Association of total and pancreatic serum amylase enzymatic activity with insulin resistance and the glucose and insulin responses to oral starch test in Mexican children.

BACKGROUND/OBJECTIVES: Little is known about the effect of serum amylase enzymatic activity on glucose metabolism. We investigated the association of serum amylase enzymatic activity with fasting plasma glucose, insulin resistance (IR), and the plasma glucose and insulin response to an oral starch test (OST) in Mexican children. METHODS: Anthropometric data, glucose and insulin levels, and the serum enzymatic activity of total (AMYt), salivary (AMY1), and pancreatic (AMY2) amylase were analysed in 764 children (Nnormal weight  = 427/Nobesity  = 337). After categorization into low (LA) and hig

Type 3 resistant starch from Canna edulis improves Parkinson's symptoms, including behavioral function and nerve damage, in Parkinson's model rats by regulating the gut-brain axis.

Intestinal microorganisms affect the pathogenesis of Parkinson's disease (PD) and, therefore, are a new research focus of PD treatment. Type 3 resistant starch from Canna edulis (Ce-RS3) regulates gut microbes and has prebiotic properties. The effects include decreased blood lipids, weight, and inflammation, as well as increased glycolipid metabolism. There are few reports on the use of prebiotics alone in the treatment of Parkinson's. In this study, based on the gut-brain axis, the pharmacological effects and related mechanisms of Ce-RS3 on PD were studied by analyzing intestinal bacteria and…

Improving the processability and digestion resistance of rice starch gel through linear chain-ferulic acid V-type complexation and branch-mediated crosslinking.

Ferulic acid (FA) reduces starch digestibility but disrupts gel network, limiting its application in gel strength-reliant products. Rice starch was sequentially modified by pullulanase and transglucosidase to generate linear chains with a few branches (LCFBs), which were complexed with 0-2.0% FA to clarify how intermolecular interactions regulated the trade-off between gel strength and digestion resistance. At 0.5% FA, the hardness of modified starch gel remained at 98.2 g, whereas that of native starch gel decreased from 122.0 to 76.1 g. Moreover, the latter could not be shaped into blocks at

Impact of resistant starch from high amylose wheat compared to inulin on human gut microbiota and host immune responses using an artificial colon associated to human leukocytes.

Cooked High Amylose Wheat (HAW) products are dietary sources of resistant starch (RS). While its effects on gut microbiota are known, links to immune responses remain unclear. This study evaluated the antioxidant and anti-inflammatory properties of RS from HAW compared to inulin using a human colon model (ARCOL) coupled with blood leukocytes. ARCOL was supplemented for 7 days and fermentation supernatants incubated with leukocytes to measure inflammatory markers. Both RS and inulin increased short chain fatty acids, gas production, and modified bacterial profiles in a donor-dependent manner. B…

Synergistic co-processing of heat-moisture treated resistant rice starch with HPMC and Eudragit® S100: A novel multifunctional excipient for direct compression and colon-targeted delivery.

A novel multifunctional excipient was developed based on co-processed resistant rice starch (RRS) for direct compression (DC) tablet and targeted colonic drug delivery. While rice starch (RS) is biodegradable and exhibits good compressibility for DC, its poor flowability and low resistant starch content limit its applicability in both DC and controlled-release formulations. To address these limitations, RS was modified using heat-moisture treatment (HMT) with optimal process parameters (moisture content, temperature, and treatment time) were determined through a design of experiments (DoE) app…

Preparation of flexible poly(l-lactide)-b-poly(ethylene glycol)-b-poly(l-lactide)/talcum/thermoplastic starch ternary composites for use as heat-resistant and single-use bioplastics.

Poly(l-lactide)-b-poly(ethylene glycol)-b-poly(l-lactide) block copolymer (PLLA-PEG-PLLA) is a highly flexible bioplastic, yet its use in practical applications is limited due to its poor heat resistance and high production cost. In this study, talcum was used as a nucleating agent to improve the heat resistance, and thermoplastic starch (TPS) was used as a low-cost filler to reduce the cost of production. PLLA-PEG-PLLA/talcum/TPS and PLLA/talcum/TPS ternary composites with 4 wt% talcum and various TPS contents were prepared by melt blending before injection molding and were then evaluated. Wh

Baseline co-occurrence of Faecalibacterium prausnitzii and Ruminococcus bromii is associated with greater increases in selected short-chain fatty acid-related pathways following resistant starch type 2 intervention across diverse populations.

Type 2 resistant starch (RS2) can be fermented by the gut microbiota and contribute to short-chain fatty acid (SCFA) production; however, microbial responses to resistant starch vary substantially across individuals and populations, and the role of baseline microbiota in this variability remains incompletely understood. In this study, we integrated four publicly available paired metagenomic datasets representing diverse experimental and host contexts and performed cross-dataset analyses of taxonomic and functional responses. Faecalibacterium prausnitzii was among the taxa consistently enriched…

Structural-state-dependent effects of RVA-treated waxy starch on multiscale evolution and freeze-thaw resistance of myofibrillar protein emulsion gels: a perspective from processing to storage.

Physically modulating the structural state of waxy starch offers a clean-label strategy to improve the freeze-thaw (FT) tolerance of high-fat myofibrillar protein (MP) emulsion gels. In this study, waxy corn starches with distinct structural states, including native crystalline starch (NCS), partially disordered starches with gelatinization degrees of 45% (PDS45) and 70% (PDS70), and amorphous starch (AS), were prepared by thermal-shear treatment. These starches were incorporated into MP emulsions to elucidate their roles during emulsification, thermal gelation, and FT storage. During emulsifi

New insights into starch, lipid, and protein interactions - Colon microbiota fermentation.

Starch, lipids, and proteins are essential biological macromolecules that play a crucial role in providing energy and nutrition to our bodies. Interactions between these macromolecules have been shown to impact starch digestibility. Understanding and controlling starch digestibility is a key area of research. Investigating the mechanisms behind the interactions of these three components and their influence on starch digestibility is of significant practical importance. Moreover, these interactions can result in the formation of resistant starch, which can be fermented by gut microbiota in the …

Sequential effects of autoclaved heat treatment and electron beam irradiation on acorn starch: Multiscale structural differences and related mechanisms.

In this study, the differences in the modification effects and related mechanisms of different times (20 and 40 min) of autoclaved heat (AH) treatment and different doses (2 and 4 kGy) of electron beam irradiation (EBI) in different sequences of effects on acorn starch were investigated. The results showed that both AH and EBI reduced the amylose content (22.70 to 19.59%) and enthalpy (10.28 to 1.84%) of starch but increased the resistant starch content (53.69 to 64.11%). AH treatment made the crystalline regions of the residual starch granules denser, which was resistant to the action of amyl…

Ileal starch digestibility and the postprandial glycemic response to faba beans in healthy humans.

Legumes are a sustainable protein source and a recommended starchy food in healthy diets. Resistant starch has shown beneficial effects on glycemic control. However, the digestibility of the starch, indicating the amount of resistant starch, in legumes has been poorly studied in humans. We aimed to assess the ileal starch digestibility from faba beans in healthy volunteers and the associated glycemic response. Nine healthy volunteers were equipped with a naso-ileal tube. They ingested 250 g of dehulled, cooked mashed faba beans as a test meal. Ileal contents, plasma and urine samples were coll…

Butylated starch alleviates polycystic ovary syndrome by stimulating the secretion of peptide tyrosine-tyrosine and regulating faecal microbiota.

Polycystic ovary syndrome (PCOS) is an endocrine disorder that affects women of reproductive age. Resistant starch can be fermented by gut microbes then produce short-chain fatty acids (SCFAs). It was reported SCFAs might play a key role in gut microbiota-dependent therapy of PCOS. However, the effects of resistant starch on PCOS symptoms have not yet been investigated. Here, maize starch was acylated with specific SCFAs. Our results indicated that acylated starch can release acetate, propionate, and butyrate into the caecum and colon. Treatment with butylated starch (BS) alleviated abnormal o…
