PURPOSE: Cerebral cavernous malformations (CCMs) are hamartomatous vascular lesions. A part of CCMs is estimated to occur after cranial irradiation. However, there have been no reports documenting clinical picture of such CCMs. This study aimed to explore it.
METHODS: A total of 51 patients with multiple CCMs who visited our hospital, between 2006 and 2024, were explored. Among them, five were considered to have radiation-induced CCMs, four developing after cranial irradiation and one after endovascular therapy.
RESULTS: The male-to-female ratio of radiation-induced CCM cases was 3:2. The me

The detrimental effects associated with exposure to ionizing radiation have long been thought to result from the direct targeting of the nucleus leading to DNA damage; however, the emergence of concepts such as radiation-induced genomic instability and bystander effects have challenged this dogma. After cellular exposure to ionizing radiation, we have isolated a number of clones of Chinese hamster-human hybrid GM10115 cells that demonstrate genomic instability as measured by chromosomal destabilization. These clones show dynamic and persistent generation of chromosomal rearrangements multiple

Recent adaptive radiations provide experimental opportunities to parse the relationship between genomic variation and the origins of distinct phenotypes. Sympatric radiations of the charr complex (genus Salvelinus) present a trove for phylogenetic analyses as charrs have repeatedly diversified into multiple morphs with distinct feeding specializations. However, charr species flocks normally comprise only two to three lineages. Dolly Varden charr inhabiting Lake Kronotskoe represent the most extensive radiation described for the genus, containing at least seven lineages, each with defining morp

Cellular response to DNA damage, including ionizing radiation (IR) and UV radiation, is critical for the maintenance of genomic fidelity. Defects of DNA repair often result in genomic instability and malignant cell transformation. Centrosomal protein Nlp (ninein-like protein) has been characterized as an important cell cycle regulator that is required for proper mitotic progression. In this study, we demonstrate that Nlp is able to improve nucleotide excision repair (NER) activity and protects cells against UV radiation. Upon exposure of cells to UVC, Nlp is translocated into the nucleus. The

Chronic oxidative stress has been associated with genomic instability following exposure to ionizing radiation. However, results showing direct causal linkages between specific ROS (reactive oxygen species) and the ionizing radiation-induced mutator phenotype are lacking. The present study demonstrates that ionizing radiation-induced genomically unstable cells (characterized by chromosomal instability and an increase in mutation and gene amplification frequencies) show a 3-fold increase in steady-state levels of hydrogen peroxide, but not superoxide. Furthermore, stable clones isolated from pa

Genomic integrity is constantly insulted by solar ultraviolet (UV) radiation. Adaptative cellular mechanisms called DNA damage responses comprising DNA repair, cell cycle checkpoint, and apoptosis, are believed to be evolved to limit genomic instability according to the photoperiod during a day. As seen in many other key cellular metabolisms, genome surveillance mechanisms against genotoxic UV radiation are under the control of circadian clock systems, thereby exhibiting daily oscillations in their catalytic activities. Indeed, it has been demonstrated that nucleotide excision repair (NER), th

The Radiation Oncology-Biology Integration Network (ROBIN) initiative addresses critical gaps in radiation oncology by integrating advanced biological research, technologic innovation, and clinical practice. ROBIN leverages "omics" technologies, data science, and integrative analyses to elucidate the mechanisms governing tumor and normal tissue responses to radiotherapy (RT). Through five specialized centers-OligoMET, ImmunoRad, GenRad, METEOR, and KIDSROBIN-the network covers a broad spectrum of cancer and radiation biology research. Each center conducts translational programs linked to clini

According to the results of recent studies, parental exposure to ionizing radiation not only leads to mutation induction in the germline of irradiated animals but also affects their non-exposed offspring. These radiation-induced transgenerational effects belong to an epigenetic phenomenon that could not be defined as a transmission of altered phenotypes from the irradiated parents to their non-exposed offspring. In this review, we present the results of laboratory studies aimed to evaluate the transgenerational effects of parental irradiation on a number of traits in the offspring of exposed p

INTRODUCTION: Microorganisms can have major impacts on the success of NASA's missions, including the integrity of materials, the protection of extraterrestrial environments, the reliability of scientific results, and maintenance of crew health. Robust cleaning and sterilization protocols for spacecraft and associated environments are currently in place in NASA facilities, but microbial contamination should be further controlled and its impact on NASA's missions and science must be minimized. To address this, air and surfaces across cleanrooms and uncontrolled spaces at the Marshall Space Fligh

Arabidopsis thaliana from the Chernobyl Exclusion Zone showed altered stress responses, reduced germination, and genomic signatures of microevolution affecting DNA repair, redox signalling and cell-cycle-related genes. Chronic exposure to ionising radiation (IR) in the Chernobyl Exclusion Zone (CEZ) has created a field experiment for plant evolution. We collected Arabidopsis thaliana seeds from a reference plot (Babchin) and two radioactively contaminated plots (Vygrebnaya Sloboda and Masany), established in vitro seed lines for each plot and studied their physiology and genomes. Seeds were ch

BACKGROUND: Tumor genomic prognostic assays estimate 10-year local recurrence risk in ductal carcinoma in situ (DCIS) and can guide treatment decisions. This study aimed to evaluate which DCIS patients treated with breast-conserving surgery (BCS) underwent DCIS score genomic testing and the influence of the results on adjuvant treatment recommendations.
METHODS: The study identified patients from the National Cancer Database (NCDB) who had DCIS treated with BCS from 2010 to 2016.
RESULTS: Of 141,047 patients, 4255 (3%) had a DCIS score assessed, 0.3% in 2010 increasing to 5.8% in 2016 (p < 0

The origin of the extensive phenotypic divergence characterizing adaptive radiation could often be geographically localized and genetically simple. In a classic case of a trophically polymorphic cichlid fish (Herichthys minckleyi), we investigated alternative genomic processes that could have produced its extreme within-population variation in pharyngeal jaw tooth size. First, we generated a high-quality reference genome for its close relative (H. cyanoguttatus) to dissect the genetic architecture of this dental polymorphism. Then, using whole genome resequencing across the small Cuatro Ciéneg

Exposure to ionizing radiation can increase the risk of cancer, which is often characterized by genomic instability. In environmental exposures to high-LET radiation (e.g. 222Ra), it is unlikely that many cells will be traversed or that any cell will be traversed by more than one alpha particle, resulting in an in vivo bystander situation, potentially involving inflammation. Here primary human lymphocytes were irradiated with precise numbers of 3He2+ ions delivered to defined cell population fractions, to as low as a single cell being traversed, resembling in vivo conditions. Also, we assessed

Exposure to ionizing radiation results in a variety of genome rearrangements that have been linked to tumor formation. Many of these rearrangements are thought to arise from the repair of double-strand breaks (DSBs) by several mechanisms, including homologous recombination (HR) between repetitive sequences dispersed throughout the genome. Doses of radiation sufficient to create DSBs in or near multiple repetitive elements simultaneously could initiate single-strand annealing (SSA), a highly efficient, though mutagenic, mode of DSB repair. We have investigated the genetic control of the formati

BACKGROUND: Pediatric high-grade glioma (pHGG) is largely incurable and accounts for most brain tumor-related deaths in children. Radiation is a standard therapy, yet the benefit from this treatment modality is transient, and most children succumb to disease within 2 years. Recent large-scale genomic studies suggest that pHGG has alterations in DNA damage response (DDR) pathways that induce resistance to DNA damaging agents. The aim of this study was to evaluate the therapeutic potential and molecular consequences of combining radiation with selective DDR inhibition in pHGG.
METHODS: We condu

Apoptosis is a necessary form of cell death for all multicellular organisms and is controlled by the action of genes that initiate, facilitate or inhibit the process. In addition to physiological triggers, apoptosis can follow cellular injury, such as exposure to ionizing radiation. Cell death following radiation serves as a means by which the possibility of cells surviving with potentially harmful genomic damage can be reduced. Apoptosis can be identified in tissues shortly after whole body radiation; but apoptotic cells are rapidly cleared by phagocytosis, and apoptosis is unlikely to serve

Exposure to ionizing and non-ionizing radiation from environmental and clinical settings can significantly threaten genomic stability, especially when combined. This ex vivo study investigates the potential combined effects of gamma radiation and ultraviolet B (UVB) exposure on human peripheral blood mononuclear cells (PBMCs) from healthy donors by exposing whole blood and isolated PBMCs to 1 Gy of gamma rays, to an absolute dose of approximately 100 J/m2 of UVB, or to their combination. Combined exposure resulted in significantly elevated γH2AX foci formation and chromosomal aberrations relat

Exposure to ionizing radiation may induce a heritable genomic instability phenotype that results in a persisting and enhanced genetic and functional change among the progeny of irradiated cells. Since radiation-induced bystander effects have been demonstrated with a variety of biological end points under both in vitro and in vivo conditions, this raises the question whether cytoplasmic irradiation or the radiation-induced bystander effect can also lead to delayed genomic instability. In the present study, we used the Radiological Research Accelerator Facility charged-particle microbeam for pre

Deinococcus species, noted for their exceptional resistance to DNA-damaging environmental stresses, have piqued scientists' interest for decades. This study dives into the complex mechanisms underpinning radiation resistance in the Deinococcus genus. We have examined the genomes of 82 Deinococcus species and classified radiation-resistance proteins manually into five unique curated categories: DNA repair, oxidative stress defense, Ddr and Ppr proteins, regulatory proteins, and miscellaneous resistance components. This classification reveals important information about the various molecular mec

Behavioral and cognitive traits have a genetic component even though contributions from individual genes and genomic loci are in many cases modest. Changes in the environment can alter genotype-phenotype relationships. Space travel, which includes exposure to ionizing radiation, constitutes environmental challenges and is expected to induce not only dramatic behavioral and cognitive changes but also has the potential to induce physical DNA damage. In this study, we utilized a genetically heterogeneous mouse model, dense genotype data, and shifting environmental challenges, including ionizing r
