Adult sea lamprey tolerates biliary atresia by altering bile salt composition and renal excretion.

The sea lamprey (Petromyzon marinus) is a genetically programmed animal model for biliary atresia, as it loses its bile ducts and gallbladder during metamorphosis. However, in contrast to patients with biliary atresia or other forms of cholestasis who develop progressive disease, the postmetamorphosis lampreys grow normally to adult size. To understand how the adult lamprey thrives without the ability to secrete bile, we examined bile salt homeostasis in larval and adult lampreys. Adult livers were severely cholestatic, with levels of bile salts >1 mM, but no evidence of necrosis, fibrosis, or

Genome-wide identification of the RLCK gene family in sugarcane and functional analysis of ScRLCK53 in salt tolerance.

Sugarcane (Saccharum spp.) is a major sugar crop, yet its growth and yield are frequently limited by soil salinization. Receptor-like cytoplasmic kinases (RLCKs) act as central signaling hubs in plants, regulating growth, development, immunity, and abiotic stress responses. However, the evolution of the RLCK gene family in sugarcane and its role in salt tolerance remain unclear. Using the high-quality genome of elite cultivar 'Zhongzhe No.1', we identified 262 ScRLCK genes, grouped into 15 subfamilies, all of which contain a typical protein kinase domain. Synteny analysis revealed that whole-g…

Genome-Wide and GWAS Dissection of Maize Fibrillin Genes Reveals Plastid Regulators of Drought and Salt Stress Tolerance.

Fibrillins (FBNs) are conserved plastid-associated proteins implicated in plant development and abiotic stress responses; however, their roles in maize remain unclear. In this study, through a genome-wide bioinformatic analysis, we identified 14 ZmFBN genes in the maize genome and characterized their phylogeny, chromosomal distribution, gene structure, conserved motifs, and promoter cis-elements. ZmFBN members were grouped into several subfamilies that all retain a conserved PAP_fibrillin domain, whereas the variation in exon-intron organization, motif composition, and regulatory elements sugg

Identification and preliminary functional characterization of MsPAL2, a key salt-tolerance gene during seed germination in Medicago sativa.

BACKGROUND: Alfalfa (Medicago sativa) is an important legume forage crop. Salt stress severely impairs seed germination, compromising field establishment and subsequent seedling growth. However, the key genes underlying alfalfa seed responses to salt stress during germination remain largely unidentified, hindering targeted genetic improvement of salt tolerance within the genus Medicago. RESULTS: In this study, transcriptomic sequencing was performed on germinated and ungerminated seeds of alfalfa treated with 150 mmol/L NaCl, using the 'Zhongmu No. 4' reference genome. Through integrated KEGG…

Cloning and functional characterization of OsERFSR1, a negative regulator of salt stress tolerance in rice (Oryza sativa L.).

Salt stress is one of the major abiotic factors limiting rice growth, development and yield. In this study, a member of the AP2/ERF transcription factor family, OsERFSR1, was cloned from rice. Overexpression of OsERFSR1 (OE) significantly reduced salt tolerance, with survival rates of 53.2% compared with 70.2% in wild-type (WT) plants, whereas CRISPR/Cas9-mediated knockout (ko) markedly enhanced salt tolerance, with survival rates of 54.6% compared with 28.2% in WT, suggesting that OsERFSR1 acts as a negative regulator of salt stress tolerance. Physiological analyses revealed that, under salt …

Abscisic acid-regulated stability of CmABF1 and CmBRM modulates salt tolerance in chrysanthemum via epigenetic regulation of CmHSFA4.

Soil salinization poses a major threat to global agricultural productivity and plant biodiversity. The phytohormone abscisic acid (ABA) is central to plant adaptation to abiotic stress; however, the mechanisms by which ABA coordinates posttranslational modifications of signaling proteins with epigenetic regulation remain poorly understood. Here, we show that salt stress-induced ABA accumulation up-regulates Heat Shock Factor 4 (CmHSFA4), a gene that is known to enhance chrysanthemum salt tolerance. The ABA responsive transcription factor ABRE binding factor 1 (CmABF1) binds to the CmHSFA4 prom…

Soybean Inositol Polyphosphate 5-Phosphatase 8 Confers Salt Tolerance by Reducing Sodium Influx Through Inositol 1,4,5-Trisphosphate Signalling.

Evidence suggests that the metabolism of inositol and its derivatives may be involved in various biological processes including salt tolerance, but there has been limited understanding. Ectopic expression of Gs5PTase8, an inositol polyphosphate 5-phosphatase cloned from wild soybean (Glycine soja), significantly enhanced salt tolerance in cultivated soybean (Glycine max). In this follow up study, the overexpression of Gs5PTase8 was shown to improve salt tolerance in transgenic Arabidopsis thaliana, soybean hairy roots and composite plants, by preventing sodium (Na+) accumulation and maintainin…

Overexpression of PeMIPS1 confers tolerance to salt and copper stresses by scavenging reactive oxygen species in transgenic poplar.

Myo-inositol is a vital compound in plants. As the key rate-limiting enzyme in myo-inositol biosynthesis, l-myo-inositol-1-phosphate synthase (MIPS) is regarded as a determinant of the myo-inositol content in plants. The up-regulation of MIPS genes can increase the myo-inositol content, thereby enhancing the plant's resistance to a variety of stresses. However, there are few reports on the roles of myo-inositol and the identification of MIPS in woody trees. In this study, a MIPS gene, named as PeMIPS1, was characterized from Populus euphratica Oliv. The heterologous expression of PeMIPS1 compe

Mercury mobility and effects in the salt-marsh plant Halimione portulacoides: Uptake, transport, and toxicity and tolerance mechanisms.

The plant Halimione portulacoides, an abundant species widely distributed in temperate salt-marshes, has been previously assessed as bioindicator and biomonitor of mercury contamination in these ecosystems. The present study aims to assess uptake and distribution of total mercury (THg) and methylmercury (MMHg) within H. portulacoides, potential mercury release by volatilization through leaves, and toxicity and tolerance mechanisms by investigating plant photochemical responses. Stem cuttings of H. portulacoides were collected from a salt-marsh within the Tagus estuary natural protected area, a

Approaches in modulating proline metabolism in plants for salt and drought stress tolerance: Phytohormones, mineral nutrients and transgenics.

Major abiotic stress factors such as salt and drought adversely affect important physiological processes and biochemical mechanisms and cause severe loss in crop productivity worldwide. Plants develop various strategies to stand healthy against these stress factors. The accumulation of proline (Pro) is one of the striking metabolic responses of plants to salt and drought stress. Pro biosynthesis and signalling contribute to the redox balance of cell under normal and stressful conditions. However, literature is meager on the sustainable strategies potentially fit for modulating Pro biosynthesis

Producing alcohol and salt stress tolerant strain of Saccharomyces cerevisiae by heterologous expression of pprI gene.

INTRODUCTION: Ethanol is considered a comparatively clean biofuel, and its large scale production has been a long time concern. Saccharomyces cerevisiae has proven to be the suitable microorganism for large scale ethanol production, but production of other alcohols like butanol and using lignocellulosic substrates is restricted due to lacking tolerance toward toxicity of alcohols, and compounds released from substrates. This study aimed to produce a tolerant strain by using pprI gene of Deinococcus radiodurans. MATERIAL AND METHOD: pprI gene was introduced into Saccharomyces cerevisiae. To ev

The glutathione-related metabolism and the AhGST23 gene mediate drought and salt stresses tolerance in peanut (Arachis hypogaea L.).

BACKGROUND: Abiotic stresses such as drought and salinity significantly constrain peanut productivity. However, their underlying molecular response mechanisms remain unclear. RESULTS: This study identified the AhGST23 gene, together with its involvement in glutathione metabolism and the ascorbic acid-glutathione (AsA-GSH) cycle, as a key component in alleviating drought and salt stresses in peanut. Under drought (15% PEG6000 for 6 days) and salt (200 mM NaCl for 6 days) stresses, peanut seedlings exhibited a marked reduction in biomass, net photosynthetic rate, transpiration rate, and chloroph

S-nitrosoglutathione enhances rice tolerance to salt-submergence by coordinating ethylene, GA, and ABA accumulation and improving ion transport.

With unprecedented climate change, coastal overtopping is projected to increase up to 50-fold by 2100. Unlike complete flooding, overtopping imposes a salt-submergence stress for plants by combining oxygen (O2) deficiency with elemental toxicity. To address this, we investigated whether S-nitrosoglutathione (GSNO), a stable nitric oxide (NO) donor, could mitigate these dual stresses-submergence (Sub) and salt-mixed Sub (Salt + Sub)-in two Korean japonica rice varieties, Ilmi and Samgwang. Under Sub stress, both varieties relied on internode elongation and aerenchyma formation as escape strateg
