ionization incident causes another. In some cases, damage to DNA can lead to cellular death at mitosis. In other cases, activation of the genetic machinery can lead to a genetic cascade potentially leading to mutations

The genetic consequences resulting from environmental exposure to ionizing radiation have a significant impact on both radiation regulatory policies and the comprehension of the human health risks associated with radiation exposure. The primary objectives of the study were to assess 1) genotoxicity of exposure to radiation as a function of absorbed dose and dose rate, and 2) induction of a radio-adaptive response following a priming dose at varying dose rates. Results demonstrated that sub-acute environmental exposures of 10cGy gamma radiation resulted in indistinguishable levels of chromosoma

Radiation therapy, including conventionally fractionated external beam radiation therapy, stereotactic radiosurgery, and fractionated stereotactic radiation therapy, is a cornerstone in the interdisciplinary management of meningiomas. Recent advances in radiation oncology and also in other fields, such as neuropathology and imaging, have various implications for meningioma radiation therapy. This review aims to summarize current and anticipated developments, as well as active clinical trials related to the use of radiation therapy for meningiomas. In imaging, positron emission tomography has p

Subspecialty fellowship training is becoming more commonplace and medical practice more multidisciplinary. However, specialized training programs are challenged to provide a core curriculum that covers the interdisciplinary aspects of their field. Here we present a joint, interdisciplinary, interactive didactic program for breast fellowships in surgery, pathology and imaging. A yearlong core curriculum composed of topics of relevance to breast fellows involved in diagnostics and clinical care was created. Two hour long fellow-run interactive in-person sessions occur monthly. Additionally, facu

Virotherapy is currently undergoing a renaissance, based on our improved understanding of virus biology and genetics and our better knowledge of many different types of cancer. Viruses can be reprogrammed into oncolytic vectors by combining three types of modification: targeting, arming and shielding. Targeting introduces multiple layers of cancer specificity and improves safety and efficacy; arming occurs through the expression of prodrug convertases and cytokines; and coating with polymers and the sequential usage of different envelopes or capsids provides shielding from the host immune resp

PURPOSE: This review is to evaluate the use of biomarkers as an indication of past exposure to radiation or other environmental insults, individual sensitivity and risk for the development of late occurring disease.
OVERVIEW: Biomarkers can be subdivided depending on their applications. Markers of exposure and dose can be used to reconstruct and predict past accidental or occupational exposures when limited or no physical measurements were available. Markers of risk or susceptibility can help identify sensitivity individuals that are at increased risk for development of spontaneous disease an

BACKGROUND/AIM: Biomonitoring is currently applied in the estimation of health risks after overexposure to ionizing radiation (IR). The aim of this study was to compare the association of dicentric chromosomes and acentric fragments (AF) with cancer risk in subjects exposed to IR, as well as in control subjects.
MATERIALS AND METHODS: The study was performed on 3,574 subjects (2,030 subjects exposed to IR and 1,544 control subjects). The mean follow-up period was 8 years.
RESULTS: In subjects reporting exposure to IR, the presence of AFs and dicentric chromosomes was associated with a signif

Irradiation temperature, changed from 37 degrees C to 4 degrees C, acts as a dose-modifying factor with regard to the dose-yield relationship for dicentric chromosome aberrations in human lymphocytes irradiated with 150 kV X-rays. The temperature dependence of the aberration yield observed at constant dose is S-shaped, with a sharp rise near 15 degrees C from a lower plateau below 12 degrees C to a higher plateau beyond 17 degrees C. The aberration yield is determined by the irradiation temperature, irrespective of fast temperature changes from 4 degrees C to 37 degrees C or from 37 degrees C

The fractionation experiments were carried out on resting lymphocytes. Non-stimulated lymphocytes were X-irradiated at the total dose of 200 r separated into two equal parts by either 5-hour or 20-hour intervals. The whole blood samples were kept during the intervals between the exposure at the temperature of 20 degrees centigrade or 37 degrees centigrade. All the cultures were made after the last radiation exposure at 37 degrees centigrade. Dicentrics and centric rings were scored. It is shown that a fractionation effect takes place in resting lymphocytes when the whole blood was kept for int

Antarctic notothenioid fishes are the classic example of vertebrate adaptive radiation in a marine environment. Notothenioids diversified from a single common ancestor ∼22 Mya to between 120 and 140 species today, and they represent ∼90% of fish biomass on the continental shelf of Antarctica. As they diversified in the cold Southern Ocean, notothenioids evolved numerous traits, including osteopenia, anemia, cardiomegaly, dyslipidemia, and aglomerular kidneys, that are beneficial or tolerated in their environment but are pathological in humans. Thus, notothenioids are models for understanding a

PURPOSE: Since the genotoxicity of ionizing radiation was demonstrated in the 1920s, its hereditary effects have remained a serious concern for human society. The International Commission on Radiological Protection has highlighted the need for appropriate protection against hereditary effects of radiation in humans. In this paper, we review the literature on the possible multigenerational and transgenerational effects following testicular exposure to radiation, focusing on sperm epigenetic alterations as possible mechanisms.
RESULTS: This mini-review highlights that hereditary effects followi

A study was made of the frequency of chromatid aberrations in lymphocyte culture of subjects affected by the Chernobyl accident as a function of dose estimated by the incidence of dicentrics. The average number of chromatid aberrations was nearly the same within the dose range from 0 to 5 Gy exhibiting a tendency towards growth with dose. A high individual variability of the chromatid aberration frequency was observed.

A subset of patients receiving radiation therapy for pelvic cancer develop radiation cystitis, a complication characterized by mucosal cell death, inflammation, hematuria, and bladder fibrosis. Radiation cystitis can reduce bladder capacity, cause incontinence, and impair voiding function so severely that patients require surgical intervention. Factors influencing onset and severity of radiation cystitis are not fully known. We tested the hypothesis that genetic background is a contributing factor. We irradiated bladders of female C57BL/6, C3H, and BALB/c mice and evaluated urinary voiding fun

Two of the major cutaneous consequences of ultraviolet (UV) radiation exposure are immunosuppression and the development of skin cancer. This study examined whether these effects are genetically determined. Suppression of contact hypersensitivity by local, low-dose UV radiation was examined in what have been termed "UV-susceptible" and "UV-resistant" strains of mice. C3H/HeJ mice ("UV resistant") were resistant to the adverse effects of low-dose UV radiation when normal doses of hapten were applied to UV-irradiated skin; however, they were sensitive when the amount of hapten used for sensitiza
